兔IL-15蛋白在毕赤酵母中的表达及其免疫学活性

用特异性引物扩增出兔IL-15基因片段,用KpnI和NotI双酶切后将其定向克隆到pPICZαA中,构建出重组质粒pPICZαAIL-15。将pPICZαAIL-15用SacI内切酶线性化后,电转化感受态GS115酵母细胞,用PCR法筛选阳性重组子。用1.5%的甲醇诱导重组酵母菌,取培养物上清进行重组蛋白的检测。结果SDS-PAGE电泳可在重组酵母菌培养物上清中检测到分子量为18.6kDa大小的重组蛋白。凝胶薄层扫描分析表明,3株重组酵母菌在1.5%甲醇诱导144h后,重组IL-15表达量约占培养物上清总蛋白量的6.1%~8.6%。将重组IL-15用Ni离子亲和层析纯化后,在MTT试验中表现出明显的促进淋巴细胞的增殖反应,证实表达的重组IL-15具有天然蛋白的免疫学活性。     

TA15合金热压缩流变应力修正及神经网络预测

文章采用Gleeble-1500热模拟试验机对TA15合金进行了等温热压缩试验;对试验数据进行了摩擦修正,以修正后的流变应力为基础,研究了在不同的热变形条件下,TA15合金流变应力的变化规律;采用人工神经网络的方法建立了该合金在不同温度范围内流变应力曲线的预测模型.预测值与实测值吻合较好,表明该方法能够很好地预测TA15合金在热变形过程中流变应力的变化.     

萘并冠醚配合物[K（N15C5）2]2[Cd（SCN）4]CH2CL2的合成与晶体结构表征

研究了萘并15冠5与CdCl2·2．5H2O,KSCN的反应,得到标题配合物[K（N15C5）2]2[Cd（SCN）4]CH2Cl2,并通过元素分析、红外光谱、X-射线单晶衍射对配合物进行了表征．配合物为单斜晶系,空间群P^-1,晶体学数据：a=15．521（2）A、b=15．617（3）A、c=20．582（3）A,α=73．528（3）°、β=80．355（3）°、γ-61．295（2）°,V=4193．6（12）A3,Z=4,Dcaled=1．411Mg／m3,F（000）=1848,R1=0．0579,wR2=0．1227,R1=0．1757,wR2=0．1708．标题配合物由两个[K（N15C5）2]＋配阳离子和一个[Cd（SCN）4]2-配阴离子组成．两个N15C5分子和钾离子通过10个K—O键形成夹心式配位结构,[Cd（sCN）4]2-中的Cd2＋与四个NCS基团中的四个N配位,形成变形四面体结构．配阳离子子[K（N15C5）2]＋与配阴离子[Cd（SCN）4]2-通过静电作用形成中性配合物．

Control of the cardiovascular system ofAplysia by identified neurons

The neural network that controls the cardiovascular system ofAplysia adapts cardiovascular function to a variety of different physiological and behavioral situations. It (1) coordinates the cardiovascular system with the renal and respiratory systems; (2) modifies both systemic and regional blood flow during food-elicited arousal and feeding; and (3) changes the tension of longitudinal vascular muscle to adapt the arterial tree to changes in body shape. Indirect evidence suggests that the cardiovascular control circuit may also play a role in maintaining homeostasis during egg laying. Several putative neurotransmitters, including acetylcholine, serotonin, R151 and R152 peptides, have been localized to identified neurons in this circuit.     

(+)-4-(2-甲基丁基)-4′-氰基联苯的合成

以(+)-对甲苯磺酸-2-甲基丁酯与联苯基溴化镁反应,在催化剂作用下实现手性戊基与联苯偶联,再经碘化和氰化,合成了(+)-4-(2-甲基丁基)-4′-氰基联苯(CB-15)。总收率为15%(以左旋戊醇计算)。同时,就手性戊基与联笨偶联的机理等问题作了讨论。

Effect of P15INK4b/MTS2 on the proliferation of human hepatoma cells SMMC-7721

The full length cDNA coding for P15 INK4b, which is a cyclin-dependent kinase inhibitor, was cloned to plasmid PXJ41-neo (Eco RⅠ/XhoⅠ site) and the new constructed plasmid pXJp15 was obtained. pXJp15 was transferred into the human hepatoma SMMC-7721 cells by lipofectine reagent. After G418 selection, a series of cell lines stably expressing high levels of P15 (named SHT) and the clone containing vector PXJ41-neo only (named SVXJ) were obtained by Northern and Western analysis. The results showed that the proliferation of SHT cells is inhibited compared with that of SVXJ cells. Cell cycle analysis indicated that overexpressing of P15 inhibited the growth of SHT cells by decreasing progrssion of cells from G1 to S and G2 to M phases. The levels of c-Myc and c-Fos were obviously decreased in SHT cells compared with control cells by Western blotting. The decreased expression of oncogene may be one of the molecular mechanisms of the effect of P15 on the proliferation of in SHT cells.
