Matrix metalloproteinase-26 (MMP-26, endometase and matrilysin-2), a novel member of the MMPs family, is detected not only
in the placenta and uterus, but is widely expressed in malignant tumors from different sources as well as in diverse tumor
cell lines. However, the function of MMP-26 in the reproductive system has never been reported. Expression of MMP-26 in mouse
embryos and the function of the MMP-26 antibody during mouse embryo implantation was examined for the first time by injecting
the uterine horn, immunohistochemistry,in situ hybridization, co-culture of mouse blastocysts and uterine monolayer epithelial cells, Western blot, RT-PCR, Northern blot
and zymography. Our results show that there is strong expression of MMP-26 mRNA and protein in the mouse embryo. Furthermore,
the MMP-26 antibody dramatically inhibited mouse embryo implantation and significantly inhibited adhesion and outgrowth of
mouse blastocysts onin vitro uterine monolayer epithelial cells. At the same time, the MMP-26 antibody inhibited the expression of integrin αV mRNA and
protein in a dose-dependent manner. These data suggest that MMP-26 may play a role in some of the tissue-remodeling events
associated with the invasion of the endometrium by trophoblast cells and facilitate successfully embryo implantation. 相似文献

以N6为基本培养基,对四川栽培小麦品种、品系以及农家品种共45个基因型的幼胚离体培养,进行了植株再生能力变异性的研究。结果表明,供试材料间的形态发生能力存在明显变异,表现为器官建成速度明显不同,芽器官和根器官再生率存在极显著差异,分别变异在15.00%-95.45%和0-95.45%之间,由每个幼胚外植体起源的愈伤组织再生出1.0cm以上的芽数及其芽长均存在极显著差异。 相似文献

对果蝇胚胎低表达和高表达水平基因内含子的序列结构进行分析,发现2种表达水平的基因内含子序列特征有明显差异.高表达基因的内含子一般比低表达基因的长,其中高表达基因第1内含子的平均长度是低表达基因的2.62倍,第2内含子的平均长度是低表达基因的1.79倍.两类基因第1内含子中的CpG岛含量最高,并且高表达基因内含子中CpG岛含量要高于低表达基因.此外,与低表达基因相比,TATA box、CAAT box和GC box在高表达基因内含子中出现的频数明显要高些,尤其是在第1内含子中.作者还提取出果蝇胚胎2种表达水平基因第1内含子中高频出现的6-mer简单重复序列,发现一些重复序列与实验得到的转录因子结合位点相符合.这些结果提示内含子特别是第1内含子有可能调控果蝇胚胎基因的转录从而影响基因的表达水平. 相似文献

A large number of experiments about induction of callus, embryo and germination of regeneration plants were done in soybean anther culture. Screening for genotypes, low_temperature pretreatment for floral buds, constituent of hormone in medium and effects of different temperatures on induction rate of callus and embryo were studied. It was found that the TDZ played an important role in embryo induction and germination. 28 genotypes had reaction competent to the anther culture in all 110 genotypes. The inherited -hemoglobinopathies (sickle cell disease and thalassemia) are the result of a mutation in the adult () globin gene. The fetal globin chain, encoded by the globin genes, can substitute for the mutated or defective globin chain, but expression of the globin gene is developmentally inactivated prior to birth. Reinducing expression of the normal fetal globin genes is a preferred method of ameliorating sickle cell disease and the thalassemias. Stimulation of as little as 4–8% fetal globin synthesis in the bone marrow can produce >20% fetal hemoglobin in the peripheral circulation, due to enhanced survival of red blood cells containing both sickle and fetal hemoglobin, compared to those containing sickle hemoglobin alone. Butyric acid and butyrate derivatives are generally safe compounds which induce fetal hemoglobin production by stimulating the promoter of the fetal globin genes. An initial trial with the parent compound, delivered as Arginine Butyrate, has demonstrated rapid stimulation of fetal globin expression to levels that have been shown to ameliorate these conditions. Phase 1 trials of an oral butyrate derivative with a long plasma half-life have just begun. These agents now provide a specific new apporach for ameliorating these classic molecular disorders and merit further investigation in larger patient populations. 相似文献

Lysophospholipids have long been recognized as membrane phospholipid metabolites, but only recently has their role as intercellular
signaling molecules been appreciated. Two of the best-studied lysophospholipids, LPA and S1P, signal through cognate G-protein-coupled
receptors to activate many well-known intracellular signaling pathways, leading to a variety of biologically important cell
responses. Lysophospholipids and their receptors have been found in a wide range of tissues and cell types, indicating their
importance in many physiological processes, including reproduction, vascular development, cancer and nervous system function.
This article will focus on the most recent findings regarding the biological functions of lysophospholipids in mammalian systems,
specifically as they relate to health and disease.
