In the development of transgenicBt + CpTI cotton cultivars, one male and female sterile mutant has been found in a homozygous T4 strain in our laboratory. The mutant plant, as well as its leaves, buds and flowers, is only 1/2–1/3 as large as that of
the wild transgenicBt + CpTI bivalant cotton plants. Cytological observation found that the chromosome number of the mutant is 2n = 52; however, there
are 4–8 univalents observed in meiosis I of pollen mother cells. Laboratory bioassay indicated that the mutant was highly
resistant to bollworm as the wild plants. PCR amplification revealed thatBt andCpTI genes in the mutant were still intactly inserted. However, small deletion of flanked area had been observed in the mutant
by Southern blotting analysis. So it is proposed that the mutant phenotype might result from either the DNA deletion or T-DNA
transferring in plant genome. No such report has been presented that the rearrangement of chromosome structure in a homozygous

New approaches of making single chain Fv antibodies against O6-methyl-2′-deoxyguanosine (O6MdG) have been demonstrated by using the phage antibody display system. Using O6MdG as an antigen, 21 positive clones were identified by ELISA from this library, one of which, designated H3, specifically binds to O6MdG with high affinity. The H3 scFv antibody has an affinity constant (Kaff) of 5.94×10O11(mol/L)-1. H3 scFv has been successfully used to detect O6MdG in DNA hydrolyses from yeast or E. coli cells treated with a DNA methylating agent. To our knowledge, this is the first report of the selection of a specific scFv against DNA adducts. The results demonstrate the potential applications of the phage display technology for the detection of DNA lesions caused by mutagens and carcinogens. 相似文献

Accelerator mass spectrometry (AMS) is an ultrasensitive measure for tracing14C labeled moleculesin vivo or detecting the biomarker for assessment of carcinogenesis. In this review, basic principles, wide applications and new
progresses of14C-bio-AMS are presented. It has been a new advanced tool for measuring the adduction of biologcial molecules with xenobiotics,
including carcinogens, drugs, agrochemicals, nicotine, etc. The successful applications have proven the effectiveness of AMS
to assessing cancer risk, screening drug toxicity and studying nutrients uptake. In particular, AMS is characterized by measuring
xenobiotics at very low dose levels relevant to human environmental exposure. It is sensitive and precise to an attomole (10−18 mole) or less of14C per mg carbon. Although it has some shortcomings, undoubtedly, AMS possesses an evident merit of high sensitivity and will

CpG DNA is DNA sequence that has immune stimulatory effects. Several lines of investigation over the past few years indicate that CpG DNA plays an important role in the induction of immune responses to DNA vaccines. In this study, CpG DNA-containing synthetic oligodeoxynucleotide (CpG-ODN) was cloned into the eukaryotic expression plasmid encoding a fusion protein containing b- galactosidase from E. coli and immunogenic epitopes of foot- and-mouth disease virus (FMDV) type O, and the immune responses induced by the plasmid were assayed. The results showed that guinea pigs immunized with the recombinant plasmid containing CpG-ODN generated a higher level of FMDV-neutralizing antibody and a stronger T cell proliferative response and protection against viral challenge than those receiving the plasmid containing no CpG-ODN. Our study demonstrated that it is an effective route to enhance the efficacy of DNA vaccines by inserting exogenous CpG DNA into the plasmids, and the DNA vaccine developed here is a promising candidate to prevent FMDV infection. 相似文献

采用RAPD技术分析了16个球孢白僵菌菌株的遗传分化。由160个随机引物中筛选出27个引物对各菌株进行了PCR扩增。结果表明从16个 孢白僵菌菌株中共获301个RAPD标记,其中多态性标记288个,占95.7%,表明菌株间具有较丰富的遗传多态性,聚类分析把16个球孢白僵菌菌株分为三大类型(1)Blh,,Bgd和Bpy;(3)By7,F-263,RECBb01,B12和B13;(3)B7,B11,Bxs,B6,By2,Bpc和Bzs。菌株DNA多态性与采集地之间有一定的相关性,但与寄主来源,孢子形态和对松墨天牛幼虫的毒力间未表现出明显的相关性。 相似文献
