Function of resveratrol derived from transgenic plant expressing resveratrol synthase gene

Two genes from grapevine coding for resveratrol synthase, named RS1 and RS2, were cloned by RT-PCR. AnEscherichia coli expression vector was constructed by insertion of RS1 into pBV221. A specific protein with the same molecular weight (42 ku) as the resveratrol synthase was expressed and used to prepare the rabbit antiserum. A plant expression vector was constructed by inserting the RS1 gene into pBin438 downstream of the doubled CaMV 35S promoter and TMV-Ω fragment. PCR-positive transgenic tobacco plants were obtained after transformation withAgrobacterium tumefaciens LBA4404 harboring the plant expression vector. Southern blot analysis demonstrated that the foreign gene was integrated into the tobacco genome. The results of RT-PCR and Western blot indicated that the RS1 gene was transcribed and expressed. Formation of resveratrol in transgenic tobacco was further determined by thin-layer chromatography of silica gel and HPLC. Increased accumulation of human breast adenocarcinoma cells in G₀ and G₁ phases of cell cycle was observed in cells treated with resveratrol purified from transgenic tobacco as compared to the untreated cells.     

大学生体质现状调查分析

以宁夏大学为例,对2002,2003,2004年度31384名在校大学生进行了体质测试,远用SPSS10．00统计软件对有效数据进行纵向（同年度不同年级）和横向（不同年度同年级）的方差比较分析,探讨了大学生身体形态特征、机能发育水平和身体素质状况等方面的指标和问题．从学校体育的角度剖析了大学生体质动态变化的多层面原因,并提出相关建议,为促进高校体育教育改革的深化和科学研究的发展。更好地培养新世纪所需要的高科技、有创造性、有竞争力的健康的建设人才,为制定出适合大学生增强体质、增进健康的体育教学理念、内容和方法,提供了科学的依据．     

Aromatase and steroid sulfatase mRNA expression in fine needle aspirates from benign and malignant breast disorders

The fine needle aspiration (FNA) test is a convenient and tolerable technique with minimal invasion that is accepted by most women.Local estrogen synthesis depends mainly on the aromatase and steroid sulfatase pathways that are believed to play important roles in breast carcinogenesis.However,little is known about the level of aromatase and steroid sulfatase mRNA expression in FNA samples which contain only small amounts of tissue.The nested Q-PCR assay has been proven to be a highly sensitive and specific method to assess the aromatase expression of breast tissue.In this study,aromatase and steroid sulfatase mRNA expression in 74 patients with benign or malignant disorders was evaluated and compared using nested Q-PCR and non-nested Q-PCR assays.The expression levels were analyzed and correlated with clinical parameters.No difference in the aromatase expression levels between nested and non-nested Q-PCR was noticed.Age and aromatase mRNA expression level were two independent risk factors for breast cancer (P=0.04 and P=0.00,respectively),while menopausal status and steroid sulfatase mRNA expression levels were not associated with breast cancer.This study showed that both nested and non-nested Q-PCR assays were effective methods for research using FNA breast samples.     

基于人工神经网络的火控系统故障诊断

阐述了人工神经网络的基本原理,研究分析了BP神经网络的缺陷并提出了优化策略。在此基础上,将BP神经网络应用于火控系统故障诊断之中,提出了基于此网络的火控系统故障诊断模型、实现方法和优点。     

乳腺组织的位相衬度成像

将位相村度成像用于乳腺肿瘤的结构研究后,发现:正常乳腺组织的吸收图像的清晰度比较高,原因是正常乳腺组织比较致密,对于对X射线的吸收相对较强。而位相衬度成像在分辨肿瘤组织内部结构方面的优势是比较明显的,能够清晰地显示出乳腺组织中的一些精细结构,并可以反映出正常组织、良性肿瘤组织和癌变组织结构的特异性。在位相衬度成像中,图像的村度是与样品到探测器的距离有关的,在不同样品到探测器的距离下,位相村度成像的分辨率是不同的。根据衬度传递函数(CTF),可以估计出乳腺癌组织中的钙化尺度在30μm左右。     

新型多酸化合物对人乳腺癌细胞增殖的抑制作用

采用MTT法、形态学方法和Western blot方法,检测了3种新型多酸化合物[SbW9、（SbW9）2-（SnR）4、（SbW9）2-（SnR-CH3）4]对人乳腺癌MCF7和MDA-MB-231细胞生长抑制作用,细胞形态学变化及细胞PCNA蛋白表达变化,并探讨该3种化合物抑癌的机制.实验结果表明：3种化合物对人乳腺癌细胞MCF7和MDA-MB-231的生长均具有明显的抑制作用（P〈0.01）,细胞形态发生明显变化,细胞皱缩并且增殖速度明显减慢;其中,（SbW9）2-（SnR）4可使MCF7细胞PCNA蛋白表达下降（P〈0.05）,且呈浓度剂量依赖性.说明该3种新型多酸化合物均具有抗肿瘤活性,其活性部位可能是以{SbW9}为基本建筑单元的聚阴离子,作用机制可能与其抑制细胞DNA的合成有关.     

HER-2基因特异性核酶在MDA-MB-453细胞内的瞬时效应

目的确定人表皮生长因子受体-2(HER-2)特异性锤头状核酶(hammerhead ribozyme,RZ)对雌激素受体阴性乳腺癌细胞MDA-MB-453的影响.方法应用RT-PCR、免疫细胞化学检测RZ对MDA-MB-453细胞HER-2表达的影响;MTT法检测RZ对细胞增殖的影响.结果RZ转染入MDA-MB-453细胞中,RT-PCR检测出转染RZ的MDA-MB-453细胞中HER-2 mRNA表达量明显下降;免疫细胞化学方法检测细胞内的HER-2蛋白表达下降;MTT法检测细胞增殖活性明显受抑.结论RZ在MDA-MB-453细胞内有效抑制靶基因HER-2mRNA和蛋白的表达,同时可抑制细胞增殖,有助于进一步研究雌激素受体阴性乳腺癌细胞HER-2信号转导通路.     

FTIR assessment of the secondary structure of proteins in human breast benign and malignant tissues

The compositions of the secondary structures of protein in the human breast normal, hyperplasia, fibroadenoma and invasive ductal carcinoma tissues have been estimated from the Fourier self deconvolved spectra, the second derivative spectra and the curve-fitting analysis of the amide I bands in their spectra. Some parameters of the secondary structures of proteins in these 4 types of tissues are significantly different and located in separate ranges. Fibroadenoma tissue demonstrates the most remarkable difference in the compositions of α-helical and atypical helical structures. The carcinoma tissue has the highest ratio of the intermolecular β-sheet content and the intramolecular β-sheet content. This noticeable difference in the secondary structure may have important implications for analyzing the properties and types of human breast disease.     

基于数据挖掘诊断事件方法的网络入侵检测系统

简单论述了在入侵检测系统中数据挖掘技术的应用,并结合了数据挖掘挖等新概念,根据数据挖掘技术用于事件诊断的基本思想,利用粗糙集进行数据归纳,过滤大量事件数据中的冗余属性,得到精简事件数据集,并将得到的精简数据进行分类成所需的规则,并结合实例说明了该方法的工作步骤。由实例可知该方法用于事件诊断的可行性,最后指出实际应用过程中的一些技术难题。
