Molecular evolution of connective tissue growth factor in Cyprinidae (Teleostei: Cypriniformes) 总被引:1,自引:0,他引:1
Connective tissue growth factor (CTGF) plays an important role in regulation of cell growth, differentiation, apoptosis and individual development in animals. The study of sequences variation and molecular evolution of CTGF gene across various species of the cyprinid could be helpful for understanding of speciation and gene divergence in this kind of fish. In this study, 19 novel sequences of CTGF gene were obtained from the representative species of the family Cyprinidae using PCR amplification, cloning and sequencing. Phylogenetic relationships of Cyprinidae were reconstructed by neighbor-joining (NJ), maximum parsimony (MP), maximum likelihood (ML) and Bayesian method. Oryzias latipes from the family Cyprinodontidae was assigned to be the outgroup taxon. Leuciscini and Barbini were clustered into the monophyletic lineages, respectively, with the high nodal supports. The estimation of the ratio of non-synonymous to synonymous substitution (dN/dS) for the various branches indicated that there stood the different evolution rates between the Leuciscini and the Barbini. With the ratio of dN/dS of the Leuciscini being lower than that of the Barbini, species within the Barbini were demonstrated to be subjected to the relatively less selection pressure and under the relaxable evolution background. A 6 bp indel (insertion/deletion) was found at the 5' end of CTGF gene of Cyprinidae, and this 6 bp deletion only appeared in the Leuciscini, which is a typical characteristic of the Leuciscini and provides evidence for the monophylogeny of the Leuciscini. For the amino acid sequences of CTGF protein, the most variations and indels were distributed in the signal region and IGFBP region of this protein, implying that these variations were correlated with the regulation of the CTGF gene expression and protein activity. 相似文献

In order to understand the genomic changes during evolution of hexaploid wheat, two sets of synthetic hexaploid wheat from hybridization between maternal tetraploid wheat (AABB) and paternal diploid goat grass (DD) were used for DNA-AFLP and single strand conformation polymorphism (SSCP) analysis to determine the genomic and genic variation in the synthetic hexaploid wheat. Results indicated that more DNA sequences from paternal diploid species were eliminated in the synthetic hexaploid wheat than from maternal tetraploid wheat, suggesting that genome from parental species of lower ploidity tends to be eliminated preferentially. However, sequence variation detected by SSCP procedure was much lower than those detected by DNA-AFLP, which indicated that much less variation in the genic regions occurred in the synthetic hexaploid wheat, and sequence variations detected by DNA-AFLP could be derived mostly from non-coding regions and repetitive sequences. Our results also indicated that sequence variation in 4 genes can be detected in hybrid F1, which suggested that this type of sequence variation could be resulted from distant hybridization. It was interesting to note that 3 out of the 4 genes were mapped and clustered on the long arm of chromosome 2D, which indicated that variation in genic sequences in synthetic hexaploid wheat might not be a randomized process. 相似文献

Electromagnetic signals in deep reservoir are very weak so that it is difficult to predict about the presence of hydrocarbon in seabed logging(SBL) environment.In the present work,Mn0.8Zn0.2Fe2O4 nanoferrites were prepared by a sol–gel technique at different sintering temperatures of450 °C,650 °C and 850 °C to increase the strength of electromagnetic(EM) antenna.XRD,FESEM,Raman spectroscopy and HRTEM were used to analyze the phase,surface morphology and size of the nanoferrites.Magnetic properties of the nanoferrites were also measured using an impedance network analyzer.However,nanoferrites sintered at 850 °C with initial permeability of 200 and Q factor of 50 were used as magnetic feeders with the EM antenna.Lab scale experiments were performed to investigate the effect of magnetic field strength in scale tank.SPSS and MATLAB softwares were also used to confirm the oil presence in scale tank.It was observed that the magnitude of the EM waves for the antenna was increased up to 233%.Finally,the correlation values also show 208% increase in the magnetic field strength with the presence of the oil.Therefore,antenna with Mn0.8Zn0.2Fe2O4 nanoferrites based magnetic feeders can be used for deep water and deep target hydrocarbon exploration. 相似文献

天然岩体在温度、水分变化的基础上发生冻融循环,引起岩石微观损伤、冻融碎裂以及破坏失稳等劣化现象,是中西部寒冷地区岩体的主要风化过程。将取自云冈石窟砂岩分成干燥组、饱水组、饱和度对比组进行冻融试验研究,得到试验前后云冈石窟砂岩吸水率、纵波波速、单轴抗压强度等变化参数。结果表明:砂岩饱和度30%时,变化参数基本不受冻融作用影响;砂岩饱和度为30%~70%时,变化参数开始受冻融作用影响;砂岩饱和度70%时,变化参数出现突变现象。定义饱和S=70%为云冈石窟砂岩冻融损伤的"阈值"。 相似文献

液化天然气(liquefied natural gas, LNG)在充装和储运过程中,密度差分层现象易导致翻滚行为,对LNG储罐安全造成极大威胁。当初始密度差小于临界密度差时,储罐内相邻两层LNG的混合过程较平稳;当初始密度差大于临界密度差时,LNG翻滚强度显著增大,混合过程趋于剧烈。因此,研究不同罐容储罐的翻滚现象与临界密度差之间的联系尤为重要。采用商业数值软件FLUENT分别对5 000、10 000、30 000、50 000、80 000、100 000、120 000、160 000 m3储罐内分层LNG的翻滚现象进行数值分析,获得多罐容LNG储罐内的翻滚现象随初始密度差的变化规律,并确定各罐容的临界密度差范围。研究发现:LNG翻滚流速随着分层初始密度差的增加而增加,初始密度差是影响LNG翻滚的直接原因;同时,LNG翻滚的最大速度与初始密度差呈线性关系,即:当初始密度差越大,翻滚时最大速度越大,表征翻滚越剧烈,该线性关系式中的系数并非定值,其大小与储罐高度和直径等尺寸参数相关;此外,LNG发生翻滚时存在临界密度差,与储罐直径满足二次方函数关系,临界密度差... 相似文献
