转基因小鼠与肿瘤研究

转基因小鼠的出现有力地推动了肿瘤研究的发展。具有癌症倾向的转基因小鼠,是从DNA水平开展有关人类恶性肿瘤发生、生长、转移和防治研究的良好活休模型。肿瘤形成与控制细胞生长和分化的多种基因特别是癌基因、抑癌基因的遗传改变有关。在利用转基因小鼠进行实验研究时,对由相同结构基因建立的不同转基因小鼠家系在癌基因表达上的时空差异性,应当给予足够的重视。此外,实现转基因动物生产的专业化、产业化,将会加速我国生命科学领域中转基因动物应用研究的进程。     

基因工程小鼠剖宫取胎的适宜条件

目的　净化升级基因工程小鼠。方法　采用统计学方法对基因工程小鼠剖宫取胎净化过程中的大量数据进行分析。结果　①按规律准确判断妊娠天数 ,可使剖得的仔鼠成活率达到 81 0 % ,②CD_1小鼠作为代乳母鼠剖得的仔鼠离乳成活率为 5 1 5 %。结论　适宜的剖宫取胎条件是提高剖出仔鼠和离乳仔鼠成活率的关键。     

Obtaining High Pest-resistant Tobacco Plants Carrying B.t. insecticidal Gene

To increase the expression level of CryIA(c) gene in transgenic plants, a plant expression vector pBinMoBc carrying the CryIA(c) gene under control of chimeric OM promoter and Ω factor was constructed. As a control, pBinoBc carrying the CryIA(c) gene with the CaMV 35S promoter was also constructed. The vectors were transferred into tobacco plants respectively via Agrobacterium-mediated transformation. ELISA assay showed that the expression level of the CryIA(c) gene in pBinMoBc transgenic tobacco plants was 2.44-times that in pBinoBc transgenic tobacco plants, and it could be up to 0.255% of total soluble proteins. Bioassay showed that pBinMoBc transgenic tobacco plants had more notable insecticidal effect than pBinoBc transgenic tobacco plants. The above results showed that the chimeric OM promoter was a stronger promoter than CaMV 35S promoter that was widely used in plant genetic engineering, and this is very useful in pest-resistant plant genetic engineering.     

pGH基因在F_1代转基因金鱼中的整合及表达的分析

本实验对转基因金鱼进行分子生物学鉴定,所用实验材料转基因金鱼是采用显微注射的方法,将羊金属硫蛋白基因启动子与猪生长激素基因(pGH)重组而成的线形DNA片段导入金鱼的受精卵中,在合适的条件下培养获得的。为了鉴定外源基因在金鱼体内整合、表达和遗传的情况,我们选择了相同生长期的F_1代转基因金鱼6条和普通金鱼2条,进行总DNA的提取和纯化,用特异引物对pGH基因进行PCR扩增,结果表明:外源基因(pGH)在部分受体金鱼中得到了整合,并通过有性繁殖稳定地遗传给下一代。同时将阳性金鱼与阴性金鱼进行身长和体重等形态上的比较,初步断定:pGH基因在部分受体鱼中得到了表达,且有显著的促生长作用。     

Transgenic mice carrying chimeric or mutated type III intermediate filament (IF) genes

Mice carrying chimeric, truncated or mutated genes encoding intermediate filament (IF) proteins type III do not show any detectable severe pathology. However, upon (over)expression of the transgene in the eye lens all animals develop lens opacification (cataract). At the cellular level the loss of visual acuity is preceded by interference with the terminal differentiation of lens fibre cells, plasma membrane damage, distorted assembly of the IF cytoskeleton and perturbation of the cytoskeleton-membrane complex. The degree of expression is paralleled by the extent of the damages. Received 4 November 1996; accepted 15 November 1996     

小鼠转基因及传代研究

综述了湖北省农科院生物技术研究所近年来有关转基因小鼠的主要研究进展。应用原核注射技术 ,将POMT PGH、hDAF、pBHSA、pSHSA、PT HBV 1 3、Bcl xL、hCD5 9、hCD5 9+hMCP等 8种外源基因 ,注入并移植 4 874枚小鼠的受精卵 ,得到 5 5 6只小鼠 ;经PCR和Southern杂交检测 ,确认原代转基因小鼠 10 8只。基因整合率平均 19.4 % ,转基因效率平均 2 .2 %。应用混合注射的方法得到了转双基因小鼠 ,双基因共整合率 2 2 .2 %。表达外源蛋白的转基因小鼠在 5 0 %～ 10 0 %之间。研究了小鼠的超数排卵和影响转基因效率的几个因素。通过转Bcl xL小鼠与非转基因小鼠连续五个世代的传代交配和检测 ,研究了转基因小鼠外源基因的遗传规律 ,表明四只原代小鼠中 ,只有一只能稳定地将外源基因传递给后代。并非所有的转基因小鼠都具有遗传的稳定性 ,欲建立小鼠的转基因品系 ,尚需对原代转基因小鼠进行筛选。
