Structural view of cadherin-mediated cell-cell adhesion

Following the multiplication of biochemical, biophysical and structural studies describing cadherin molecules and their interactions, several ideas have emerged to explain the mechanisms of cadherin-mediated cell adhesion. Although different models were proposed for cadherin interactions, a consensus has come forth considering lateral dimerization of cadherins as being a central component of the cell-cell adhesion process. 五六十年代的科研管理干部与科学家--罗登先生访谈录

通过对中国科学院细胞生物学研究所前所务秘书罗登先生的访谈，展示了五六十年代以王仲良、罗登为代表的少量当时被视为“右倾”的科研管理干部执行党的相关政策的情况。尽管作为党领导科学的具体体现者，他们在执行政策时有一定的自由度，但在当时的政治气氛下，他们“纠左”的能力是很有限的。     

MicroRNA-25对大鼠骨关节炎软骨细胞增殖和凋亡调节作用

探讨miR-25对白细胞介素1β(IL-1β)促进OA软骨细胞凋亡和增殖的影响。以木瓜蛋白酶膝关节腔注射诱导骨性关节炎动物模型和SD大鼠骨性关节炎软骨细胞原代培养。IL-1β体外刺激OA软骨细胞,模拟骨性关节炎体外环境。MiR-25mimic或inhibitor转染至OA软骨细胞。利用Annexin V-FITC/PI和CCK-8法检测软骨细胞细胞凋亡和细胞增殖。miR-25过表达明显抑制软骨细胞凋亡而miR-25低表达促进软骨细胞凋亡(P0.05);miR-25 mimic促进软骨细胞增殖,而miR-25 inhibitor抑制软骨细胞增殖(P0.05)。miR-25介导OA发病发生,进展中发挥着保护性调节作用,抑制OA软骨细胞凋亡和促进其增殖作用,可能对OA发病机制有新的认识和提示OA新治疗策略。     

不同还原性巯基氨基酸对人血管平滑肌细胞增殖的影响

目的:观察三种还原性巯基氨基酸对人血管平滑肌细胞增殖的影响.方法:采用组织贴块法体外培养人脐静脉血管平滑肌细胞(VSMCs),在培养液中分别加入三种不同浓度的还原性巯基氨基酸(Hcy,Cys,GSH)对VSMCs进行刺激,24h后同MTT比色法测定细胞的活力,<'3>H-TdR测定细胞的增殖率.结果:随浓度增加.Hcy和Cys明显促进VSMCs增殖,Hcy的作用略强于Cys;GSH只有在极高浓度时(1000umol/L)才显著促进VSMCs增殖.结论:极高浓度的Hcy、Cys、GSH均可促进VSMCs增殖.

Selective antiproliferative effects of thymidine

A substance with antiproliferative bioactivity in an aqueous extract ofCordyline terminalis was purified and identified by mass spectrometry to be the natural nucleoside, thymidine. 10^–5M Thymidine inhibited EL4 cell replication and decreased cell viability after 12–24 h. The effect was highly specific for this nucleoside. Treated cell cultures showed a significant increase in S phase cells and a corresponding decrease in G₁ phase cells. Nitrobenzylthioinosine (which prevented facilitated entry of thymidine) protected cells from the antiproliferative action of thymidine. A human breast cancer cell line (MCF7) was also growth-inhibited by 10^–5M thymidine but a murine lymphoma cell line (K36) was not. Thus, submillimolar thymidine has effects on cell proliferation which are selective both with respect to specificity for the compound and for different tumour cell lines.
