全长人PPAR-γ的表达、纯化与结构确证

目的建立表达、纯化结构完整的全长人PPAR-γ的方法。方法构建pReceiver-B01-PPAR-γ质粒并转化E．coliBL21（DE3）细胞,诱导表达重组蛋白,优化细胞生长条件;利用亲和色谱和尺寸排阻色谱纯化重组蛋白;胶内酶解重组蛋白,用离子阱质谱仪分析二维AgilentHPLC-Chip纯化的酶解片段;基于MS／MS搜索IPI、Swiss．Prot、NCBInr和MSDB数据库,鉴定重组蛋白。结果在优化的细胞生长条件（TB介质、37℃、0．8mMIPTG、诱导3h）,从每升TB中可获得280mg重组蛋白;两步纯化后,获得176mg、纯度为95％的均质重组PPAR-γ蛋白;经质谱分析和搜索蛋白质数据库,获得均匀地分布在整个PPAR-γ多肽链中的33个阳性肽段,覆盖率为60％,表明重组蛋白为结构完整的全长人PPAR-γ。配体结合活性位点S289、H323、H449和Y473无突变,保证全长人PPAR-γ与配体结合的生物学活性。结论本文所建立的方法能成功用于大量表达结构完整的全长人PPAR-γ,有利于进一步的结构、功能研究和活性配体的筛选。     

Histone methylation and ubiquitination with their cross-talk and roles in gene expression and stability

Methylation of lysine residues of histones is associated with functionally distinct regions of chromatin, and, therefore, is an important epigenetic mark. Over the past few years, several enzymes that catalyze this covalent modification on different lysine residues of histones have been discovered. Intriguingly, histone lysine methylation has also been shown to be cross-regulated by histone ubiquitination or the enzymes that catalyze this modification. These covalent modifications and their cross-talks play important roles in regulation of gene expression, heterochromatin formation, genome stability, and cancer. Thus, there has been a very rapid progress within past several years towards elucidating the molecular basis of histone lysine methylation and ubiquitination, and their aberrations in human diseases. Here, we discuss these covalent modifications with their cross-regulation and roles in controlling gene expression and stability. 岛型工程场地在地震波荷载下的动力响应分析

基于三维显式有限元算法并结合粘弹性人工边界条件,针对某岛型大型工程场地,研究了岛型地质、地形条件对地震动反应的影响,分析了该工程场地地表地震动峰值加速度的分布规律。在对该工程场地进行地震动小区划的基础上,分析了不同区的地表加速度反应谱特性。研究结果为该大型工程场地上建筑及设备布置的地震安全性评价提供参考。     

用GEP实现复杂函数的自动建模

GEP是一种新颖的遗传算法,在函数建模的应用中取得良好的结果.给出计算有效基因长度的伪代码,结合GRCM方法阅读基因,快速计算出染色体的适应值.在算法中增加了参数估计模块,用GEP得到较好模型后,用参数估计模块进行参数优化,试验显示这种混合的GEP方法比传统的最小二乘法、神经网络以及遗传程序设计等方法具有更好的性能.     

DNA methylation of fly genes and transposons

The use of anti-5-methylcytosine antibodies in affinity columns allowed the identification of methylated sequences in the genome of Drosophila melanogaster adults. In view of the presence of transposable elements amongst the identified sequences, it has been suggested that DNA methylation is involved in transposon control in the fly genome. On the contrary, a reanalysis of these data furnishes several intriguing elements that could raise new questions about the role that DNA methylation plays in the fly genome. The aim of the present paper is to discuss some features that emerge from the analysis of the identified methylated sequences. 企业经营者的动态激励机制设计

相对业绩比较信息能改进委托－代理关系中的激励与效率,设计一个对企业经营者动态的机制既要考虑经营者的前期业绩,也要参照其他经营者的业绩,经营者的努力水平不仅来自于显性报酬契约的激励,还来自于两种隐性激励－声誉效应和棘轮效应的影响,本文运用博弈论的方法研究基于相对业绩比较的动态激励机制设计问题。

焦循的戏曲理论著作<花部农谭>、<剧说>,尤为赞赏花部戏曲的重要价值,在中国戏曲批评史上产生了重要影响.焦循认为戏曲应具有示范作用,应重视观赏与演出紧密结合,并能同诗歌一样"言志",而某些优秀的花部戏曲确实符合他的审美标准.
