AN ALTERNATIVE APPROACH TO GLOBAL ROBUST OUTPUT REGULATION OF OUTPUT FEEDBACK SYSTEMS

The global robust output regulation problem of the output feedback systems has been extensively studied under various assumptions of the complexity and uncertainty. All these approaches boil down to a stabilization problem of a so-called augmented extended system. This paper will describe an alternative approach which converts the original problem into a stabilization problem of a so-called extended augmented system. As the extended augmented system is somewhat simpler than the augmented extended system, this alternative approach is also simpler than the first approach.

Genetic background of phenotypic variation

A noteworthy feature of the living world is its bewildering variability. A key issue in several biological disciplines is the achievement of an understanding of the hereditary basis of this variability. Two opposing, but not necessarily irreconcilable conceptions attempt to explain the underlying mechanism. The gene function paradigm postulates that phenotypic variance is generated by the polymorphism in the coding sequences of genes. However, comparisons of a great number of homologous gene and protein sequences have revealed that they predominantly remained functionally conserved even across distantly related phylogenic taxa. Alternatively, the gene regulation paradigm assumes that differences in the cis-regulatory region of genes do account for phenotype variation within species. An extension of this latter concept is that phenotypic variability is generated by the polymorphism in the overall gene expression profiles of gene networks. In other words, the activity of a particular gene is a system property determined both by the cis-regulatory sequences of the given genes and by the other genes of a gene network, whose expressions vary among individuals, too. Novel proponents of gene function paradigm claim that functional genetic variance within the coding sequences of regulatory genes is critical for the generation of morphological polymorphism. Note, however, that these developmental genes play direct regulatory roles in the control of gene expression.

Different patterns of agonist-induced modulation of α1B-adrenoceptor density at different initial expression levels

We compared the norepinephrine (NE) induced α_1B-adrenoceptor (α_1B-AR) expression modulation between two transfected human embryonic kidney (MEK) 293 cell lines in which α_1B-AR densities were (6 336 ± 913) and (773 ± 164) fmol ▪ mg¹, respectively. Treatment of cells with NE (10 μmol ▪ L₁) for 48 h decreased high-level expressed α_1B-AR density, but increased low-level expressed α_1B-AR density. The protein kinase C inhibitor Calphostin C or Ro-31-8220 reversed, and its activator PMA mimicked the NE-induced down-regulation of high-level expressed α_1B-AR. Moreover, PMA induced a down-regulation of low-level expressed α_1B-AR. The endoplasmic reticulum Ca²⁺-ATPase inhibitor cyclopiazonic acid (CPA) and the calcium chelator BAPTA/AM did not affect the down-regulation of high-level expressed α_1B-AR, but inhibited the up-regulation of low-level expression α_1B-AR induced by NE. These results suggest that α_1B-adrenoceptor densities at different initial expression levels are differentially regulated by NE and their signal transduction pathways are different.
