An exploratory study was made of the mechanisms underlying the toxic action of carbadox in young pigs: dehydration, loss of appetite and at autopsy seemingly specific and selective structural alterations of the glomerular zone of the adrenal cortex. Administration of carbadox in the feed, in dosages of 150 ppm (approximately 6 mg X kg-1 b. wt X day-1) caused a rapid decline in the plasma aldosterone levels (to 10% of control) followed by significant changes in the sodium and potassium levels in blood. Characteristic for the toxic action of carbadox are the rapid and seemingly selective and specific alterations in the aldosterone-releasing zona glomerulosa of the adrenals. Our results indicate that with carbadox a functional and possibly reversible extirpation of the adrenal zona glomerulosa can be achieved in pigs. 相似文献

In excised inside-out membrane patches of the human colon carcinoma HT-29cl.19A cells a large conductance (373±10 pS) chloride channel was found. Channel activity could only be observed after excision of patches from cells incubated with calcium ionophore. The channel was never observed in cell-attached patches. The channel was strongly voltage dependent, being open only between +30 and –30 mV clamp potentials. The selectivity sequence among anions, deduced from reversal potentials, was I>Br>Cl>F>gluconate. The PNa/PCl was 0.09. Although a similar type of channel, has been described earlier, this is the first report stating its appearance in patches of intestinal epithelial cells requiring the combined action of Ca2+ ionophore and excision, suggesting its control by an intracellular compound. 相似文献

Summary Quercetin is an effective inhibitor of human myeloperoxidase (MPO) activity, both with purified enzyme (IC50=3.5 M) and in a system using stimulated human neutrophils. Quercetin is significantly more potent than three other related compounds (rutin, rutin sulfate and troxerutin) and than methimazole, a previously-known myeloperoxidase inhibitor. The inhibitory activity of quercetin is of the competitive type. Moreover, quercetin is directly able to scavenge hypochlorous acid (HOCl), a chlorinated species generated by the, MPO/H2O2/Cl– system. 相似文献

Dominantly inherited familial amyloidosis, Finnish type (FAF) is caused by the accumulation of a 71-amino acid amyloidogenic fragment of mutant gelsolin (GSN). FAF is common in Finland but is very rare elsewhere. In Finland and in two American families, the mutation is a G654A transition leading to an Asp to Asn substitution at residue 187. We found the same mutation in a Dutch family but a Danish FAF family had a G654T mutation, predicting Asp to Tyr at residue 187. We also found the G654T transversion in a Czech family. Using GSN polymorphisms, different haplotypes were found in the Danish and Czech families. We conclude that substitution of the uncharged Asn or Tyr for the acidic Asp at residue 187 creates a conformation that may be preferentially amyloidogenic for GSN. 相似文献

TSV5 clone 2 cells in normal conditions of culture contain only an expressed RNA virus (R-type virus). However, exposure of the cells to 5-bromo-2'-deoxyuridine with dexamethasone, induced synthesis of a syncitium-forming ("Foamy") virus. In other hamster cell lines, the same treatment fails to induce a "foamy" virus. The origin of this "foamy" virus is discussed. 相似文献

Résumé Dans 18 cas de neuroblastome humain la noradrénaline (NA) et la dopamine--hydroxylase présentent des profils de distribution intracellulaire identiques, suggérant un stockage dans la même particule. Le contenu en NA de ces particules n'est pas inférieur à celui des particules du nerf splénique; on peut en conclure qu'un stockage défectueux de NA dans les cas de neuroblastome est peu probable.
