Effect of dipyridamole on glomerular mesangial cell ecto-5′-nucleotidase expression

Although dipyridamole has been extensively studied as an anti-aggregating agent, its mechanism of action has not been elucidated. Cultured mesangial cells were treated with dipyridamole 1–100 M from 6–72 h. Ecto-5-nucleotidase activity approximately doubled (from 115±11 to 226±14 nmol/min/mg) after treatment with 100 M dipyridamole for 72 h. This effect was concentration- and time-dependent. Cycloheximide, an inhibitor of protein synthesis, did not alter basal 5-nucleotidase activity. However, it prevented stimulation by 5 M dipyridamole. Adenosine availability at the receptor sites was increased by dipyridamole and S-(p-nitrobenzyl)-6-thioinosine (NBTI), which inhibit adenosine uptake into the cell. Addition of dipyridamole or NBTI to the adenosine-treated mesagial cells produced an additive increase in ecto-5-nucleotidase activity. Dipyridamole, through its effect on extracellular adenosine and ecto-5-nucleotidase, may have an influence upon regulation of the glomerular microcirculation.

微波辅助萃取腺苷技术的研究

研究了微波提取蛹虫草固体培养物中腺苷的条件.用HPLC法测定提取液中的腺苷含量,通过正交试验,确定了微波提取腺苷的最佳条件.结果表明,在提取液用量10 mL,固液比1∶200,微波强度为中火,处理时间2 min,样品中腺苷的测定值达0.50 mg/g时,微波提取时间仅为超声波提取的1/45,且提取率高于超声波提取.     

Molybdenum cofactor biosynthesis and deficiency

The molybdenum cofactor (Moco) forms the active site of all molybdenum (Mo) enzymes, except nitrogenase. Mo enzymes catalyze important redox reactions in global metabolic cycles. Moco consists of Mo covalently bound to one or two dithiolates attached to a unique tricyclic pterin moiety commonly referred to as molybdopterin (MPT). Moco is synthesized by an ancient and conserved biosynthetic pathway that can be divided into four steps, according to the biosynthetic intermediates precursor Z (cyclic pyranopterin monophosphate), MPT and adenylated MPT. In a fifth step modifications such as attachment of nucleotides, sulfuration or bond formation between Mo and the protein result in different catalytic Mo centers. A defect in any of the steps of Moco biosynthesis results in the pleiotropic loss of all Mo enzyme activities. Human Moco deficiency is a hereditary metabolic disorder characterized by severe neurodegeneration resulting in early childhood death. Recently, a first substitution therapy was established. 家兔在发育过程中外侧腓肠肌内MyHCs变化与诱发电位的关系

 为研究在生后不同年龄组家兔外侧腓肠肌各亚体肌纤维MyHCs变化与诱发电位的关系,探讨生后发育期间肌纤维MyHCs组成和作用差异与表型之间的相关联系,采用电生理记录仪结合十二烷基硫酸钠-聚丙烯酰胺凝胶电泳（SDS PAGE）检测外侧腓肠肌各亚体。结果表明中间亚体在刺激时各年龄组的峰值电压基本相同,而外侧亚体和内侧亚体分别有些变化。同时发现成年家兔的持续时间都较幼年的长,这可能与各亚体的肌纤维型构成比例有很大关系。外侧腓肠肌各亚体的肌球蛋白重链异构体（MyHCs）电泳条带分别显示MyHCsⅡa、Ⅱd（或Ⅱx）、Ⅱb、Ⅰ共4种,对应于骨骼肌纤维表型ⅡA型（FOG型）、ⅡX（FO型）、ⅡB型（FG型）及Ⅰ型（SO型）4类。

腺苷发酵液中有机酸的代谢规律

选取了优化过程中典型的3种含有不同碳源的培养基配方(其中配方A中含有100g／L葡萄糖,配方B中含100g／L蔗糖,配方C中含100g／L蔗糖和10g／L谷氨酸),进行摇瓶发酵生产腺苷(AR),以测定发酵液的生化参数。通过研究发酵液中的有机酸与腺苷的积累在时序性上的代谢规律,在此基础上提出了通过增强TCA循环为手段以提高目的产物产量的观点,即TCA强化论假说。
