Characterization of the Antigenicity and Immunogenicity of the Escherichia Coli Produced RNase Domain of the Classical Swine Fever Virus Glycoprotein E~(rns)

Erns is a highly glycosylated envelope protein of classical swine fever virus (CSFV) with RNase activity. Erns can induce neutralizing antibodies and provide immune protection against CSFV infection. In this study, the RNase domain of the Erns was produced in Escherichia coli. 杀鲑气单胞菌单克隆抗体的制备及其特性分析

应用杂交瘤单克隆抗体技术制备了6株分泌抗杀鲑气单胞菌杀鲑亚种的单抗细胞株,并对其特性进行分析。结果显示：6株单抗中IgM有3株,IgG1有2株,IgG2a有1株,且抗体效价为1：12800-1：51200,检测灵敏度为1．0×10^5-1．0×10^8cfu·mL^-1。进一步实验证实这些单抗与其他病原菌都无交叉反应。但单抗5C7、7H6与杀日本鲑亚种有交叉反应;单抗8A2与无色亚种存在阳性反应。表明杀鲑气单胞菌亚种之间既有独特的抗原决定簇,又有共同抗原位点。制备的单抗可用于杀鲑气单胞菌的快速诊断和亚种鉴定,为该菌的进一步研究提供必要手段。     

Efficacy of anti-CD20 chimeric Fab’ fragment on proliferation of B lymphoma cells

The variable domain of heavy chain (V_H) and light chain (V_L) genes of anti-CD20 monoclonal antibody HI⁴⁷ were cloned from anti-CD20 ScFv expression vector pCANBTEcd20 by PCR and ligated into vector pYZF to construct chimeric anti-CD20 Fab’ fragment expression vector pYZFcd20. Chimeric anti-CD20 Fab’ fragment was expressed inE. coli 16C9 and purified by protein G affinity chromatography. Competitive inhibition assay showed that anti-CD20 Fab’ fragment inhibited binding of HI⁴⁷ to CD20 on the surface of Daudi cells. Results from MTT assay indicated that chimeric anti-CD20 Fab’ fragment inhibited the proliferation of Daudi cells, IC₅₀ = 69 μg/mL. Affinity of chimeric anti-CD20 Fab’ fragment was determined, K_a was about 8.9×10⁸ (mol/L)⁻¹.     

Induction of multi-epitopespecific antibodies against HIV-1 by multi-epitopevaccines

Some neutralizing antibodies against HIV-1 envelope proteins were highly effective to inhibit the infection of different strainsin vitro, and existed in the infected individuals with very low levels. We suggested multi-epitope-vaccine as a new strategy to increase levels of neutralizing antibodies and the abilities against HIV mutationin vivo. Two candidate multi-epitope-vaccines induced antibodies with predefined multi-epitope-specificity in rhesus macaque. These antibodies recognized corresponding neutralizing epitopes on epitope-peptides, gp41 peptides, V3 loop peptide, rsgp41 and rgp120. Besides, three candidate epitope-vaccines in combination (another kind of multi-epitopevaccines) showed similar potency to induce predefined multiple immune responses in rabbits. These results suggest that multi-epitope-vaccines may be a new strategy to induce multi-antiviral activities against HIV-1 infection and mutations.     

ELISA对毒理实验动物血清中神经生长因子抗体动态的研究

用ＥＬＩＳＡ法对神经生长因子（ＮＧＦ）毒理实验动物犬和大鼠血清作了抗－ＮＧＦ抗体检测。结果表明在大鼠血清中没有发现抗－ＮＧＦ抗体,但犬血清中有相应的抗体产生,并随注射剂量增加和注射时间延长而逐渐升高,另外,对ＥＬＩＳＡ间接法和竞争抑制法进行了比较,结果表明,竞争抑制法明显优于间接法,而且是一种简便、特异的好方法。     

特异性抗体和特异性免疫复合物的检测对结核病的诊断意义

以ＥＬＩＳＡ检测１０８例活动性肺结核、３９例稳定期肺结核、３０例肿瘤、１０１例非结核性肺部疾病患者及２０２例正常人血清的抗结核菌纯蛋白衍生物（ＰＰＤ）ＩｇＧ抗体（简称抗－ＰＰＤ）和循环免疫复合物（ＣＩＣ）中的抗－ＰＰＤＩｇＧ抗体（简称ＣＩＣ－ＰＰＤ）。结果表明，活动性肺结核患者的抗－ＰＰＤ和ＣＩＣ－ＰＰＤ水平明显高于正常人和各疾病组患者的水平（Ｐ＜０．００１），但与其病变范围无关。单一测定活动性肺结核患者的抗－ＰＰＤ，敏感性为８５．２％，特异性为９８．５％；测定ＣＩＣ－ＰＰＤ，敏感性为８６．１％．特异性９６．９％。以两者任一项阳性作为诊断活动性肺结核的标准，则敏感性为９３．５％。此外，对１１例活动性肺结核患者的动态观察表明，随着治疗天数的增加和临床症状的改善，患者的抗－ＰＰＤ和ＣＩＣ－ＰＰＤ水平会逐渐降低，前者于治疗后第２０～２４周降至正常水平，后者则于治疗后第１２～１６周降至正常值。研究结果提示测定患者的抗－ＰＰＤ和ＣＩＣ－ＰＰＤ的水平，可用于结核病诊断，对结核病的疗效判定亦有帮助。     

小鼠血清甲状腺激素水平与免疫功能的衰老变化

利用放射免疫测定法和免疫功能指标的检测方法研究了 2 0月龄老年小鼠血清甲状腺激素水平和免疫功能的衰老变化 ,并用外源性甲状腺激素胃内给药的方法探讨了甲状腺机能和免疫功能变化间的因果关系 .结果证明 ,老年期甲状腺机能和免疫功能都有明显的衰退变化 ,且二者间存在有明显的因果关系 .甲状腺机能衰退导致免疫功能紊乱可能是机体衰老的基本过程之一     

Piezoelectric immunoassay of thyroidal triiodothyronine

A re-utilizable piezoelectric crystal (PC) immunosensor for thyroidal triiodothyronine (T3) has been developed. The crystal was coated with protein A and then reacted with T3 antibody. T3 could be detected in the range of 0.5×10⁻⁶ g/L to 10.0×10⁻⁶g/L by the, system. Crystal was regenerated by saturating with T3 and subsequent binding of a new anti-T3 antibody layer. 抗人凝血VIII因子的单克隆抗体制备及其鉴定

经凝胶过滤和亲和层析分离得到的人抗血友病A因子——VIII因子抗原VIII:CAg,在用一期法和火箭免疫电泳证实其纯度后,免疫Balb/c小鼠和包被免疫测定板。采用常规鼠杂交瘤技术经间接ELISA法筛选、克隆化得到四个稳定分泌抗VIII:C的单克隆抗体的细胞株,即DC8,DE4,DE7和DE8,腹水效价为1:1×10~4～1:1×10~5,一期法抑制反应表明,单克隆抗体对VIII:C活性有显著的抑制作用,但其抑制是不完全的,对抗体进行了纯化及分析,并将单抗用于血友病A多态性的分析。
