将透明颤菌(Vitreoscilla)血红蛋白基因(vgb)插入几丁质酶基因表达质粒中,然后转入大肠杆菌(Escherichia coli)内,研究vgb基因的表达对几丁质酶基因表达和产物分泌的影响.结果表明,在不同的通氧条件下,vgb基因的表达产物均能不同程度地促进细胞的生长,而且发酵上清液的几丁质酶活性也有提高;若溶氧浓度越低,则效果越明显,可比对照提高达12.1%.同时,由于vgb在大肠秆菌的表达,还可进一步促进几丁质酶在大肠杆菌中的分泌。 相似文献

This presentation introduces the advances in biological nitrogen fixation research abroad, in particular,describes the great progress and achievements on its research in China as follows: collection of rhizobial resources and establishment of the largest database of Rhizobium in China, correction and development of Rhizobium taxonomy in international; discovery of a couple of nif genes, identification and unification of linkage among the nif gene operons of Klebsiella pneumoniae, finding of regulative mechanism of positive regulation nif gene and its sensitivity to oxygen,temperature; finding of the activity of nodulation gene nodD3 product in Sinorhizobium meliloti which is not controlled by flavonoid produced from its host alfalfa; finding of the association between expression of genes coding the products for carbon utilization and nitrogen metabolism and their regulations; chemical synthesis of nodulation factor of Sinorhizobium meliloti; constructions of engineered nitrogen fixers and utilization in practice based on the research of gene expression and regulation; chemical simulation of the structure and function of nitrogenase and bringing forward the model of nitrogenase active center for the first time in international and synthesis of model compounds which were paid attention by colleagues abroad. 研究建立起一套简单、高效的辣椒遗传转化系统。利用根癌农杆菌介导带柄子叶,将豇豆胰蛋白酶抑制剂(CpTI)基因转入辣椒栽培品种“益都羊角椒”中。对获得的33株卡那霉素(Km)抗性植物进行PCR检测,证明有6株为转CpTI基因植株。通过对转基因植株进行抗虫性研究,发现R1代植株对棉铃虫表达出一定的抗性,但不同转化体之间其抗性存在着差异。 相似文献

The differentiation process of round spermatids to spermatozoa during the late stage of spermatogenesis is called spermiogenesis. To explore spermiogenesis-related genes, cDNA microarray was used to study expression patterns of 1176 genes in pachytene spermatocytes, round spermatids and elongating spermatids of Balb/c mice. The results showed that 208 genes were detected in all the three cell types. Most of them were down-regulated from pachytene spermatocytes to round spermatids and elongating spermatids. However, up-regulation of 7 genes expression in round spermatids and 3 genes in elongating spermatids were found. Expression of 7 differentially expressed genes in cDNA arrays was further confirmed by semi-quantitative RT-PCR study. The RT-PCR results indicated that the expression of 6 genes was consistent with that in cDNA arrays, only one gene did not show differential expression by RT-PCR. These results may provide important clues for studying of expression, regulation, and function of spermiogenesis-related genes. 相似文献

It was demonstrated that parental gene expression of maize in hybrid genetic background was different from that in their inbred
lines. RT-PCR revealed a cDNA overexpressed in hybrid maize. The cDNA was reamplified and used to screen a corresponding cDNA
library. A full length of a cDNA insert ZH01 was isolated and shown to have an open reading frame encoding 414a. a. Sequence
comparison shows that ZH01 has extremely high similarity with the previously reported plant initiator factor elF-4A and distinct
difference from another maize elF-4A gene MIF4A. Ovcrexpression of ZH01 in hybrid may involve vigorous growth of hybrid maize
