Role of Cys221 and Asn116 in the zinc-binding sites of the Aeromonas hydrophila metallo-β-lactamase

The CphA metallo-h6elkpyfdlcp4jq4/xxlarge946.gif" alt="beta" align="MIDDLE" BORDER="0">-lactamase produced by Aeromonas hydrophila exhibits two zinc-binding sites. Maximum activity is obtained upon binding of one zinc ion, whereas binding of the second zinc ion results in a drastic decrease in the hydrolytic activity. In this study, we analyzed the role of Asn116 and Cys221, two residues of the active site. These residues were replaced by site-directed mutagenesis and the different mutants were characterized. The C221S and C221A mutants were seriously impaired in their ability to bind the first, catalytic zinc ion and were nearly completely inactive, indicating a major role for Cys221 in the binding of the catalytic metal ion. By contrast, the binding of the second zinc ion was only slightly affected, at least for the C221S mutant. Mutation of Asn116 did not lead to a drastic decrease in the hydrolytic activity, indicating that this residue does not play a key role in the catalytic mechanism. However, the substitution of Asn116 by a Cys or His residue resulted in an approximately fivefold increase in the affinity for the second, inhibitory zinc ion. Together, these data suggested that the first zinc ion is located in the binding site involving the Cys221 and that the second zinc ion binds in the binding site involving Asn116 and, presumably, His118 and His196.Received 3 March 2003; received after revision 4 August 2003; accepted 25 August 2003

非光滑(h,φ)-Dini-凸多目标规划解的充分性与对偶性

利用Ben-Tal广义代数运算,定义了(h,φ)-Dini右上方向导数和(h,φ)-Dini-梯度,提出了几类非光滑非凸函数的概念,在φ是严格递增函数,并且φ(0)=0相当弱的假设下,得到了(h,φ)-Dini-凸多目标规划的最优性条件和几个对偶性结果。     

高温后结构钢力学性能试验

为了研究结构钢高温后材料力学性能变化规律,对Q 235结构钢加热至不同高温,再经不同方式冷却后进行了力学性能试验。试验表明:结构钢经不同高温冷却后,有显著热变色反映;高温后的弹性模量、屈服强度、抗拉强度和断后伸长率等力学性能变化程度主要与曾经历的最高温度和冷却方式有关。根据试验结果,编制了高温后钢材热变色图谱,提出了高温后结构钢的屈服强度、抗拉强度计算公式,对高温后弹性模量的取值提出了建议。研究成果可供火灾后钢结构损伤评估和加固分析时参考。

引入强优美树概念，证明了三星体Ｒ＿［（ｎ－ｐ），ｎ，ｎ＋ｐ］（ｎ为自然数；ｐ＝１．２）是强优美树，从而得到四星体Ｒ＿［（ｎ－ｐ），ｎ，ｎ＋ｐ，ｍ］（ｍ，ｎ为自然数；ｐ＝１，２）是优美树的结果。
