银合欢壳多糖酶在转基因Khao Dawk Mali 105籼稻中的表达

通过含有pCAMBIA1300载体(抗潮霉素基因(hpt)作为选择标记基因)的根癌农杆菌菌株EHA105的介导,将326个银合欢I型碱性壳多糖酶基因(KB3,GenBank登录号为:AAM49597)的氨基酸导入到籼稻Khao Dawk Mali 105(KDML 105)植株中,获得了KDML 105籼稻的愈伤组织。pCAMBIA1300载体包含有CaMV35S启动子、银合欢壳多糖酶基因和NOS终止子。在NB培养基的水稻秧苗中愈伤组织的转化率为94%.在培养温度为28℃,以含有4 mg/l BAP和7 g/l植物凝胶再生培养基的条件下,愈伤组织的再生频率高达80%.为了建立一个KDML 105农杆菌转化方法,考察了各方面的因素。共培养期间,在培养基中添加50μM的乙酰丁香酮对提高瞬时转化率非常重要。4周龄的盾片衍生的愈伤组织是最佳的起始材料。添加40 mg/l潮霉素和400 mg/l头孢霉素的NB培养基是水稻愈伤组织选择性转化的最佳培养基,其转化率达到78%.采用PCR、Northern Blot杂交凝胶、Southern Blot杂交凝胶和GUS分析技术研究了愈伤组织和转基因水稻中的壳多糖酶和gus基因的表达。壳多糖酶和gus基因在水稻的多个部位被检测到,包括愈伤组织、叶、根、茎和秧苗,但是它们在水稻根部的含量较少。抗真菌的水稻壳多糖酶表现出抗串珠镰刀菌的性质。     

Observations on the “tenuis group” (Eutardigrada,Macrobiotidae) and description of a new Macrobiotus species

A new species of the tardigrade genus Macrobiotus is described. The species, designated M. ciprianoi n. sp., was isolated from a mixture of Provence broom leaf litter and mosses, and from rock mosses collected in the Sierra de Guadarrama, Madrid (Spain). Given that Macrobiotus ciprianoi n. sp. shares several characters to members of the “tenuis group”, we assessed the taxonomic homogeneity of the group. The new species differs from those of the “tenuis group” according to a unique set of characters related with claw shape, features of the buccal‐pharyngeal apparatus, and egg morphology. Our analysis of holotypes and/or paratypes of “tenuis group” species and other Macrobiotus species with similar characters (M. bondavallii and M. caelicola) reflects the heterogeneity of this group of species as currently described.     

Hymenoptera collections of Boyer de Fonscolombe: Apoidea in the University Museum,Oxford

An account is given of the Apoidea collected by Boyer de Fonscolombe and preserved in the University Museum, Oxford. Lectotypes are designated for three species (Osmia tricornis Latreille, Lithurgus chrysurus Fonscolombe, and Macrocera grandis Fonscolombe). The identities of the type species of the eucerine genera Tetralonia and Tetraloniella and the correct application of the former name are discussed.     

Reissner-Nordström de Sitter黑洞的电磁粒子隧穿辐射

在Reissner-Nordström de Sitter黑洞时空背景下,采用Parikh -Wilczek半经典隧穿方法,计算了带有电荷和磁荷的静质量非零粒子在黑洞视界面上的隧穿率．结果表明,黑洞事件视界和宇宙视界处粒子的隧穿率与Bekenstein-Hawking熵有关,真实的辐射谱不再是严格的纯热谱,但这是对Hawking纯热谱的正确修正,结果满足量子理论的幺正性定理．     

基于自适应信任报警关联的P2P覆盖IDS

随着计算机网络安全需求的日益增长,分布式入侵检测技术成为一个至关重要的研究方向.但传统的分布式入侵检测系统仍存在一些缺陷,如在分布性、灵活性、协作性、检测效率和内部威胁应对等方面还不尽人意.为能更好地应对各种内部威胁,提出了一种基于自适应信任报警关联的对等覆盖入侵检测系统.在JXTA对等架构基础上实现了一个对等覆盖IDS原型,并在仿真网络环境中评估了该原型在检测应对Internet蠕虫传播和感染中的有效性.实验结果显示,该对等覆盖IDS能明显提高脆弱网络节点的平均幸存率.     

De Novo Assembly Methods for Next Generation Sequencing Data

The recent breakthroughs in next-generation sequencing technologies, such as those of Roche 454,Illumina/Solexa, and ABI SOLID, have dramatically reduced the cost of producing short reads of the genome of new species. The huge volume of reads, along with short read length, high coverage, and sequencing errors, poses a great challenge to de novo genome assembly. However, the paired-end information provides a new solution to these problems. In this paper, we review and compare some current assembly tools, including Newbler, CAP3, Velvet,SOAPdenovo, AllPaths, Abyss, IDBA, PE-Assembly, and Telescoper. In general, we compare the seed extension and graph-based methods that use the overlap/lapout/consensus approach and the de Bruijn graph approach for assembly. At the end of the paper, we summarize these methods and discuss the future directions of genome assembly.
