PX domain and CD domain play different roles in localization and vacuolation of Sorting Nexin 10

Sorting nexins （SNXs） are PX domain containing proteins and essential for intracellular protein sorting, trafficking and signal transduction. The PX domains of SNXs can bind to various phosphorelated phosphoinositides （PIs） and target the host proteins to endosomes. Recently, we have reported that overexpression of SNX10 in mammalian cells could induce giant vacuoles. In this study, we aimed to identify regions in SNX10 critical for the vacuolation activity. We found that both the PX domain and the CD1 region were essential for vacuolation. We provided evidence that the PX domain was able to specifically bind to Ptdlns（3）P and target SNX10 to endosomes. A mutation in the 131 region of the PX domain （V15A） disrupted the Ptdlns（3）P binding ability and the endosomal localization of SNX10. However, correct subcellular localization alone was not sufficient for SNX10 to induce vacuoles. We found that the CD1 region, which was not required for the localization, was indispensable for the vacuolation activity of SNX10. In summary, both the PX domain and the CD1 region are necessary for SNX10 to induce vacuoles but they play different roles in this process.

手性Schiff碱配合物在催化、生物活性、药物合成和功能材料等方面具有重要的应用。本文合成了两对新的手性Schiff碱镍（Ⅱ）配合物,并通过X-单晶衍射、红外光谱对其结构进行了测定,X-单晶衍射分析表明它们都属于P212121手性空间群,并通过圆二色谱对合成化合物的光学对映体性质进行了一步表征。     

浅谈随书光盘的管理与利用——以太原市图书馆为例

介绍了随书光盘的分类以及太原市图书馆对随书光盘的加工方式,探讨了光盘在使用过程中所遇到的问题,提出了随书光盘利用和保护措施。     

稀释水应用于匀浆辊流浆箱的数值模拟

为了确定匀浆辊流浆箱能否采用稀释水来调节纸页横幅定量偏差,文中采用计算流体动力学软件Fluent对匀浆辊流浆箱内的流场进行数值模拟,利用Multiphase-Mixture模型分析了两种流体在匀浆辊流浆箱中的混合情况,利用旋转坐标系统(RRF)模型考察流场内旋转域对流场的影响,计算在不同进浆流速和匀浆辊转速的情况下,稀释水的调节效果.模拟结果表明:稀释水的调节效果与匀浆辊转速符合一次函数关系.实验证明,采用稀释水调节后,横幅定量偏差降低了3个百分点以上,该结果与数值模拟结果相符,说明稀释水调节单元可用于匀浆辊式流浆箱.     

HBME-1、Galectin-3和CD44v6在甲状腺滤泡癌与不典型腺瘤中的表达

 探讨间皮瘤相关抗体-1（HBME-1）、半乳糖凝集素-3（Galectin-3）及粘附分子CD44v6在甲状腺滤泡癌（Follicular Thyroid Carcinoma,FTC）与不典型腺瘤（Atypical Thyroid Adenoma,ATA）中的表达差异,寻找有助于二者临床病理诊断的肿瘤标记物及探索其部分发病机制,拟解决在甲状腺临床病理中对良、恶性难以确定的滤泡性肿瘤的准确判定及对其预后的预示作用。采用免疫组化SP三步法检测37例FTC及18例ATA中HBME-1、Galectin-3和CD44v6的表达,以10例嗜酸性腺瘤（OncocyticThyroid Adenoma,OTA）、8例普通滤泡性腺瘤（Follicular Thyroid Adenoma,FTA）、7例甲状腺乳头状癌（Papillary ThyroidCarcinoma,PTC）及5例尸检正常甲状腺组织作为对照。结果显示,HBME-1、Galectin-3及CD44v6在FTC中的阳性表达率分别为70.3%、64.9%和62.2%,在ATA中阳性表达率分别为66.7%、61.1%和61.1%,三者在FTC与ATA中的表达差异均无统计学意义（χ2分别为0.074、0.094、0.245,P值分别为0.786、0.759、0.620,P值均＞0.05）;在10例OTA中的表达均为100%;8例FTA中的表达分别为3/8例、6/8例及4/8例;7例PTC的表达分别为7/7例、6/7例及7/7例;5例正常尸检甲状腺组织均无表达。由此得出结论： HBME-1、Galectin-3和CD44v6对FTC与ATA的临床病理鉴别诊断无统计学意义,但提示三者可能在甲状腺滤泡癌与不典型腺瘤的发病机制及进展过程中起着某种共同作用。由于此3种标记物已被证实在诸多肿瘤的发生、发展过程中起着重要作用,故在实际甲状腺临床病理诊断中对表达此3种标记物的良、恶性难以确定的病例可能提示其恶性程度增高,尽管不足以诊断为癌,但仍应加强随访。有关FTC与ATA的临床病理诊断与鉴别诊断及二者的发病机制尚有待于进一步的探索和研究.     

Effect of MTECl on the functional expression of TCRαβ~ + 3G11~- 6C10~-CD4~+CD8~- thymocyte subset

A murine CD4+ thymocyte subset with phenotype of TCRαβ + 3G11- 6C10- CD4 + CD8- CD69 +/- HSAmed/locontains the cells in relatively functional matured status. The functional property of the cells in this subset is characterized by the unique pattern of cytokine production at transitional stage from Th0 to Th2 type with the latter being the dominant type. After being co-cultured with murine thymic medullary epithelial cell line (MTEC1) cells, a murine thymic medullary type epithelial cell line, the TCRαβ(T 3G11 6C10-CD4 + CD8- CD69+/- HSAmed/l? thy-mocytes, has exhibited significantly higher levels of proliferation capability and IL-6 production, whereas the production of IL-4 and IL-10 is suppressed after co-culturing with MTECl. By contrast, MTECl could not induce thymocytes to secrete Th1 type of cytokines. The results suggest that MTECl can regulate functional status of this thymocyte subset and induce them to develop into a specialized Th2 subset.     

Important amino acid in the hemagglutinin glycoprotein of measles virus (MV) that governs hemadsorption

Two strains of measles virus IMA and SMD that were isolated using B95a cell line show hemadsorption negative. After adaptation to Vero cells, these strains gain the ability to agglutinate AGM-RBC and show hemadsorption positive. Sequence analysis of these two kinds of virus reveals that the two strains all have a Ser (HAD negative) to Gly (HAD positive) mutation at position 546 in the H protein. Site-directed mutagenesis and expression in COS cells were used to confirm that the mutation Ser→Gly is responsible for hemadsorption alteration. Then the monoclonal antibody against CD46 was used to identify that this mutation also governs the binding function of MV H protein to CD46 receptor. The data provide a new important amino acid of MV H protein that governs the hemadsorption and binding to CD46 receptor.
