High-level expression and secondary structure analysis of the bovine mature prion protein

By using the recombinant DNA technology, the gene of the bovine mature prion protein (bPrPCL) has been cloned into pET30a and the resulting plasmid has been expressed in E.coli BL21(DE3). After solubilizing in 8 mol/L urea, the expression product was purified by cation ion exchange chromatography. The purified product was refolded by dilution and the recovery was about 15%. Analysis of mass spectrum, circular dichroism (CD) spectrum and Fourier transform infrared (FTIR) spectrum demonstrate that the molecular weight of the bPrPCL is 23 630 u, the bPrPCL has a high α-helix content (36.1%) and low β-sheet content (11.9%).     

Circular dichroism spectroscopic studies on structures formed by telomeric DNA sequencesin vitro

Telomere plays an important role in cellular processes, such as cell aging, death and carcinogenisis. Having special sequences, it can form quadruplex structurein vitro. Circular dichroism (CD) spectroscopic studies show that TTAGGG, (TTAGGG)₂ and (TTAGGG)₄ can all form quadruplexin vitro and exist mainly as parallel quadruplex without metal ions. Both K⁺ and Na⁺ can stabilize the tetrameric structure and facilitate the forming of anti-parallel conformation. Furthermore, the conformations of quadruplex can also be affected by sequence length, the nature and concentration of metal ions.     

Enhancement of T cell immune response to lymphoma cells by CD28/CD80 alternative pathway

The aim of this study is to determine whether myeloma and lymphoma tumor cells can function as efficient antigen presenting cells (APC) to enhance the co-stimulation of T cells. The expression and function of T cell activation-related molecules, especially CD80, CD28, CD40 and CD40 ligand (CD40L), were studied on nine human myeloma cell lines (HMCL) and two B lymphoma cell lines. In the case of myeloma cell lines, the cells generally lacked CD80 antigen and expressed a heterogeneous CD40, and the expressions of CD40 and CD80 molecules could not be induced by either CD28 stimulation or CD40 ligation. Conversely, in the two B lymphoma cell lines, tumor cells expressed both CD80 and CD40 to some extent. CD28 stimulation could obviously increase the expression of CD80, CD40 and some adhesion molecules, and therefore generate a more efficient anti- tumor cell immunity. In conclusion, CD28 stimulation combined with CD40 antibody or soluble CD40 ligand may be a promising immunotherapeutic approach to B lymphoma.     

非小细胞肺癌CD44v6、VEGF与C-erbB-2和nm23基因蛋白的表达及其临床意义

应用链霉菌抗生物素蛋白 过氧化物酶 (SP)法染色技术测定 65例非小细胞肺癌(NSCLC)组织标本的CD4 4v6、VEGF、C erbB 2和nm2 3基因蛋白的表达水平 .显示CD4 4v6、VEGF、C erbB 2及nm2 3在NSCLC组织中表达的阳性率分别为 66.2 %、78.5%、63.1%及73.8% ,均显著高于癌旁组织 ;上述四项指标阳性率与NSCLC的病理类型无明显关系 ;区域性淋巴结转移组CD4 4V6及C erbB 2的阳性表达率均显著高于无淋巴结转移组 ;VEGF在肿瘤3.0≤Φ <5.0cm及Φ≥ 5.0cm组的阳性率均显著高于Φ <3.0cm组 (P <0 .0 5) ;提示检测CD4 4v6、VEGF和C erbB 2基因蛋白的表达 ,有可能为判断NSCLC淋巴结转移、病变的发展以及评估预后提供依据     

硬盘镜像技术在光盘网络中的应用

硬盘镜像是将CD-ROM光盘按ISO 9660的光盘格式压缩或不压缩地“镜像”到硬盘中的一种新技术,硬盘镜像完整地保留了CD光盘的属性,并且具有防病毒侵袭和数据被改写的功能。目前硬盘镜像系统正以存储容量大,支持大规模光盘在线共享、读取速度快、性价比高的特点逐步取代传统的光盘塔和光盘库,成为光盘网络应用中的新方向。     

