甘薯HXK基因的克隆、组织表达及生物信息分析

通过RT-PCR技术从甘薯的总RNA中克隆得到了全长的己糖激酶基因,测序结果表明HXK(hexokinase)基因编码区长1491bp,编码496个氨基酸,其翻译的蛋白质分子量为53.4kD,其氨基酸序列与其它已知高等植物HXK基因具有很高的同源性.构建了原核表达载体pET-HXK,经IPTG诱导后可表达获得相对分子量约为71kD的外源融合蛋白,与预测结果一致.并对HXK基因在不同组织中的转录水平进行了数字表达谱和荧光定量分析.     

基于EFICA的AD微阵列数据基因网络分析

将EFICA(Efficient Variant of Algorithm FastICA)方法与基因网络相结合分析一组阿尔茨海默病(AD)微阵列数据.根据分类结果提取特征基因集并探寻与早期AD相关的基因网络,实验结果表明,EFICA方法比传统的Fastica方法能够获得更好的分类效果.并且通过对基因网络的研究,扩展了EFICA在生物信息学中的应用,为AD疾病的进一步研究提供新思路.     

Neuroglobin mutation associated with hypoxia adaptation in Tibet chicken

Globin protein family plays an important role in storing and transporting oxygen. As a newly reported globin, the revealed function of neuroglobin includes binding and storing oxygen as well as facilitating the utilization of oxygen in neuronal cells. In the present study, mutations in exons of chicken Ngb gene were identified with the method of sequencing and created restriction site PCR in Tibet chicken and other four lowland chicken breeds. The mutations of Lys-2224(E4)-Asn and Ser-2279(E4)-Gly were identified in exon 4 of the gene. The Lys-2224(E4)-Asn mutation existed only in Tibet chicken and the mutation frequencies increased with increasing altitude. Analysis of the haplotype and diplotype of the two mutations in Tibet chicken populations of different altitudes showed that the frequencies of TG haplotype and TTGG diplotype also increased with increasing altitude, while the reverse tendency was found on GGAA diplotype. Under the hypoxic simulation incubation, the main haplotype was TG in living embryos and GA in dead embryos. The results showed that the Lys-2224(E4)-Asn mutation may be a specific mutation associated with hypoxia adaptation in Tibet chicken.     

基因与细胞工程大实验教学体系的建立与实践

对生物技术专业的基因与细胞工程大实验的教学体系进行了改革.构建了新的教学体系,改验证性实验为探索性实验,学生选择实验材料设计实验.实践表明,基因与细胞工程大实验课程的改革有效地提高了实验教学质量,培养了学生的实验能力和创新能力.     

肝再生

肝脏在受到损害或切除之后,可以显示出强大的再生能力。肝再生这一现象早已被人们发现并研究。肝脏再生的启动因素有哪些,又是如何适时终止的?在肝再生过程中,哪些基因的表达发生了变化?哪些信号通路参与了肝再生过程?这些问题一直是研究者关注的焦点。本文将对肝再生研究的这些热点问题进行综述。     

DNA methylation of fly genes and transposons

The use of anti-5-methylcytosine antibodies in affinity columns allowed the identification of methylated sequences in the genome of Drosophila melanogaster adults. In view of the presence of transposable elements amongst the identified sequences, it has been suggested that DNA methylation is involved in transposon control in the fly genome. On the contrary, a reanalysis of these data furnishes several intriguing elements that could raise new questions about the role that DNA methylation plays in the fly genome. The aim of the present paper is to discuss some features that emerge from the analysis of the identified methylated sequences. 企业文化评价与识别研究是管理学科的热点和难点问题.在回顾企业DNA研究应用的基础上,提出并界定了企业文化基因概念,对企业文化基因识别进行了实证研究.     

一种简便有效的PCR扩增片段直接回收克隆方法

作者提供一种简便快速的克隆方法,即直接对琼脂糖凝胶电泳分离的PCR产物进行克隆,可获得较高的阳性克隆率.结果显示,300～700 bp大小的片段阳性克隆率为70.72%,300 bp以下和大于700 bp的片段阳性克隆率分别达到60.38%和45.33%.该方法适用于含有大量不同大小DNA片段的PCR产物并需要同时对其进行回收和克隆的样品,不仅可以大大简化实验过程,也可以降低假阳性出现的机会.     

Construction and anti-tumor effects of recombinant fowlpox virus expressing Newcastle disease virus hemagglutinin-neuramidinase gene

Hemagglutinin-neuramidinase (HN), a Newcastle disease virus-derived protein, not only mediates receptor recognition but also possesses neuraminidase (NA) activity, the ability to cleave a component of those receptors, N-acetylneuraminic acid (NAcneu, sialic acid). It is known that this protein in mammalian species, including human beings, has interesting anti-neoplastic as well as immune stimulating properties. To explore the use of the HN gene in cancer gene therapy, we constructed a recombi-nant fowlpox virus expressing the HN protein (vFV-HN) and compared the anti-tumor activity of the recombinant virus with that of wild-type fowlpox virus (FPV) in vivo and in vitro. Here we found that although B16 cells were somewhat resistant to the basal cytotoxic effect of wild-type fowlpox virus, infection with vFV-HN caused a pronounced cytotoxic effect and, the survival of tumor-bearing mice immunized with vFV-HN was significantly increased compared with the survival of mice immunized with the FPV alone. Furthermore, the immunization of mice with vFV-HN elicited a B16 tumor-specific cytotoxic T lymphocyte (CTL) response and clonal expansion of both CD4 and CD8 T cell populations in vivo. In addition, T cells from lymph nodes of mice vaccinated with vFV-HN secreted high levels of the Th1 cytokine IL-2 and IFN-γ, indicating that the regression of tumor cells is related to a Th1-type dominant immune response. These results demonstrate that vaccination with vFV-HN may be a potential strategy for cancer gene therapy.     

枝状枝孢霉MD2的Unigene A03231的克隆及原核表达分析

为研究真菌紫杉醇合成相关基因的功能,从产紫杉醇真菌枝状枝孢霉MD2中克隆了Unigene A03231的DNA和c DNA全长序列(918 bp).结果表明:该基因不含内含子,其编码产物含305个氨基酸,与短链脱氢酶/还原酶一致性为64%,预测蛋白分子量为33071.5 Da,不存在信号肽,含有2个跨膜结构.Unigene A03231以单拷贝形式存在于基因组,在0~30 h内其表达水平随茉莉酸甲酯诱导表现为先升高后降低.构建了该基因的原核表达菌株,初步优化了其在大肠杆菌BL21中的诱导表达条件为:诱导温度28℃,IPTG浓度0.6 mmol/L,诱导时间8 h.为深入分析该基因的催化功能奠定了基础.
