为了解重金属Hg2+对青海湖裸鲤肾脏组织的细胞毒性;以含有Hg2+的浓度为0.0025、0.005、0.01、0.02mg/L的培养液培养裸鲤肾脏组织细胞;结果表明,0.005mg/L的Hg2+浓度为裸鲤肾脏细胞毒性死亡的临界浓度,且裸鲤肾脏细胞的死亡率随Hg2+的浓度的增加而增大,当培养基的Hg2+浓度达到0.02mg/L时细胞全部死亡. 相似文献

Role of dystrophin and utrophin for assembly and function of the dystrophin glycoprotein complex in non-muscle tissue 总被引:5,自引:0,他引:5
The dystrophin glycoprotein complex (DGC) is a multimeric protein assembly associated with either the X-linked cytoskeletal
protein dystrophin or its autosomal homologue utrophin. In striated muscle cells, the DGC links the extracellular matrix to
the actin cytoskeleton and mediates three major functions: structural stability of the plasma membrane, ion homeostasis, and
transmembrane signaling. Mutations affecting the DGC underlie major forms of congenital muscle dystrophies. The DGC is prominent
also in the central and peripheral nervous system and in tissues with a secretory function or which form barriers between
functional compartments, such as the blood-brain barrier, choroid plexus, or kidney. A considerable molecular heterogeneity
arises from cell-specific expression of its constituent proteins, notably short C-terminal isoforms of dystrophin. Experimentally,
the generation of mice carrying targeted gene deletions affecting the DGC has clarified the interdependence of DGC proteins
for assembly of the complex and revealed its importance for brain development and regulation of the ’milieu intérieur. Here,
we focus on recent studies of the DGC in brain, blood-brain barrier and choroid plexus, retina, and kidney and discuss the
role of dystrophin isoforms and utrophin for assembly of the complex in these tissues.

The present work is concerned with the relationship of the different colors of NSE (non-specific esterase) reaction product,
in which alpha-naphthyl acetate and diazotate of o-dianisidine (Fast Blue B salt) were used as a substrate and a coupler respectively
to describe the histochemical characteristics of NSE in chick embryonic mesonephros and metanephros. Our preliminary results
throw light on the nature of NSE reaction produced colors, under light microscope, which were different in various sections
for different airdrying time-length. The reaction product on the slices of the convoluted tubules of chick embryonic kidney
was bright yellow after less than 30 min air-drying time-length, brown after 1.5≈2 h air-drying, time-length and black after
more than 3 h of air-drying. The color-change process of NSE precipitates from bright yellow to black is progressive. The
phenomennon has not been described previously.

We compared the norepinephrine (NE) induced α1B-adrenoceptor (α1B-AR) expression modulation between two transfected human embryonic kidney (HEK) 293 cell lines in which α1B-AR densities were (6 336±913) and (773±164) fmol.mg-1, respectively. Treatment of cells with NE (10 μmol.L-1) for 48 h decreased high-level expressed α1B-AR density, but increased low-level expressed α1B-AR density. The protein kinase C inhibitor Calphostin C or Ro-31-8220 reversed, and its activator PMA mimicked the NE-induced down-regulation of high-level expressed α1B-AR. Moreover, PMA induced a down-regulation of low-level expressed α1B-AR. The endoplasmic reticulum Ca2+-ATPase inhibitor cyclopiazonic acid (CPA) and the calcium chelator BAPTA/AM did not affect the down-regulation of high-level expressed α1B-AR, but inhibited the up-regulation of low-level expression α1B-AR induced by NE. These results suggest that α1B-adrenoceptor densities at different initial expression levels are differentially regulated by NE and their signal transduction pathways are different. 相似文献

Summary Nocloprost, a new synthetic prostanoid, elicited a vasodilator action when given through the renal artery of the isolated perfused rabbit kidney. Lower concentrations of the compound inhibited the pressor response to sympathetic stimulation or to angiotensin II, but potentiated the pressor effect of exogenous noradrenaline. These results were taken as an evidence that nocloprost prevents the release of neurotransmitter by a presynaptic mechanism.This work was supported by a grant from the Turkish Scientific and Technical Research Council (TAG-578).
