Isolation and partial characterization of cuticular collagen from the parasitic nematodeGaigeria pachyscelis

Summary The cuticle from adultGaigeria pachyscelis was isolated by solubilizing the internal tissues with sodium dodecyl sulphate (SDS) at 37°C. Cuticular protein was extracted with guanidine-HCl and -mercaptoethanol and purified by ammonium sulphate fractionation and DEAE-cellulose chromatography. SDS-polyacrylamide gel electrophoresis of purified protein revealed 2 polypeptides with apparent mol. wts of 58,000 and 74,000. As judged from their hydroxyproline content both of them are collagenous in nature. Results of gel filtration indicate that cuticular collagen exists in two forms, a non-associated form at low concentration and an associated form at high concentration.Acknowledgments. Effect of hormones and cyclic AMP on γ-glutamyltranspeptidase activity of rat mammary gland explants

Summary -Glutamyl transpeptidase activity was assayed in midpregnant rat mammary gland explants at 14, 22 and 38 h, in the presence and absence of insulin, prolactin and corticosterone. With these 3 hormones the explants attained the characteristics of a secretory gland after 22 h of tissue culture, at which time the enzyme exhibited its maximal activity. The addition of dibutyryl cyclic AMP in the presence of the 3 hormones produced a significant increase in enzyme activity, which was maximal with a 1 mM concentration of the cyclic nucleotide. A similar effect was observed when theophylline or theophylline plus dibutyryl cyclic AMP were added to the culture medium.This work was supported by grant B1138-8333 from the Departamento de Desarrollo de la Investigación, Universidad de Chile.     

Secreted oleanolic acid on the cuticleOlea europaea (Oleaceae); a chemical barrier to fungal attack

Summary The leaf of the bitter oliveOlea europaea (Oleaceae) is heavily coated by almost pure oleanolic acid, which forms part of a multichemical defense against fungal attack.We thank Mr K. A. Hoelmer for taking the electron micrographs and also Professor T. Kamikawa for NMR measurements.     

Excretion of acid hydrolases during molting inPhilosamia ricini

Summary The level of the acid hydrolases -glucuronidase (EC 3.2.3.11), acid phosphatase (EC 3.1.3.2) and acid protease (EC 3.4.4) was studied during larval growth and molting inP. ricini. The level of activity of these enzymes remained low during larval growth; however, the level increased sharply at the time of molting and declined sharply thereafter in the newly ecdysed insect. Interestingly, the diminished activity of these enzymes was almost quantitatively recovered in the cast-off cuticle. The excretion of acid hydrolases through the cast-off cuticle has hitherto not been reported in insects during molting.     

‘Chloride-cell’ — like mitochondria-rich cells of salamander larva gill epithelium

Summary Two types of mitochondria-rich cells (MRC) are described ultrastructurally in the gill epithelium of salamander larva. They resemble MRC found in larval ventral epidermis. Histochemical localization of carbonic anhydrase indicated numerous positive reacting cells, most of them flask-shaped. Morphological and functional similarities to fish chloride cells are discussed.     

Estrogen and accessory sex gland lipids

Summary Administration of estradiol-17 elicited differential responses on accessory sex glands of rats. In caput epididymis, the estrogen treatment led to an accumulation of glycerides and phospholipids while in cauda epididymis, there was no significant change. However, in seminal vesicles, depletion of phospholipids was observed In prostate, the treatment, resulted in an accumulation of glycerides.Part of Ph.D. Thesis. Umapathy, E., Thesis. Oxygen consumption in rat skeletal muscle at various rates of oxygen delivery

Summary Resting oxygen consumption of the vascularly isolated gracilis anticus muscle of the rat perfused by natural circulation via the femoral artery with diluted or undiluted blood depends on oxygen delivery (the product of flow rate and arterial oxygen concentration-DO₂) only when DO₂ falls below 16 l/min/g.     

High-affinity binding ofβ-alanine to cerebral synaptosomes might involve glycine-receptors

Summary High-affinity, Na⁺-independent binding of -alanine to a synaptosomal fraction of rat brain was potently inhibited by glycine and by some other -amino acids, but not by taurine or GABA. This binding mechanism, which was also sensitive to both bicuculline and strychnine, might involve synaptic receptors for both -alanine and glycine.This study was supported in part by Centro Ramón y Cajal and Fundacion Juan March.     

Control of obesity inA vy /a mice by 5α-androstan-17-one

Summary Treating VY/WfL-A ^vy /a mice with 5-androstan-17-one, a mammalian glucose-6-phosphate dehydrogenase inhibitor, prevented the mice from becoming obese. The weight difference between treated and controlA ^vy /a mice was mainly due to a decreased accumulation of triacylglycerol. The compound did not suppress appetite, had no detectable toxicity and did not affect the lipogenesis rates in the liver and carcass. The weight-controlling effect of 5-androstan-17-one inA ^vy /a mice was reversible upon withdrawal of treatment.The authors wish to thank Mr W.R. Gibson and Drs C.G. Culbertson and P.N.
