A BAC clone of MDV strain GX0101 with REV-LTR integration retained its pathogenicity

The complete genome of Marek＇s disease virus （MDV） strain GX0101, which was integrated with the LTR sequences of REV, was cloned in Escherichia coli as a bacterial artificial chromosome （BAC）. BAC vector sequences were introduced into the US2 locus of the MDV genome by homologous recombination. The viral DNA containing the BAC vector was used to transform Escherichia coli strain of DH10B. Then the recombinant virus was successfully rescued by transfection of the recombinant BAC DNA into primary chicken embryo fibroblast （CEF）. This BAC viral clone was named bac-GX0101. When the reconstituted virus was inoculated into 1-day-old birds, visceral tumors could be detected as early as 62 d post infection. There was no difference in growth ability and pathogenicity to birds between the BAC derived virus and its parental virus. The BAC derived virus maintained its oncogenicity and immunosuppressive effects. In conclusion, the complete genome of GX0101 strain was successfully cloned into BAC and the infectious clone was rescued. With the powerful BAC manipulation system, the infectious clone will provide a useful tool for further understanding the functional roles of the inserted REV-LTR sequence in the GX0101 strain of MDV,     

Subtyping of type A influenza by sequencing the variable regions of HA gene specifically amplified with RT-PCR

The outbreak of a novel influenza A(H1N1) virus across the globe poses a threat to human health.It is of paramount importance to develop a rapid,reliable and inexpensive diagnostic procedure.Based on the bioinformatic information from public database,primers specific for influenza A virus surface protein haemagglutinin(HA) of several subtypes(including H1,H2,H3,H5,H7 and H9) were designed.Primer-specific PCR products were subjected to sequencing for accurately distinguishing H1 and H3 subtypes from others.T...     

2株甲型肝炎病毒蛋白酶2A核苷酸序列的测定与比较

从甲型肝炎病毒２个不同株感染的ＫＭＢ１７细胞中提取总ＲＮＡ,经ＲＴ－ＰＣＲ特异性扩增出此２毒株病毒蛋白２Ａ基因,将２个２Ａ基因克隆到ｐＧＥＭ－Ｔ载体上,经ＤＮＡ序列测定,得到２个２Ａ基因的核心苷酸序列,序列比较发现它们及与ＨＡＶ野毒株相对应序列有突变存在,Ｈ２株与ＨＭ１７５相比有一个点突变,同源性为９９．７２％;与ＨＭ１７５比较,Ｈ２株和Ｌ８株第３１９７位核苷酸均由Ａ突变为Ｇ,相应的氨基酸由丝氨酸     

There is the second virus that causes tobacco leaf curl disease (not TbLCV-CHI) in the field

Sequence analysis of virus isolation DNA of tobacco leaf curl disease shows that there is the second geminivirus (not Chinese Tobacco Leaf Curl Virus, TbLCV-CHI) that causes tobacco leaf curl disease in the field in the Guangxi Zhuang Autonomous Region, China. This virus DNA-A contains 2 734 nt. Large intergenic region (LIR) contains 269 nt, the virus sense strand contains 2 open reading frames (ORFs): AV1 (115 aa) and AV2 (coat protein gene, CP, 256 aa), and the complementary sense strand contains 4 ORFs: AC1 (replicase gene, 361 aa), AC2 (transactivator, 134 aa), AC3 (134 aa) and AC4 (97 aa). The virus belongs to one kind of subgroup III geminiviruses from old world, and could be the Chinese tomato yellow leaf curl virus (TYLCV-CHI).     

Construction of cDNA library, nucleotide sequence and analysis of entire genome of classical swine fever virus strain Shimen

According to the previously published CSFV sequences, 18 pairs of primers have been designed and synthesized, which cover the entire genome of CSFV strain Shimen. Each cDNA fragment has been amplified by RT-PCR from the anticoagulant blood of strain Shimen infected pig. The PCR products have been cloned respectively and sequenced. Results show that the cDNA library of strain Shimen and its nucleotide sequence have been obtained. The genomic RNA of strain Shimen is 12 298 nucleotides in length, containing a 5′ and a 3′ noncoding region 373 and 231 nt long respectively. The center of genome is a single large open reading frame of 11 697 nt which encodes a polyprotein of 3 898 amino acids. The entire sequence of strain Shimen has also been compared with that of other CSFV strains.     

玉米矮花叶病毒原的生物学鉴定

对我国西北的玉米矮花叶病毒Ｂ株系（ｍａｉｚｅ ｄｗａｒｆ ｍｏｓａｉｃ ｖｉｒｕｓ－Ｂ,ＭＤＭＶ－Ｂ）进行了系统的鉴定。ＭＤＭＶ－Ｂ侵染玉米（Ｚｅａ ｍａｙｓ）、高粱（Ｓｏｒｇｈｕｍ ｖｕｌｇａｒｅ）和狗尾草（Ｓｅｔａｒｉａ ｖｉｒｉｄｉｓ）等禾 本科杂草,但不侵染普通烟（Ｎｉｃｏｔｉａｎａ ｔａｂａｃｕｍ）、心叶烟（Ｎｉｃｏｔｉａｎａ ｇｌｕｔｉｎｏｓ）、苋色藜（Ｃｈｅｎｏｐｏｄｉｕｍ ａｍａｒ     

鸡传染性法式囊病毒VP2原核表达蛋白的纯化和复性

对原核表达的鸡传染性法式囊病毒(IBDV)VP2蛋白进行可溶性分析与不可溶性分析,结果表明蛋白主要以包涵体形式存在,利用溶菌酶、Triton-100、尿素等试剂进行纯化和复性后,对复性前后的蛋白分别与特异性多抗进行Dot-ELISA,复性后蛋白的反应活性增强了10倍,对复性后的蛋白与IBDV16株单抗进行Dot-ELISA,其中有11株与其发生特异性反应.     

计算机网络病毒传播的概率模型

研究网络拓扑对病毒传播的影响.提出在双星图上计算固定概率(病毒成功蔓延整个网络的概率)的教值方法,并成功将其转化为求解线性方程组的问题.通过仿真发现:1)双星网络越不平衡,病毒越容易在其中蔓延;2)让网络规模趋于无穷大,固定概率收敛.固定概率还可以简单地作为判断双星网络是否安全可信的参考指标.
