白腐菌降解氯代酚类化合物的特性研究

研究了Mn^2＋对白腐菌（Phanerochanete chrysosporium）产生的过氧化物酶（MnP和LiP）活力的影响及胞内外酶在氯代酚化合物降解过程中的作用。结果表明：Mn^2＋对MnP和LiP的活力有显著影响；白腐菌对相对分子质量低的有机化合物的降解主要在细胞内依靠胞内酶的作用进行，而胞外酶加快了降解速率；两种胞外酶MnP和LiP在降解过程中所起的作用不同，MnP起主要作用，LiP起次要作用。白腐茵对酚类化合物的基质亲和性较活性污泥低，适宜于高浓有机废水的预处理；白腐茵对毒性的忍受能力与活性污泥微生物相似，但处理方式和条件的改变可以提高其毒性忍受能力。

从南海红树林内生真菌菌体中提取到胞外多糖W2 1 ,甲醇解研究表明W2 1 由葡萄糖 ,半乳糖和少量木糖组成     

用响应面分析法优化胞外多糖的发酵工艺

采用响应面分析方法对发酵培养的蔗糖质量分数、接种量和pH等条件进行优化.优选得到较佳的发酵工艺条件:蔗糖4%～5%,pH6 5～8,接种量10%左右.摇瓶发酵胞外多糖的产量22 6g/L,蔗糖转化率45%,产品黏度2 01Pa·s.30L生物反应器放大发酵的实验结果产胶30g/L,产品黏度4 6Pa·s.     

Lecticans: organizers of the brain extracellular matrix

Lecticans are a family of chondroitin sulfate proteoglycans, encompassing aggrecan, versican, neurocan and brevican. These proteoglycans are characterized by the presence of ahyaluronan-binding domain and a C-type lectin domain in their core proteins. Through these domains, lecticans interact with carbohydrate and protein ligands in the extracellular matrix and act as linkers of these extracellular matrix molecules. In adult brain, lecticans are thought to interact with hyaluronan and tenascin-R to form a ternary complex. We propose that the hyaluronan-lectican-tenascin-R complex constitutes the core assembly of the adult brain extracellular matrix, which is found mainly in pericellular spaces of neurons as ‘perineuronal nets’. Isolation, chemical characteristics and immunity activity of an extracellular polysaccharide EPS I isolated from Antarctic bacterium Pseudoalteromonas sp.S-15-13

A new extracelluar polysaccharide （EPS） was isolated and purified from Antarctic bacterium S-15-13, identified as Pseudoalteromonas sp. After being separated and purified by DEAE-Sephadex A-50 ionexchange and Sephadex G-100 gel chromatography, two mains fractions （EPS I and EPS Ⅱ ） were ob-tained. EPS I was composed of mannose, glucose and galactose with a molecular weight of 23kDa and EPS Ⅱ was composed of mannose only with a molecular weight of 62kDa. The effect of the polysaccharide EPS I on the cellular immune response of mice was investigated. Results demonstrated that EPS I could markedly facilitate lymphocyte proliferation, and might be a strong immunomodulator.     

利用EPS膜间作用能评价不同SRT下其污染潜能

考察了不同SRT条件下EPS膜污染潜能,并分析了不同污泥龄(SRT)条件下EPS与PVDF膜间的界面能参数及相互作用能形貌.结果表明,SRT 30 d的EPS与PVDF膜之间的界面自由能(ΔGadh)比SRT 15 d的EPS与PVDF膜之间的界面自由能(ΔGadh)减少12.3%.SRT 15 d的EPS到达PVDF膜表面比SRT 30 d的EPS到达膜表面需要多克服4.2 KT的斥力相互作用能.过滤实验显示SRT 15 d的EPS导致了65.2%的通量衰减,而SRT 30 d的EPS引起了57.2%的通量衰减.这一结果进一步证实了与SRT 15 d的EPS相比较,SRT 30 d的EPS具有更低的污染潜能.     

不同环境因子对爪哇伪枝藻分泌胞外多糖的影响

以一种从荒漠土壤中分离的典型丝状蓝藻——爪哇伪枝藻为材料,在实验室条件下研究了温度、盐度、pH和培养时间对爪哇伪枝藻胞外多糖分泌的影响.结果表明:随培养温度升高,伪枝藻胞外多糖分泌量逐渐升高,在35℃时达到最大值;在一定盐度范围内,胞外多糖分泌随盐度的增大而增大;pH在7.5时,胞外多糖产量显著高于酸性和碱性条件下的胞外多糖产量;随着培养时间延长,伪枝藻胞外多糖分泌逐渐增大.     

Effects of simulated microgravity on the cell growth and extracellular matrix of cultured chondrocytes

The effects of simulated microgravity on cell growth and extracellular matrix of chondrocytes have been examined by a light microscope and a scanning electron microscope using cultured chicken embryonic chondrocytes as the model. No notable difference of the cell density between the rotation group and the control group has been found. But during the same period, the growth spots of the rotation group were sparser than those of the control group. These results indicated that the lower level of cell development and differentiation happened in the rotation group. Observation by the scanning electron microscope showed that the extracellualr matrix decreased after rotating, and the fibres in the extracellular matrix were slighter and blurrier than those of the control group. It is concluded that the simulated microgravity can affect the secreting and assembly of the extracellular matrix. The possible mechanism for them is discussed. 粘质沙雷氏菌酶催化H2O2降解对苯二酚的产酶条件研究

文章介绍采用源于粘质沙雷氏菌(Serratia marcescens)菌株AB 90027的酶催化H2O2降解对苯二酚,研究了时间、温度、pH值及摇床转速等条件对产酶的影响,探讨了酶在细胞中的存在位置。研究结果表明,产酶的适宜条件为培养时间30 h、温度28℃初始pH10.0及摇床转速150 r/min。并且发现催化H2O2降解对苯二酚的酶主要存在于细胞外,属于胞外酶。对HPLC和UV-VIS监测结果的分析表明,对苯二酚的苯环被降解开环。     

拟南芥悬浮培养细胞外钙结合蛋白的初步检测

钙离子通过与其结合的蛋白质在植物细胞信号转导中起着重要的作用,已知植物细胞外空间钙离子浓度高达毫摩尔级,而与钙结合的胞外蛋白有待进一步的分析研究.采用丙酮沉淀法分别制备了拟南芥悬浮培养细胞壁盐提蛋白及培养介质蛋白,制备的蛋白经SDS-PAGE电泳分离后,电转移至硝酸纤维素膜,45Ca2 覆盖结合,放射自显影,结果表明悬浮细胞外可能存在多条可与钙离子结合的蛋白条带,其分子量约为17,30,40,68 ku,推测这些蛋白可能参与胞外钙离子相关的生物学过程.
