A novel salicylaldehyde dehydrogenase involved in catabolism of naphthalene from Pseudomonas putida ND6, NahV, has been identified. NahV exhibited lower identity in amino acid sequence with the classic salicylaldehyde dehydrogenase, NahF, from P. putida ND6. This is the first report of an isofunctional enzyme of bacterial salicylaldehyde dehydrogenase. Comparison of Km and Vmax values of NahV and NahF demonstrated that NahF has a more efficient catalytic reaction than NahV, while NahV has much higher affinity for salicylaldehyde and NAD^+. Both enzymes exhibited broad substrate specificities and catalyzed the oxidation of salicylaldehyde, 5-chlorosalicylaldehyde, formaldehyde, m-nitrobenzaldehyde, o-nitrobenzaldehyde, o-methoxybenxaldehyde, glutaraldehyde, caprylic aldehyde, and glyoxal. However, the relative rates at which the substituted analogs are transformed differ considerably. NahV activity could be enhanced by Fe^2+, Cu^2+ and Zn^2+; whereas NahF activity could only be stimulated by Fe^2+, NahF is more stable than NahV at elevated temperatures. Dot-blot hybridization analyses showed that nahF-like genes occurred in all naphthalene-degradation bacteria isolated in this study, whereas nahV-like genes were present in only some naphthalene-degrading bacteria.  相似文献

Summary In gerbil adrenal cortex the activity of intramitochondrial NADP-linked isocitric dehydrogenase (IDH) is up to 10-fold greater than the NAD-linked IDH. The NADP-IDH, apparent Km 0.58 mM, Vmax 280 nmoles/min/mg mitochondrial protein, appears to be the major source of reducing equivalents to support adrenal mitochondrial steroid 11B- and 19-hydroxylation in this species.  相似文献

Summary Pheromone perception requires rapid enzymatic degradation of the active chemical signal in the sensory hairs. Three insects are used to illustrate chemical approaches to studying the degradation of pheromones by antennal enzymes. First, hydrolysis of acetate and haloacetate esters is examined in the diamondback moth,Plutella xylostella. Second, aldehyde oxidation and the selective inhibition of the oxidase and dehydrogenase activities are described for the tobacco budworm moth,Heliothis virescens. Finally, a stereochemical analysis of the epoxide-hydrase catalyzed addition of a water molecule to the oxirane ring of disparlure is described for the gypsy moth,Lymantria dispar.  相似文献

Summary NAD pyrophosphorylase (ATP:NMN adenylyltransferase) activity has been measured in the skeletal muscle of dystrophic mice. The amount of this enzyme in the dystrophic mice, as determined by three different methods, was about one half of that in the controls. In addition, the concentration of ATP was too low to be detected in crude extracts of dystrophic mouse skeletal muscle, which were prepared using Tris buffer alone or Tris buffer containing either 3 M KCl, or 1 mM PMSF.  相似文献

Objective: To explore the relationship between genetic polymorphisms in methylenetetrahydrofolate reductase (MTHFR), methionine synthase reductase (MTRR), the central enzymes in folate metabolism that affects DNA methylation and synthesis, and the risk of Down syndrome in China. Methods: Genomic DNA was isolated from the peripheral lymphocytes of 64 mothers of children with Down syndrome and 70 age matched control subjects. Polymerase chain reaction and restriction fragment length polymorphism were used to examine the polymorphisms of MTHFR 677C→T, MTRR 66A→G and the relationship between these genotypes and the risk of Down syndrome was analyzed. Results: The results show that the MTHFR 677C→T polymorphism is more prevalent among mothers of children with Down syndrome than among control mothers, with an odds ratio of 3.78 (95% confidence interval (CI), 1.78-8.47). In addition, the homozygous MTRR 66A→G polymorphism was independently associated with a 5.2-fold increase in estimated risk (95% CI, 1.90-14.22). The combined presence of both polymorphisms was associated with a greater risk of Down syndrome than the presence of either alone, with an odds ratio of 6.0 (95% CI, 2.058-17.496). The two polymorphisms appear to act without a multiplicative interaction. Conclusion: MTHFR and MTRR gene mutation alleles are related to Down syndrome, and CT, TT and GG gene mutation types increase the risk of Down syndrome.  相似文献

Summary Three anti-horse liver alcohol dehydrogenase (HLADH) monoclonal antibodies are described. Two are specific for ADH and cross-react with class I and II enzymes from mouse, horse and Chinese hamster. They are specific for the native enzyme but do not inhibit enzyme activity except when combined at high concentration. The third antibody was isolated as a response to rabbit metallothionein. 溶氧对谷氨酸棒杆菌发酵产谷氨酸代谢的影响   总被引:1,自引:0,他引:1  
分别控制0、5%、30%3种溶氧水平进行谷氨酸分批发酵,考察了3种溶氧水平下谷氨酸棒杆菌发酵的代谢响应。结果表明,随着溶氧降低,发酵液中柠檬酸和α-酮戊二酸的含量降低,三羧酸(TCA)循环还原臂途径中的有机酸含量增加,丙氨酸、缬氨酸、苯丙氨酸含量也有所增加,在溶氧为0时,乳酸和乙酸大量积累。通过分析不同溶氧水平下的相关酶活和胞内氧化还原状态,发现随着溶氧降低,谷氨酸脱氢酶酶活降低,乳酸脱氢酶酶活升高,胞内氧化还原电势升高,它们的共同作用使氧限制条件下发酵的代谢流流向TCA循环的还原臂途径和乳酸合成途径,导致谷氨酸产量下降。  相似文献
