Plasmepsins are aspartic proteases involved in the degradation of the host cell hemoglobin that is used as a food source by
the malaria parasite. Plasmepsins are highly promising as drug targets, especially when combined with the inhibition of falcipains
that are also involved in hemoglobin catabolism. In this review, we discuss the mechanism of plasmepsins I–IV in view of the
interest in transition state mimetics as potential compounds for lead development. Inhibitor development against plasmepsin
II as well as relevant crystal structures are summarized in order to give an overview of the field. Application of computational
techniques, especially binding affinity prediction by the linear interaction energy method, in the development of malarial
plasmepsin inhibitors has been highly successful and is discussed in detail. Homology modeling and molecular docking have
been useful in the current inhibitor design project, and the combination of such methods with binding free energy calculations
is analyzed.

Malaria, caused by members of the genusPlasmodia, is still the most prevalent parasitic disease in the world. In an attempt to understand genetic factors conferring resistance to malaria, mouse models of thalassemia, sickle trait, and ankyrin and spectrin deficiency were studied during infection with species of malaria infectious to rodents. Although growth ofP. falciparum is not inhibited in thalassemic erythrocytes in culture, mice carrying a -thalassemia mutation were protected fromPlasmodium chabaudi adami, supporting epidemiologic findings. Transgenic mice expressing s hemoglobin were also significantly protected from two species of rodent malaria. Importantly, a significant role for the spleen in protection in the s transgenic mice was found. Finally, mice deficient in spectrin and ankyrin were studied with respect to their ability to support the growth of malaria. It was found that spectrin deficient mice were almost completely refractory toP. chabaudi adami andP. berghei. These models will allow further study of host factors in resistance to malaria. 相似文献

Continuous natural exposure to Plasmodium transmission by infectious Anopheles mosquitoes leads to a gradual acquisition of immunological competence against malaria. The partial immunity, observed in adolescents and adults living in endemic areas, reduces morbidity and mortality without preventing parasite infection. In experimental animal models, long-lasting sterilizing immunity can be achieved with genetically attenuated Plasmodium liver stages. Can these findings be translated to accomplish sterile protection against natural malaria transmission in the high-risk group, young infants in sub-Saharan Africa? 相似文献