RNA干扰CD133基因对结肠癌干细胞生物学行为的影响

目的探讨CD133基因表达、活化被阻断后对结肠癌干细胞生物学行为的影响。方法从EpcAMhighCD44＋结肠癌干细胞中流式分选获得CD133＋细胞,感染LV-CD133shRNA载体慢病毒后观察CD133＋结肠癌干细胞在生长方式、成球能力、克隆形成率、成瘤能力以及ABCC2mRNA的变化;Westernblot分析CD133-细胞中CD133蛋白表达情况。结果EpcAMhighCD44＋结肠癌干细胞中CD133＋细胞比例为89．2％。实验组经过LV-CD133shRNA载体病毒感染后,在干细胞养液中细胞改悬浮生长的方式为贴壁生长,不能形成细胞球。MTT法测定发现细胞增殖减慢,克隆形成率明显下降。将感染细胞移植在Balb／C裸鼠体内,在观察期间,感染LV—CD133shRNA载体病毒的CD133＋细胞无肿瘤形成。ABCG2mRNA表达水平明显降低（P〈0．01）。从EpcAMhighCD44＋结肠癌干细胞中流式分选获得CD133-细胞,其中也有CD133蛋白的表达。结论CD133维持结肠癌干细胞生物学特性。     

Cellular uptake of amelogenin,and its localization to CD63, and Lamp1-positive vesicles

Proteins of the developing enamel matrix include amelogenin, ameloblastin and enamelin. Of these three proteins amelogenin predominates. Protein-protein interactions are likely to occur at the ameloblast Tomes’ processes between membrane-bound proteins and secreted enamel matrix proteins. Such protein-protein interactions could be associated with cell signaling or endocytosis. CD63 and Lamp1 are ubiquitously expressed, are lysosomal integral membrane proteins, and localize to the plasma membrane. CD63 and Lamp1 interact with amelogenin in vitro. In this study our objective was to study the molecular events of intercellular trafficking of an exogenous source of amelogenin, and related this movement to the spatiotemporal expression of CD63 and Lamp1 using various cell lineages. Exogenously added amelogenin moves rapidly into the cell into established Lamp1-positive vesicles that subsequently localize to the perinuclear region. These data indicate a possible mechanism by which amelogenin, or degraded amelogenin peptides, are removed from the extracellular matrix during enamel formation and maturation. 牛朊病毒正常成熟蛋白的高效表达和二级结构分析

利用ＤＮＡ重组技术,将牛朊病毒正常成熟蛋白基因插入表达载体ｐＥＴ３０ａ,在大肠杆菌ＢＬ２１（ＥＤ３）中高效表达。将表达产物（包涵体）用８ｍｏｌ／Ｌ尿素溶解,用阳离子交换柱纯化,纯化产物复性后,进行质谱、圆二色谱（ＣＤ）以及Ｆｏｕｒｉｅｒ变换红外光谱（ＦＴＩＲ）分析。结果表明,所得牛肼病毒正常成熟蛋白的分子量为２３６３０ｕ,其二级结构主要由α螺旋构成,含少量β折叠。     

荞麦花粉肽及其类似物的合成、结构和免疫活性研究

用Merrifield固相法合成了荞麦花粉肽BPP-1及其类似物BPP-1-3,BPP-1-4和BPP-2。通过FT-IR和CD谱初步讨论了BPP-1的二级结构,结果表明其水溶液构象属于无规卷曲和 β-折叠,但在三氟乙醇中,CD谱在206nm和222nm附近呈双负峰形,分子形成了α-螺旋构象。研究了BPP-1,BPP-1-3和BPP-2对动物和人淋巴细胞的增殖应答功能和抑制释放sIL-2R的作用,结果显示它们具有较强的促免疫活性。     

ROCK1 与CD40相互作用的质谱分析及在CD40信号通路中功能的初探

为鉴定和验证ROCK1和CD40的相互作用,探索ROCK1在CD40通路中可能发挥的作用,应用HD质谱分析ROCK1和CD40的相互作用,用免疫共沉淀实验验证其相互作用, 用ROCK1抑制剂研究ROCK1对CD40通路的影响. 结果ROCK1与CD40存在相互作用;并参与CD40介导的信号通路.说明ROCK1能与CD40发生相互作用,为CD40信号通路的研究提供了新的线索.
