蛋白质网络的特征研究

基于复杂网络理论研究蛋白质网络的特征.构建了蛋白质网络,当截断距离R为0.7 nm时,单体蛋白网络表现出明显的小世界性质.分析了蛋白质网络的分层结构,当截断距离增大时,分层指数b值减小,截断距离R为2.4 nm时,分层指数b与网络规模正相关.研究结果表明,单体蛋白质网络及其对应复合物中肽链网络中接近中心度和介数中心度较高的残基均是非极性、疏水性氨基酸.     

光谱法研究EDTA-Fe(Ⅲ)与牛血清白蛋白的相互作用

应用紫外光谱法和荧光光谱法研究了乙二胺四乙酸铁(Ⅲ)(EDTA-Fe(Ⅲ))与牛血清白蛋白的相互作用.结果表明:EDTA-Fe(Ⅲ)可以进入牛血清白蛋白的疏水腔,使蛋白质非极性区的生色基暴露于极性溶剂;EDTA-Fe(Ⅲ)通过静态猝灭的方式使牛血清白蛋白的荧光强度显著降低;EDTA-Fe(Ⅲ)与牛血清白蛋白主要以氢键和范德华力相结合.测定了EDTA-Fe(Ⅲ)与牛血清白蛋白的结合常数KA和结合位点数n,利用Forster非辐射能量转移机制确定了牛血清白蛋白与EDTA-Fe(Ⅲ)的结合距离和能量转移效率,并使用同步荧光技术探讨了EDTA-Fe(Ⅲ)对牛血清白蛋白构象的影响.     

Identification of proteinaceous binding media for the polychrome terracotta army of Emperor Qin Shihuang by MALDI-TOF-MS

A proteinaceous binding media for the polychrome terracotta army of Emperor Qin Shihuang has been identified using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS).Methods for the preparation of the model samples and the elimination of interferences have been evaluated,as well as ageing processes.A method involving the complexation of EDTA in combination with dialysis was used to eliminate any interference in the polychrome layers taken from the historical samples.The model samples were aged by being buried in loess to replicate the ageing process of the historical samples as closely as possible.The optimum conditions for the extraction and subsequent enzymatic hydrolysis of the extracted protein were investigated.Under the optimized conditions,the‘‘peptide mass fingerprints’’of the proteinaceous binding media from the historical and model samples were determined by MALDITOF-MS.The results revealed that animal glue was used as the binding media for the polychrome layers of Emperor Qin Shihuang’s terracotta army.It also shows that animal glue used as binding media has a long history in China.

An Integrated Workflow for Proteome-Wide Off-Target Identification and Polypharmacology Drug Design

Polypharmacology,which focuses on designing drugs to target multiple receptors,has emerged as a new paradigm in drug discovery.To rationally design multi-target drugs,it is fundamental to understand protein-ligand interactions on a proteome scale.We have developed a Proteome-wide Off-target Pipeline （POP） that integrates ligand binding site analysis,protein-ligand docking,the statistical analysis of docking scores,and electrostatic potential calculations.The utility of POP is demonstrated by a case study,in which the molecular mechanism of anti-cancer effect of Nelfinavir is hypothesized.By combining structural proteome-wide off-target identification and systems biology,it is possible for us to correlate drug perturbations with clinical outcomes.

光谱检测新技术应用研究－Ⅱ.吸光染料标记测定蛋白质

将所提出的光谱检测新技术应用于常用标记染料的分析测定, 所得光谱图形状与吸收光谱相似, 检测限比相应的分光光度法测定降低１ 个数量级左右．     

螺旋藻酶促水解提取营养成分的研究

研究了在４种ｐＨ值环境中对螺旋藻细胞酶促水解的结果．以水解液中的水溶性蛋白质含量和全氮量为指标,初步确定了酶促水解提取螺旋藻细胞内营养物质的适宜的工艺条件,包括使用的酶制剂、加酶量、ｐＨ值、加热温度和作用时间等     

曲利本蓝—蛋白质体系的共振瑞利散射及其分析应用

研究了曲利本蓝蛋白质体系的共振瑞利散射光谱,考察了体系的光谱特征、影响因素和适宜的反应条件.确立了共振瑞利散射强度与蛋白质浓度的关系,提出了用共振瑞利散射光谱测定痕量蛋白质浓度的高灵敏的分析方法.
