Descartes’s changing mind

Descartes is always concerned about knowledge. However, the Galileo affair in 1633, the reactions to his Discourse on method, and later his need to reply to objections to his Meditations provoked crises in Descartes’s intellectual development the import of which has not been sufficiently recognized. These events are the major reasons why Descartes’s philosophical position concerning how we know and what we may know is radically different at the end of his life from what it was when he began. We call this later position Descartes’s epistemic stance and contrast it with his earlier methodological, metaphysical realism. Yet Descartes’s epistemic views cannot be separated from other aspects of his work, for example, his views concerning God, causality, metaphysics, and the nature of science. A further meta-implication is that serious errors await any scholar who cites early Cartesian texts in support of late Cartesian positions, or who uses later texts in conjunction with early ones to support a reading of Descartes’s philosophy.     

Flow-induced release of adenosine 5′-triphosphate from endothelial cells of the rat mesenteric arterial bed

Adenosine 5-triphosphate (ATP) was released into the perfusate of rat isolated mesenteric arterial beds during each of two consecutive increases in flow. There was no significant difference between the amounts of ATP released on each occasion. Substance P was also released into the perfusate by increased flow, although its release was more variable. Removal of the endothelium of the mesenteric vessels with sodium deoxycholate led to a significant reduction (74%) in the amount of ATP released compared with the release before the endothelium had been removed. This suggests that the ATP released into the mesenteric arterial perfusate during increased flow arises from endothelial cells.     

Gigantecin: A novel antimitotic and cytotoxic acetogenin,with nonadjacent tetrahydrofuran rings,fromGoniothalamus giganteus (Annonaceae)

Summary Gigantecin (I), a novel tetrahydroxy-di-tetrahydrofuran fatty acid -lactone (acetogenin), was isolated from an ethanolic extract of the stem bark ofGoniothalamus giganteus Hook. f., Thomas (Annonaceae), by means of activity-directed fractionation (brine shrimp lethality test). This new compound is extremely cytotoxic to human tumor cells, inhibits crown gall tumors on potato discs, and is active in an assay designed to detect antimitotic agents (9 ASK).     

2,9-二甲基-1,10-菲罗啉络合推动Ni~(2+)离子在水/硝基苯界面传递的研究

用半微积分伏安法研究了2,9—二甲基—1,10—菲罗啉(L)络合推动Ni(Ⅱ)离子在水/硝基苯界面的传递行为。在0.1mol/L NiCl_2(或NiSO_4)(W)—0.05mol/L TBATPB,L(NB)体系中,Ni(Ⅱ)与L生成NiClL~+(或NiL_2~(2+))络合离子,从而在W/NB界面发生离子传递。当C_(Ni~(2+))>>C_L,e_p与C_L成正比,线性范围为1×10~(-6)mol/L～5×10~(-4)mol/L,可作为L的定量分析。文中进行了理论探讨,测得了络合物的形成常数等。     

V2O5和Li2CO3共掺杂对Mg4Nb2O9陶瓷烧结行为及微波介电性能的影响

采用固相法，制备了Mg4Nb2O9（MN）微波介质陶瓷，研究了V2O5和Li2CO3共掺杂对MgaNb2O9陶瓷烧结行为、相结构、显微结构和微波介电性能的影响．结果表明，采用1．5％V2O5和1．5％Li2CO3共掺杂，能够将Mg4Nb2O9（135O℃）陶瓷的烧结温度降至925℃，且有助于Mg4Nb2O9单相的形成．3．0％（V2O5，Li2CO3）共掺杂样品在925℃空气中烧结5h可获得良好的微波介电性能（介电常数为13．7，品质因数为77975GHz）．     

Cpf1 和 Cas9核酸酶靶向EGFR-L858R突变的研究

表皮生长因子受体(EGFR)单核苷酸突变(2573TG,L858R)占所有EGFR突变的90%.使突变的EGFR失活对有此突变的病人非常有利.这里,应用双荧光报告分析的方法分析规律成簇间隔短回文重复(CRISPR)系统中Cpf1和Cas9在靶向EGFR-L858R突变的编辑效率.在EGFR-L858R突变位点的附近,有两个Cpf1前间区序列邻近基序(PAMs)——TTTN.并且,2573TG突变形成了一个Cas9的PAM——NGG.因此本文通过构建两条AsCpf1的gRNAs(gRNA1和gRNA2)和一条SpCas9的gRNA(gRNA3)在体外通过双荧光蛋白分析系统去评估SpCas9和AsCpf1特异性靶向等位基因的能力.结果证实了AsCpf1和SpCas9都能够特异性的编辑突变的EGFR(2573TG).     

重组枯草芽孢杆菌谷氨酰胺合成酶蛋白在改良M9培养基中的诱导表达

以重组枯草芽孢杆菌谷氨酰胺合成酶(L-谷氨酸:氨连接酶,Glutamine Synthetase, GS EC 6.3.1.2)蛋白表达宿主菌株BL21(DE3) (pET3C/谷氨酰胺合成酶)作为研究对象,借助SDS PAGE分析方法,对于用乳糖诱导由T7lac启动子控制的重组目的产物蛋白的各种基本参数进行了初步的研究.研究了最佳本底培养基、最佳碳源及其最佳添加比例、碳源与诱导物的最适组合,并探索了适用于改良型培养基的诱导参数.实验结果表明,对于重组目的产物蛋白,改良型M9培养基完全可以替代LB培养基;以甘油为碳源,可以避免葡萄糖对T7lac启动子的屏蔽作用.以乳糖为诱导物,目的蛋白的表达量、可溶性及酶活与以IPTG为诱导物基本接近.研究结果为酶法合成谷氨酰胺的工业化生产提供了一定的参考.     

DCPD/C9热聚型石油树脂的聚合工艺研究

提取乙烯裂解副产物C5馏分中的双环戊二烯馏分，与裂解C9、C5馏分按不同配比混合，通过自由基聚合反应得到热聚型石油树脂，并确定了热聚型石油树脂的工艺参数。
