Determination of the absolute handedness of knots and catenanes of DNA   总被引:4,自引:0,他引:4  
DNA winds about itself in a right-handed or left-handed fashion at several structural levels. The double helix is generally right-handed and is given a (+) sign by convention, whereas supercoiling of the helix axis is always (-) in the cell. The winding in higher -order forms such as knots and catenanes is unknown, and this has impeded elucidation of the mechanisms of their formation and resolution by replication, recombination and topoisomerase action. We introduce here a procedure for determining the handedness of DNA winding by inspection of electron micrographs of DNA molecules coated with Escherichia coli RecA protein. We demonstrate the validity of the method and show that DNA topoisomerase I of E. coli generates an equal mixture of (+) and (-) duplex DNA knots, and that one product of recombination by resolvase of transposon Tn3 (refs 8, 9) is a catenane of uniquely (+) sign.  相似文献

Results from a prospective sero-epidemiological study initiated in Uganda in 1971 indicate that children with high antibody titres to Epstein-Barr virus structural antigens are at high risk of developing Burkitt's lymphoma. These findings strongly support a causal relationship between the Epstein-Barr virus and Burkitt's lymphoma but suggest that the oncogenic potential of the virus is realised only in exceptional circumstances.  相似文献

We examined algal flora in the aquatic system of Devils Hole, Nevada. The water is seasonally warm, near neutral in pH, and rich in dissolved carbonate, indicative of deeply circulating groundwater flowing through carbonate rock. Most algae were benthic, with only a few planktonic representatives. Eighty-four terminal identifications in 44 genera were recorded, with diatoms and blue-green algae the most speciose groups. Diatoms were major contributors to the grazing food web. Large, mat-forming filamentous algae have an important influence on the physicochemical and general structure of the benthic substrates in Devils Hole.  相似文献

Chemicals can elicit T-cell-mediated diseases such as allergic contact dermatitis and adverse drug reactions. Therefore, testing of chemicals, drugs and protein allergens for hazard identification and risk assessment is essential in regulatory toxicology. The seventh amendment of the EU Cosmetics Directive now prohibits the testing of cosmetic ingredients in mice, guinea pigs and other animal species to assess their sensitizing potential. In addition, the EU Chemicals Directive REACh requires the retesting of more than 30,000 chemicals for different toxicological endpoints, including sensitization, requiring vast numbers of animals. Therefore, alternative methods are urgently needed to eventually replace animal testing. Here, we summarize the outcome of an expert meeting in Rome on 7 November 2009 on the development of T-cell-based in vitro assays as tools in immunotoxicology to identify hazardous chemicals and drugs. Neutron stars are composed of the densest form of matter known to exist in our Universe, the composition and properties of which are still theoretically uncertain. Measurements of the masses or radii of these objects can strongly constrain the neutron star matter equation of state and rule out theoretical models of their composition. The observed range of neutron star masses, however, has hitherto been too narrow to rule out many predictions of 'exotic' non-nucleonic components. The Shapiro delay is a general-relativistic increase in light travel time through the curved space-time near a massive body. For highly inclined (nearly edge-on) binary millisecond radio pulsar systems, this effect allows us to infer the masses of both the neutron star and its binary companion to high precision. Here we present radio timing observations of the binary millisecond pulsar J1614-2230 that show a strong Shapiro delay signature. We calculate the pulsar mass to be (1.97?±?0.04)M(⊙), which rules out almost all currently proposed hyperon or boson condensate equations of state (M(⊙), solar mass). Quark matter can support a star this massive only if the quarks are strongly interacting and are therefore not 'free' quarks.  相似文献

Two pulsars (PSR J0737-3039A and B) were recently discovered in highly relativistic orbits around one another. The system contains a rapidly rotating pulsar with a spin period of 22.7 ms and a slow companion with a spin period of 2.77 s, referred to here as 'A' and 'B', respectively. A unique property of the system is that the pulsed radio flux from B increases systematically by almost two orders of magnitude during two short portions of its orbit. Here we report a geometrical model of the system that simultaneously explains the intensity variations of B and provides constraints on the spin axis orientation and emission geometry of A. Our model assumes that B's pulsed radio flux increases when illuminated by emission from A. We predict that A's pulse profile will evolve considerably over the next several years owing to geodetic precession until it disappears entirely in 15-20 years.  相似文献
