构建肝癌相关抗原HAb18G/CD147全长及缺失片段E51(第22—50位氨基酸缺失)的真核荧光蛋白表达载体,进一步研究该基因在肝细胞肝癌发生中的作用。通过PCR及Overlapping PCR的方法获得目的基因.与荧光载体pEGFP—N1双酶切、连接、转化E.coli JM109.构建含有肝癌相关抗原HAb18G/CD147全长及E51的真核荧光蛋白表达载体.并经限制性酶切及序列分析证明插入是否正确。用阳离子脂质体介导转染COS-7细胞,进行瞬时表达;荧光显微镜观察EGFP表达,通过流式细胞术检测蛋白的表达情况,明胶酶谱法鉴定其功能。成功地构建了真核荧光蛋白表达载体pEGFP—N1/HAb18G、pEGFP—N1/E51,并经限制性酶切及序列分析证明外源基因插入正确;流式细胞术鉴定该蛋白表达正确;功能实验证实缺失片段E51不具有诱导成纤维细胞分泌MMPs的功能.结果显示HAb18G/CD147基因第22—50位氨基酸与其刺激成纤细胞分泌MMP和功能有密切关系。实验结果为HAb18G蛋白分子的生物学功能研究奠定了基础。 相似文献

Among the heterogeneous population of circulating hematopoietic and endothelial progenitors, we identified a subpopulation
of CD133+ cells displaying myogenic properties. Unexpectedly, we observed the expression of the B-cell marker CD20 in blood-derived
CD133+ stem cells. The CD20 antigen plays a role in the modulation of intracellular calcium homeostasis through signaling pathways
activation. Several observations suggest that an increase in intracellular calcium concentration ([Ca2+]i) could be involved in the etiology of the Duchenne muscular dystrophy (DMD). Here, we show that a CD20-related signaling
pathway able to induce an increase in [Ca2+]i is differently activated after brain derived neurotrophic factor (BDNF) stimulation of normal and dystrophic blood-derived
CD133+ stem cells, supporting the assumption of a “CD20-related calcium impairment-affecting dystrophic cells. Presented findings
represent the starting point toward the expansion of knowledge on pathways involved in the pathology of DMD and in the behavior
of dystrophic blood-derived CD133+ stem cells.

Mast cells play pivotal roles in allergic and inflammatory processes via distinct activation pathways. Mucosal and serosal mast cells are activated by the IgE/FcɛRI pathway, while only serosal mast
cells are activated by basic secretagogues. We show that CD47 receptors are expressed on rat peritoneal mast cells. 4N1K,
a peptide agonist of CD47, rapidly caused exocytosis. Such exocytosis required increased intracellular calcium and was inhibited
by pertussis toxin and an antibody against the βγ dimer of a Gi protein. Cooperation with integrins and glycosylphosphatidylinositol-anchored proteins was necessary, since anti-integrin
antibodies and pretreatment with phosphatidylinositol-phospholipase C reduced exocytosis. Depletion of membrane cholesterol
inhibited exocytosis and decreased CD47 in lipid rafts, consistent with a CD47/integrin/Gi protein complex being located in rafts. An anti-CD47 antibody inhibited exocytosis induced by 4N1K and by mastoparan and
spermine, suggesting that basic secretagogues might target CD47. We propose that 4N1K-stimulated mast cell exocytosis involves
a CD47/integrin/Gi protein complex.

目的研究胃癌侧群(Side Population,sP)细胞对化疗药物5-Fu(氟尿嘧啶)的耐药性及可能机制,并检测干细胞相关基因Nanog、Musashi-1及cD44的表达情况。方法选择人胃癌细胞株sGc-7901,以荧光染料H0echst 33342染色,维拉帕米桔抗对照,应用流式细胞仪分选sP细胞和nonsP细胞。细胞耐药实验比较sP细胞与nonsP细胞对化疗药物5.Fu的耐药性差异;westem_h10t检测ABcG2和bcl-2蛋白表达情况;流式细胞仪分析细胞周期;荧光定量PcR检测两组细胞中干细胞相关基因Nanog、Musashi-1及cD44mRNA的表达差异。结果胃癌细胞株sGc.790l中sP细胞的比例为2.8%,sP细胞对5-Fu的耐药存活率明显高于non-sP细胞(P〈0.05),与nonsP细胞相比,sP细胞高表达耐药蛋白ABcG2和抗凋亡蛋白bcl-2,有更多的细胞处于c0/G1期(P〈0.05),并高表达干细胞相关基因Musashi-1和cD44。结论胃癌sGC_7901细胞株中sP细胞对化疗药物5.Fu的耐药性明显高于nonsP细胞,其耐药机制可能与sP细胞高表达耐药蛋白ABcG2和抗凋亡蛋白bcl-2,有更多细胞处于G0/Gl期有关;Musashi-1和cD44可能是相对特异性的胃癌干细胞标志物。 相似文献

