Role of Cys221 and Asn116 in the zinc-binding sites of the Aeromonas hydrophila metallo-β-lactamase

The CphA metallo--lactamase produced by Aeromonas hydrophila exhibits two zinc-binding sites. Maximum activity is obtained upon binding of one zinc ion, whereas binding of the second zinc ion results in a drastic decrease in the hydrolytic activity. In this study, we analyzed the role of Asn116 and Cys221, two residues of the active site. These residues were replaced by site-directed mutagenesis and the different mutants were characterized. The C221S and C221A mutants were seriously impaired in their ability to bind the first, catalytic zinc ion and were nearly completely inactive, indicating a major role for Cys221 in the binding of the catalytic metal ion. By contrast, the binding of the second zinc ion was only slightly affected, at least for the C221S mutant. Mutation of Asn116 did not lead to a drastic decrease in the hydrolytic activity, indicating that this residue does not play a key role in the catalytic mechanism. However, the substitution of Asn116 by a Cys or His residue resulted in an approximately fivefold increase in the affinity for the second, inhibitory zinc ion. Together, these data suggested that the first zinc ion is located in the binding site involving the Cys221 and that the second zinc ion binds in the binding site involving Asn116 and, presumably, His118 and His196.Received 3 March 2003; received after revision 4 August 2003; accepted 25 August 2003     

一类时滞方程的适定性

通过设立半群的方法，研究了形如：x＇（t）=A0x（t）＋∑i=1^pAix（t—hi），t≥0的一类时滞方程的解的适定性，其中A0是Banach空间X上解析半群{e^A0t，t≥0}的无穷小生成元，Ai（i=1，2，…，p）均为（γ—A0）^α-相对有界线性算子，其中0〈α〈1，γ〉ω0（A0）为解析半群{e^A0t，t≥0}的增长界。     

基于ARM7和uClinux系统的USB主机控制器的设计与实现

针对嵌入终端产品中大多不具备USB功能,提出了一种基于以ARM7处理器S3C44B0X和USB主控芯片CY7C67200为核心的可扩展设计方案,并结合uClinux操作系统给出了相应的软硬件设计方法,通过实际测试分析了该方案的性能及可行性。     

弱C_0–半群拓扑

利用有界线性算子半群及连续线性泛函,引入了一新的局部凸向量拓扑,并对其基本性质进行了讨论.     

SBA-15-SO3H的直接合成及吸附二苯并噻吩的初步研究

采用直接合成法制备了含磺酸基的介孔材料SBA-15-SO3 H,用XRD、元素分析法、酸碱滴定法、FT-IR、TGA、N2吸附-脱附对样品进行了表征.结果表明,制备的样品具备了较好的孔道结构和较高的热稳定性,并且成功地键合了一定量的质子酸中心-SO3H,介孔表面磺酸基的键合量达到了1.09 mmol/g;SBA-15-SO3H具有较高的Ag 交换率,在正辛烷和二苯并噻吩(DBT)组成的汽油模拟体系中,吸附载体用量不变的情况下,DBT的初始浓度为4000 mg/L时,测得SBA-15-SO3Ag对硫吸附量达到了12.15 mg/g.     

15届世锦赛中国男篮与对手的技战术统计分析

对中国男篮与竞赛对手在第15届世锦赛的6场比赛中的各项技术指标进行对比分析。结果表明，处于风格转换期的中国男篮内线优势尚未建立起来，却失去了小快灵的传统特点；进攻点单一，过分依赖姚明，外线实力不足，内外脱节；防守的伸缩性、及时性不够，不能适应高强度攻守对抗。     

多氯联苯醚(PCDEs)的QSPR研究

以类原子的形式处理多氯联苯醚苯环上的基团(CH和CCl),构建分子的距离矩阵,以分子基团的均衡电负性对原子进行着色建立行矩阵,结合分子的距离矩阵和行矩阵,同时考查分子的相互间的作用情况,建立分子的结构参数——拓扑距离电负性指数.通过计算化合物的拓扑距离电负性指数(TDEI),以107种PCDEs理化性质的实验值为建模样本,建立了PCDEs的定量结构/性质相关(QSPR)模型,相关性良好.最后对107种PCDEs的三个理化性质进行了预测.     

Fe0对活性艳兰K-3R的还原降解效果

研究了活性染料活性艳兰K-3R在Fe0-H2O/O2,Fe0-H2O/N2,Fe0-H2O3种实验体系中被Fe0还原降解的效果.结果表明:氧气对Fe0活性艳兰K-3R的还原降解有促进作用;Fe0与活性艳兰K-3R的最佳反应条件是在Fe0-H2O/O2体系中,体系的pH值为碱性,反应时间为10~15 min;增大转速、染料初始质量浓度以及Fe粉加入量,染料的还原降解效率提高,且Fe粉重复使用3~4次不影响处理效果.     

关于BR0-代数弱完备性的证明

目的对BR0-代数自身的完备性问题进行研究。方法研究了王国俊教授建立的模糊命题演算的形式演绎系统L*和与之在语义上相匹配的R0-代数,并结合吴洪博教授所定义的BR0-代数,从伴随的角度切入。结果在BR0-代数中定义了BR0-等式和BR0-方程,通过全序BR0-代数证明了BR0-代数自身的弱完备性。结论得到了BR0-代数的完备性定理,为相应形式逻辑系统与模糊推理的研究提供了理论框架。     

机械手模型的PLC控制系统设计

利用S7--300系列PLC对机械手进行控制，详细阐述了系统的主回路和控制回路工作原理以及接线图。
