Genome sequence and comparative analysis of the model rodent malaria parasite Plasmodium yoelii yoelii

Species of malaria parasite that infect rodents have long been used as models for malaria disease research. Here we report the whole-genome shotgun sequence of one species, Plasmodium yoelii yoelii, and comparative studies with the genome of the human malaria parasite Plasmodium falciparum clone 3D7. A synteny map of 2,212 P. y. yoelii contiguous DNA sequences (contigs) aligned to 14 P. falciparum chromosomes reveals marked conservation of gene synteny within the body of each chromosome. Of about 5,300 P. falciparum genes, more than 3,300 P. y. yoelii orthologues of predominantly metabolic function were identified. Over 800 copies of a variant antigen gene located in subtelomeric regions were found. This is the first genome sequence of a model eukaryotic parasite, and it provides insight into the use of such systems in the modelling of Plasmodium biology and disease.     

Mechanisms of self-incompatibility in flowering plants

Self-incompatibility is a widespread mechanism in flowering plants that prevents inbreeding and promotes outcrossing. The self-incompatibility response is genetically controlled by one or more multi-allelic loci, and relies on a series of complex cellular interactions between the self-incompatible pollen and pistil. Although self-incompatibility functions ultimately to prevent self-fertilization, flowering plants have evolved several unique mechanisms for rejecting the self-incompatible pollen. The self-incompatibility system in the Solanaceae makes use of a multi-allelic RNase in the pistil to block incompatible pollen tube growth. In contrast, the Papaveraceae system appears to have complex cellular responses such as calcium fluxes, actin rearrangements, and programmed cell death occurring in the incompatible pollen tube. Finally, the Brassicaceae system has a receptor kinase signalling pathway activated in the pistil leading to pollen rejection. This review highlights the recent advances made towards understanding the cellular mechanisms involved in these self-incompatibility systems and discusses the striking differences between these systems. Flow shop下批调度与设备维护的联合优化

批调度属于经典的调度问题之一,但在目前的批调度研究过程中,并未考虑到设备维护与批调度之间的相互干扰。以flow shop下批调度为研究对象,同时兼顾设备维护对调度的影响,建立了批调度与设备维护的联合优化模型。同时依据问题的特性,采用改进的遗传算法对问题进行求解。最后通过数值仿真实验验证了本模型和算法的有效性。

利用遗传算法在处理过程中不依赖问题的种类,并具有较强鲁棒性等特点,提出了一种基于交互式的求解多目标随机规划的遗传算法,算法的意思是,结合小生境技巧和构造Pareto选优过滤器的手段,通过与决策者的反复交互对话,最后得到使决策者满意的问题的Pateto有效解集。

用生物素标记核酸探针检测鼠金黄色葡萄球菌的研究

利用聚合酶链反应技术从金黄色葡萄球菌基因组DNA中扩增出 6 78bp耐热核酸酶nuc基因特异性片段 ,经生物素标记后作为核酸斑点杂交试验的探针 ,对实验动物的金黄色葡萄球菌及其临床样品进行了检测。结果显示 ,标记探针与金黄色葡萄球菌DNA呈杂交阳性 ,而与链球菌、大肠埃希氏菌和巴氏杆菌DNA呈杂交阴性 ,斑点杂交的检测灵敏度为 1pg基因组DNA。用斑点杂交试验和PCR对 2 4 0份临床样品进行了检测 ,检出率为 2 9% (7 2 4 0 ) ,符合率为 10 0 %。这些试验结果表明 ,研究制备的核酸探针及建立的斑点杂交试验具有较高的特异性和敏感性 ,可用于实验动物金黄色葡萄球菌的快速检测     

原子吸收光谱法测定七种抗癌中草药微量元素

建立微波消解火焰原子吸收法测定七种抗癌中草药中K、Na、Mg、Fe、Mn、Zn、Cu、Cr8种元素的方法。探讨了微波消解试剂和消解条件,0.5g样品用8mL硝酸和2mL过氧化氢分解。结果表明,该法准确、快速、简便,8种金属元素的检出范围为0.006～0.035μg/mL;线性关系良好,相关系数r≥0.9990;回收率为96.00%～103.98%;RSD<2%。