Dendritic cells (DCs) play a critical role in orchestrating the innate and adaptive components of the immune system so that appropriate, coordinated responses are mounted against infectious agents. Tissue-resident DCs interact with microbes through germline-encoded pattern-recognition receptors (PRRs), which recognize molecular patterns expressed by various microorganisms. Antigens use PRR activation to instruct DCs for the appropriate priming of natural killer (NK) cells, followed by specific T-cell responses. Due to the central role of DCs in regulating the activation and progression of immune responses, minor imbalances in the feedback control of Toll-like receptor (TLR)-activated cells have been associated with autoimmunity in genetically prone individuals. We review here recent findings on the role of DCs in the priming of innate and adaptive immune responses and the possible involvement of DCs in inducing and maintaining autoimmune reactions. 相似文献

Mechanisms of HIV-mediated CD4+ T cell loss leading to immunodeficiency are amongst the most extensively studied yet unanswered
questions in HIV biology. The level of CD4+ T cell depletion in HIV infected patients far exceeds the number of infected T
cells, suggesting an indirect mechanism of HIV pathogenesis termed bystander cell death. Evidence is accumulating that the
HIV envelope glycoprotein (Env) is a major determinant of HIV pathogenesis and plays a critical role in bystander cell death.
The complex structure and function of HIV Env makes the determination of the mechanism of Envmediated apoptosis more complex
than previously thought. This review will examine the complex relationship between HIV Env phenotype, coreceptor expression
and immune activation in determining HIV pathogenesis. We review data here corresponding to the role of HIV Env hemifusion
activity in HIV pathogenesis and how it interplays with other AIDS associated factors such as chemokine receptor expression
and immune activation.

Mechanism of HAb18G/CD147 underlying the metastasis process of human hepatoma cells has not been determined. In the present
study, we found that integrin α3β1 colocalizes with HAb18G/CD147 in human 7721 hepatoma cells. The enhancing effect of HAb18G/CD147
on adhesion, invasion capacities and matrix metalloproteinases (MMPs) secretion was decreased by integrin α3β1 antibodies
(p<0.01). The expressions of integrin downstream molecules including focal adhesion kinase (FAK), phospho-FAK (p-FAK), paxillin,
and phospho-paxillin (p-paxillin) were increased in human hepatoma cells overexpressing HAb18G/CD147. Deletion of HAb18G/CD147
reduces the quantity of focal adhesions and rearranges cytoskeleton. Wortmannin and LY294002, specific phosphatidylinositol
kinase (PI3K) inhibitors, reversed the effect of HAb18G/CD147 on the regulation of intracellular Ca2+ mobilization, significantly reducing cell adhesion, invasion and MMPs secretion potential (p<0.01). Together, these results suggest that HAb18G/CD147 enhances the invasion and metastatic potentials of human hepatoma
cells via integrin α3β1-mediated FAK-paxillin and FAKPI3K-Ca2+ signal pathways.

Natural killer T (NKT) cells have been shown by a number of studies to play a protective role against cancers, autoimmune
diseases and infectious diseases. Several glycolipids and phospholipids derived from mammalian, bacterial, protozoan and plant
species have recently been identified as natural ligands (antigens) for NKT cells. Some of these glycolipid/phospholipid ligands
have now been crystallized in forms bound to CD1d molecules, and the tertiary structure of these complexes has finally been
revealed. This review is intended to list natural NKT cell ligands identified to date, and discuss how their structures relate
to their propensity to bind CD1d molecules and, as a consequence, stimulate NKT cells.
