目的研究Ⅱ型糖尿病患者血清3-硝基酪氨酸(3-NT)、糖化血红蛋白(HbA1c)等生化指标的含量及临床意义.方法按病例对照研究方法,招募确诊的Ⅱ型糖尿病患者64例,正常人对照病例20例.对入选人群进行人体测量、血生化检测和血清3-NT水平的测定,并分析其临床意义.结果糖尿病组血清中3-NT、体质量指数(BMI)、HbA1c、空腹血糖(FPG)、甘油三酯(TG)、肌酐(Cre)、收缩压、舒张压等指标均明显高于正常对照组(P〈0.05),单纯糖尿病组、糖尿病并发症组与正常对照组之间进行比较,各指标如3-NT,BMI,HbA1c,FPG,TG,Cre,收缩压,舒张压均具有统计学意义.糖尿病患者血清3-NT与各变量之间的多元线性逐步回归分析结果显示:糖尿病患者血清3-NT的含量受BMI和CHO影响.结论监测糖尿病患者3-NT,HbA1c,BMI及血脂等生化指标十分重要;联合检测3-NT,HbA1c,FPG在糖尿病及并发症的临床诊断及治疗中具有重要意义.  相似文献

将43只Sprague-Dawly(SD)大鼠随机分为安静对照组、运动对照组、运动低氧暴露低氧2 h组和运动低氧暴露(1+1)h组.运动各组分别进行为期6周递增负荷跑台运动训练,低氧暴露组从第4周起各在运动后进行人工低氧(O_2体积分数为145 mL/L)暴露2 h和(1+1)h.实验结束后测试大鼠红细胞参数与血清丙二醛、血清谷胱甘肽过氧化物酶和血清超氧化物歧化酶含量.6周运动训练后,运动性低血红蛋白现象发生,常氧运动组大鼠血清谷胱甘肽过氧化物酶和血清超氧化物歧化酶显著或非常显著低于常氧安静组,而间歇低氧暴露各组大鼠未见显著下降的现象,其中运动低氧暴露(1+1)h组抗氧化酶水平最高,血清丙二醛含量呈现相反的变化.结果显示,间歇低氧能较为有效地预防运动性低血红蛋白大鼠抗氧化能力的下降,以多次反复的低氧刺激方式效果较佳,其作用机理可能与间歇低氧暴露能提高机体酶性抗氧化系统水平有关.  相似文献

基于阿魏酸钠对血红蛋白催化H2O2氧化L-酪氨酸的抑制作用提出抑制荧光法测定阿魏酸钠的含量,讨论了测定阿魏酸钠的最佳条件.结果表明:在优化的条件下,阿魏酸钠浓度在6.00×10-8~1.60×10-6 mol·L-1范围内呈良好的线性关系,方法的检出限为1.80×10-8 mol·L-1.此方法用于阿魏酸钠片剂中阿魏酸钠含量的分析,获得满意结果.  相似文献

The inherited -hemoglobinopathies (sickle cell disease and thalassemia) are the result of a mutation in the adult () globin gene. The fetal globin chain, encoded by the globin genes, can substitute for the mutated or defective globin chain, but expression of the globin gene is developmentally inactivated prior to birth. Reinducing expression of the normal fetal globin genes is a preferred method of ameliorating sickle cell disease and the thalassemias. Stimulation of as little as 4–8% fetal globin synthesis in the bone marrow can produce >20% fetal hemoglobin in the peripheral circulation, due to enhanced survival of red blood cells containing both sickle and fetal hemoglobin, compared to those containing sickle hemoglobin alone. Butyric acid and butyrate derivatives are generally safe compounds which induce fetal hemoglobin production by stimulating the promoter of the fetal globin genes. An initial trial with the parent compound, delivered as Arginine Butyrate, has demonstrated rapid stimulation of fetal globin expression to levels that have been shown to ameliorate these conditions. Phase 1 trials of an oral butyrate derivative with a long plasma half-life have just begun. These agents now provide a specific new apporach for ameliorating these classic molecular disorders and merit further investigation in larger patient populations.  相似文献

Hemoglobin was used as a mimetic enzyme for peroxidase to catalyze the oxidative reaction of o-phenylenediamine with H2O2 which functioned as an oxidant. The relationship between physicochemical properties of the intermediate and enzymatic activity of hemoglobin was studied. Since the solubility of the intermediate in the reaction is higher in butanol phase than in water phase, the intermediate itself diffused from the aqueous phase to the butanol phase. The experimental results showed that the rate of product and the stability of intermediate were associated with the temperature and the pH value of the buffer. The formation rate of intermediate and half-life period reveal the maximal in pH7, nevertheless, the whole rate of the catalytic reaction is the greatest in pH5, which the ratio of the initial rate in final product formation compared to that intermediate formation is the greatest. 在分析血红蛋白分子与氧分子结合的相互作用特点的基础上,将氧分子与血红蛋白分子结合的过程看作为协同可逆吸附过程.根据协同吸附过程的特点,利用吸附动力学方法,得到了反映血红蛋白分子氧合过程的氧饱合分数与氧分压之间的关系式.从上述关系式得到了与之自洽的推论,在一定的氧分压条件下,血红蛋白分子处于确定的部分氧合构象态.当氧分压的值从小到大变化时,血红蛋白分子将从脱氧构象态经历一系列中间构象态到达完全氧合构象态.这表明s型的氧合曲线实际显示了单个血红蛋白分子的别构效应.  相似文献

Summary Substantial progress has been made in the last ten years in understanding the structural and functional organization of parasitic protozoa and helminths and the complex physiological relationships that exist between these organisms and their hosts. By employing the new powerful techniques of biochemistry, molecular biology and immunology the genomic organization in parasites, the molecular basis of parasite's variation in surface antigens and the biosynthesis, processing, transport and membrane anchoring of these and other surface proteins were extensively investigated. Significant advances have also been made in our knowledge of the specific and often peculiar strategies of intermediary metabolism, cell compartmentation, the role of oxygen for parasites and the mechanisms of antiparasitic drug action. Further major fields of interest are currently the complex processes which enables parasites to evade the host's immune defense system and other mechanisms which have resulted in the specific adaptations which enabled parasites to survive within their host environments. Various approaches in molecular and biochemical parasitology and in immunoparasitology have been proven to be of high potential for serodiagnosis, immunoprophylaxis and drug design.This paper is based on a review presented at a workshop on Molecular Parasitology, organized by the Swiss Society of Tropical Medecine and Parasitology at the University of Neuchâtel, March 1985.  相似文献
