Recent progress in polymer-based gene delivery vectors

The gene delivery system is one of the three components of a gene medicine, which is the bottle neck of current gene therapy. Nonviral vectors offer advantages over the viral system of safety, ease of manufacturing, etc. As important nonviral vectors, polymer gene delivery systems have gained increasing attention and have begun to show increasing promising. In this review, the fundamental and recent progress of polymer-based gene delivery vectors is reviewed.     

N-terminal of L protein of vesicular stomatitis virus contains a new signal sequence

The L protein (241kD) of vesicular stomatitis virus (VSV) is the most important snbnnit of the replication complex. The existence of specific localization signal in the L protein was investigated by making recombinant constructs expressing truncated mutants of the L protein fused to green fluorescent protein (GFP) in transient transfection assays. The chimeric genes encoding varied N-terminal of L and GFP gene were put under the control of T7 promoter or CMV promoter. The fusion proteins were transiently expressed in BHK-21, COS-7, CHO or Hep G2 cells. When more than 120 residues were deleted or only 96 residues were kept on the N-terminal, the fusion proteins were shown to be distributed throughout the cells, cytoplasm and nucleus under the confocal microscope. However, other chimeric proteins with 120 or more amino acids were dotted and distributed in the perinuclear regions. And the fusion protein with 96—120 aa has the similar distribution. A thirteen-residue peptide QGYSFLHEVDKEA (108—120) was identified as localization signal, whose function would be absolutely distributed with the deficiency of D or V. Our results show that there is an independent localizing signal in N-terminal domain of L protein of VSV and this functional signal is conserved in different cell lines.     

Analysis of cDNA sequence, protein structure and expression of parotid secretory protein in pig

Parotid secretory protein (PSP) secreted abundantly in saliva, whose function is related with the anti-bacterial effect. The PSP cDNA has been isolated from pig parotid glands by 3′ and 5′ rapid amplification of cDNA end (RACE), based on the conserved signal peptide region among the known mammalian PSP. The result of homologous comparison shows that pig PSP and human PSP shares the high identity at the level of the primary, secondary and tertiary protein structure. A search for functionally significant protein motifs revealed a unique amino acid sequence pattern consisting of the residues Leu-X(6)-Leu-X(6)-Leu-X(7)-Leu-X(6)-Leu-X(6)-Leu near the amino-terminal portion of the protein, which is important to its function. RT-PCR, Dot blot and Northern blot analysis demonstrated that PSP was strongly expressed in parotid glands, but not in other tissues.     

Abnormal piezoresponse behavior of Pb（Mg1／3Nb2／3）O3-30% PbTiO3 single crystal studied by high vacuum scanning force microscopy

The piezoresponse behavior dependence of the Pb(Mg1/3Nb2/3)O3-30%PbTi03 single crystal on the vacuum degree has been investigated by scanning force microscopy in the piezoresponse mode under high vacuum. Unusual piezoresponse behavior related to the screening charges compensation mechanism is observed on the (111) crystal face. The significant piezoresponse degradation behavior with low piezoresponse signal under high vacuum is attributed to the instability of the polarization state due to the insufficient compensation of the intrinsic screening charges for the polarization charges in PMN-30%PT single crystal. In contrast, the remarkable domain contrast of the sample at ambient pressure is owing to the dominant surface screening charges deriving from surface adsorption, which plays an important role in determining the stability of the domain behavior and in achieving the optimal properties.     

Holocene lake deposits of Bosten Lake, southern Xinjiang, China

A 9.25-m-long sediment core from Bosten Lake,Xinjiang, provides detailed information about changes in the water budget and biological acticity over the last 8400 cal-endar years. The chronology is constructed from six AMS radiocarbon dates on the terrestrial plant remains. Based onanalyses of TOC, CO3, detrital compounds and biogenic SiO2,lake level fluctuations and periods of remarkably-negative water budget appeared at 8.4-8.2 cal ka, 7.38-7.25 cal ka,5.7-5.5 cal ka, 3.7-3.4 cal ka and 3.3-2.9 cal ka, respec-tively. As they are in-phase with low lake levels at Sumxl Co and Bangong Co in western Tibet Plateau and with paleo-lakes in Inner Mongolia, a climate-induced change to some-what drier and warmer conditions is inferred. A further drop in lake level after 1320 AD of about 200 yr duration may heattributed to a negative water balance prior to the main phase of the Little Ice Age. Deep and stable lake phases of 1500 yr and 1800 yr duration at 7.2-5.7 cal ka and 5.5-3.7cal ka coincide with maximum moisture during the Holocene Megathermai in China. The long term trend towards aridity since about 4.3 cal ka can dearly be recognised. The reduced water budget of Bosten Lake from 640-1200 AD may be attributed to local effects.

Triassic U-Pb age for zircon from granites in the Tonghua area and its response to the Dabie-Sulu ultrahigh-pressure collisional orogenesis

Single-grain zircon U-Pb dating was carried out to constrain the emplacement timing of granitic plutons at Chaxinzi, Xiaoweishahe and Longtou in the Tonghua area, south of Jilin Province. The results show that these plutons formed in the Triassic with ages of 203—217 Ma. Geological and geochemical characteristics indicate that the plutons are composed of quartz diorite and granite. The former was derived from partial melting of mafic lower crust, whereas the latter originated from thickened crust with garnet as the residue in the source. It appears that protoliths of these two types of granitits are different although they have the same emplacement age. Considering that these plutons are petrologically different from the coeval granites in the Xingmeng (Xing‘an-Mongolian) to Jihei (Jilin-Heilongjiang) orogenic belt in the north, it is suggested that their formation was related to the Dabie-Sulu ultrahigh-pressure collisional orogenesis since their ages are only 10—20 Ma younger than timing of the ultrahigh-pressure metamorphism, but comparable to that of the first rapid exhumation of the ultra-high-pressure metamorphic rocks and the emplacement of the post-collisional granites.     

Time-division phase modulated single-photon interference in a Sagnac interferometer

We introduce a stable, long-distance singlephoton Sagnac interferometer, which has a balanced configuration to efficiently compensate phase drift caused by change of the fiber-optic path. By using time-division phase modulation, single-photon interference was realized at 1550nm in a 5-km-long as well as 27-km-long Sagnac fiber loops,with a fringe visibility higher than 90% and long-term stability. The stable performance of the single-photon interference indicated that the time-division phase-modulated Sagnac interferometer might readily lead to practical applications in single-photon routing and quantum cryptography.     

Effects of hyaluronic acid－chitosan－gelatin complex on the apoptosis and cell cycle of L929 cells

With the development in the field of tissue engineering, the interaction between biomaterials and cells has been deeply studied. Viewing the cells seeded on the surface of materials as an organic whole, cell cycle and apoptosis are analyzed to deepen the study of cell compatibility on biomaterials, while cell proliferation and differentiation are studied at the same time. In this paper, hyaluronic acid is incorporated into the chitosan-gelatin system. Propidium iodide (PI) was used in cell cycle analysis and the double-staining of cells with annexin-V and PI was applied in cell apoptosis analysis. The results show that incorporated hyaluronic acid shortens the adaptation period of cells on the material surface, and then cells enter the normal cell cycle quickly. In addition, added hyaluronic acid inhibits cell apoptosis triggered by the membranes. Therefore, hyaluronic acid improves the cell compatibility of chitosan-gelatin system and benefits the design of biomimetic materials.
