The proximal NPXY and distal NPXYXXL motifs in the intracellular domain of LRP1 play an important role in regulation of the
function of the receptor. The impact of single and double inactivating knock-in mutations of these motifs on receptor maturation,
cell surface expression, and ligand internalization was analyzed in mutant and control wild-type mice and MEFs. Single inactivation
of the proximal NPXY or in combination with inactivation of the distal NPXYXXL motif are both shown to be associated with
an impaired maturation and premature proteasomal degradation of full-length LRP1. Therefore, only a small mature LRP1 pool
is able to reach the cell surface resulting indirectly in severe impairment of ligand internalization. Single inactivation
of the NPXYXXL motif revealed normal maturation, but direct impairment of ligand internalization. In conclusion, the proximal
NPXY motif proves to be essential for early steps in the LRP1 biosynthesis, whereas NPXYXXL appears rather relevant for internalization. 相似文献

The evolution of colorectal cancer suggests the involvement of many genes. To identify new drivers of intestinal cancer, we performed insertional mutagenesis using the Sleeping Beauty transposon system in mice carrying germline or somatic Apc mutations. By analyzing common insertion sites (CISs) isolated from 446 tumors, we identified many hundreds of candidate cancer drivers. Comparison to human data sets suggested that 234 CIS-targeted genes are also dysregulated in human colorectal cancers. In addition, we found 183 CIS-containing genes that are candidate Wnt targets and showed that 20 CISs-containing genes are newly discovered modifiers of canonical Wnt signaling. We also identified mutations associated with a subset of tumors containing an expanded number of Paneth cells, a hallmark of deregulated Wnt signaling, and genes associated with more severe dysplasia included those encoding members of the FGF signaling cascade. Some 70 genes had co-occurrence of CIS pairs, clustering into 38 sub-networks that may regulate tumor development. 相似文献

We carried out a genome-wide association study of breast cancer predisposition with replication and refinement studies involving 6,145 cases and 33,016 controls and identified two SNPs (rs4415084 and rs10941679) on 5p12 that confer risk, preferentially for estrogen receptor (ER)-positive tumors (OR = 1.27, P = 2.5 x 10(-12) for rs10941679). The nearest gene, MRPS30, was previously implicated in apoptosis, ER-positive tumors and favorable prognosis. A recently reported signal in FGFR2 was also found to associate specifically with ER-positive breast cancer. 相似文献

Quantum superposition lies at the heart of quantum mechanics and gives rise to many of its paradoxes. Superposition of de Broglie matter waves' has been observed for massive particles such as electrons, atoms and dimers, small van der Waals clusters, and neutrons. But matter wave interferometry with larger objects has remained experimentally challenging, despite the development of powerful atom interferometric techniques for experiments in fundamental quantum mechanics, metrology and lithography. Here we report the observation of de Broglie wave interference of C(60) molecules by diffraction at a material absorption grating. This molecule is the most massive and complex object in which wave behaviour has been observed. Of particular interest is the fact that C(60) is almost a classical body, because of its many excited internal degrees of freedom and their possible couplings to the environment. Such couplings are essential for the appearance of decoherence, suggesting that interference experiments with large molecules should facilitate detailed studies of this process. 相似文献

RNA interference is an evolutionarily conserved surveillance mechanism that responds to double-stranded RNA by sequence-specific silencing of homologous genes. Here we show that transgene expression can be suppressed in adult mice by synthetic small interfering RNAs and by small-hairpin RNAs transcribed in vivo from DNA templates. We also show the therapeutic potential of this technique by demonstrating effective targeting of a sequence from hepatitis C virus by RNA interference in vivo. 相似文献

Ginkgo biloba extract is known to be efficient in diseases associated with free radical generation. The purpose of this work was to study, under in vitro conditions, the action of Ginkgo biloba extract (Gbe) against superoxide anion (O2-.), which is directly or indirectly implicated in cell damage. Gbe appears to have both an O2-. scavenging effect and also a superoxide dismutase activity. Its antiradical effect was demonstrated by low temperature electron spin resonance and in a non-enzymatic system (phenazine methosulfate-NADH), and its enzymatic activity was shown by polarographic determination. 相似文献
