重组牛肠激酶轻链在大肠杆菌中的表达和纯化

密码子优化全基因合成了牛肠激酶轻链基因(sBEKLC),采用pET-39b( )构建了融合表达载体,在大肠杆菌中进行了成功表达.37℃在含30 mg/L卡那霉素的LB培养基中培养,当细胞密度(D600)达到0.5时,降低温度到28℃,加入终浓度为0.1 mM的异丙基硫代-βD-硫代半乳糖苷诱导外源蛋白表达,继续培养6 h后收集菌体,融合蛋白(DsbA-sBEKLC)产量达到151.2mg/L,相当于80.6 mg/L sBEKLC.使用渗透冲击技术从细胞周质中提取sBEKLC,经过亲和层析可从每升发酵液中得到6.8 mg sBEKLC,经检测sBEKLC具有生物活性.     

法莫替丁对鼠胃肠肌电活动的影响

观察１２只大鼠法莫替丁对体胃肠肌电活动的影响，结果发现：腹腔注射法莫替丁５ｍｉｎ左右胃窦肌电活动开始增强，４０ｍｉｎ慢皮和快波振幅明显增大；十二指肠电活动除快经有所提高外，漫波频率、慢波和快波振幅均未见明显改变。提示法莫替丁可兴奋胃窦肌电活动     

气液色谱法测定有机溶质在碳酸丙烯酯-NaI溶液中的活度系数

<正> 本文报导了298.15、303.15、308.15、313.15K时,正已烷、苯、四氯化碳、氯仿、二氯甲烷在碳酸丙烯酯(PC)-NaI溶液中的无限稀释活度系数γ_1~∞和盐效应常数的气液色谱(GLC)法测定结果。并考察了温度的影响。     

茶油烷醇酰胺的制备及其表面活性

茶油和甲醇反应生成茶油脂肪酸甲酯，再和二乙醇胺反应得到茶油烷醇酰胺。测定了它的表面张力、发泡力、润湿力、浮化力、钙皂分散力。结果表明：茶油烷醇酰胺具有良好的表面活性。     

Induction of the synthesis of thymidine kinase by aldosterone in the kidney of the immature male rat

Summary Thymidine kinase activity was studied in kidneys from immature male rats after administration of aldosterone. Kinetic studies showed that the enzyme activity reached its maximum level 24 h after aldosterone injection. That increase was specific for aldosterone and could be related to the synthesis of new molecules of enzyme.     

氟化氢熏气对蚕豆叶片的影响（Ⅱ）

蚕豆受氟化氢污染后,叶片含氟量与硝酸还原酶活性呈显著负相关,总氮含量也明显下降.叶绿素a、b含量下降.叶片亚细胞器也受到破坏.叶片边缘和尖端出现典型的褐色坏死伤斑,造成蚕豆产量和品质下降.     

Strain differences in the pattern and intensity of wheel running activity in laboratory rats

Summary Wheel running activity rhythms of three inbred rat strains, ACI/Ztm, BH/Ztm, and LEW/Ztm, were compared in order to evaluate the effect of genetic differences on circadian rhythm parameters. Significant strain differences were found in the general pattern of the activity rhythms and their characteristic periodicities as well as in the amount and duration of wheel running activity and the timing of activity onsets and offsets. The results suggest that genetic differences exist in the coupling of the multiple circadian oscillators that generate the overall pattern of wheel running activity.     

Malignant hyperthermia: Molecular defects in membrane permeability

Summary Malignant hyperthermia (MH), a genetically inherited disorder of skeletal muscle, is due to molecular defect in membrane permeability. The alteration in membrane permeability is suggested to be due to enhanced phospholipase A₂ activity which is responsible for the increased level in sarcoplasmic Ca²⁺. The excess Ca²⁺ is responsible for muscle hyper-rigidity and enhanced rate of glycolysis, resulting in a rapid rate of lactic acid production and a low pH in MH muscle.
