高护专业基础实验教学的分析与探讨

目的探索高护专业基础实验教学最佳方法.方法通过问卷调查分析现有基础实验教学的优劣.结果实验课收效以验证理论知识为主,同时强化了学生的操作能力,提高了观察能力,但对综合能力及创新能力提高不够;对实验内容设置倾向综合性实验.结论应深化基础实验教学改革,提高基础实验教学质量及学生的综合能力.     

Characterization and expression of a cDNA, AmphiSDHD,encoding the amphioxus cytochrome b small subunit in mitochondrial succinate-ubiquinone oxidoreductase

In this study, an amphioxus cDNA, AmphiSDHD, encoding the cytochrome b small subunit in mitochondrial succinate-ubiquinone oxidoreductase, was isolated from the gut cDNA library of amphioxus Branchiostoma belcheri tsingtauense. It is 1429 bp in length, with an open reading frame of 465 bp coding for a protein of 154 amino acids. The deduced protein contains a mitochondrial targeting presequence of 65 amino acids rich in basic residues like arginine and hydroxy residues such as serine and threonine. Alignment of the amino acid sequences of AmphiSDHD and other eukaryotic SDHD proteins showed that AmphiSDHD has three transmembrane segments, and includes two histidine residues in the second transmembrane segment that are the putative binding sites for the heme b molecule. The phylogenetic tree constructed suggests that AmphiSDHD appears more closely related to vertebrate SDHD proteins than invertebrate ones. Northern blotting demonstrated that AmphiSDHD is ubiquitously expressed in amphioxus, being in line with the fact that SDHD is a house-keeping protein.

回顾、反思、求索、徘徊、出游——《离骚》的分段及其叙事与幻想

关于《离骚》的分段，自古以来众说纷纭，莫衷一是。迄今流传最广、影响最大的三大段、两部分的分法，仍然留下未辨过脉之失和割裂全诗之憾。根据《离骚》诗中叙述的情节，笔者认为将全诗分为五大段才为合理，即依次为回顾、反思、求索、徘徊、出游。也可以分为两部分，即第一、二大段为前半部分，写诗人有生以来的不懈追求与奋斗，以及遭致失败之后所引发的检视与思考，第三、四、五大段为后半部分，写诗人矢志不渝地继续努力，以及努力无果之后所产生的矛盾与苦闷。全诗在体裁形式和表现方式方面表现出两大基本特色，其一是结合叙事以抒情，在体裁形式上通篇都是叙事的结构，其二是借助“虚写”以“写实”，在表现方式上通篇都是浪漫的幻想。     

Phase relationships in 500℃ isothermal section of Ho-Ni-Ti Ternary system and the fundamental characteristics of the related compounds

By means of X-ray powder diffraction (powder XRD), differential thermal analysis (DTA), scanning electron microscopy (SEM) and X-ray electron dispersed spectrum microanalysis (EDS), the isothermal section at 500℃ in Ho-Ni-Ti ternary system is determined. Intermetallic phases in this section and the relationships among them are studied. In addition, the characteristics of some of these compounds are analyzed. This section consists of 13 singlephase regions, 23 two-phase regions and 11 three-phase regions. The existence of 10 binary compounds is confirmed.No ternary. compound is found.

C-banding pattern and nucleolar organizer regions of Cynoglossus semilaevis Günther, 1873

The C-banding pattern and nucleolar organizer regions of the metaphase chromosomes of Cynoglossus semilaevis are reported. The interstitial regions in all chromosomes including the pair of sex chromosomes had positive C-bands, and the 6th and 12th pairs of chromosomes were entirely stained in most cases. Some chromosomes such as the 1st, 2nd, 7th, 8th, 9th and 10th pairs showed C-bands at centromeric or distal ends. The C-banding heretochromatin occupies 30.03% of the total chromosome surface in C. semilaevis, which is similar to that of amphibians such as Mixophyes fasciolatus (30. 2% ) and M. schevilli (20. 7% ), but is rather lower than that of cephalochordate Branchiostoma belcheri (54. 3 % ). Silver staining revealed a single pair of nucleolar organizer regions ( NORs) located in the telomeric regions of Chromosome 2. The association of NORs with heterochromatin observed in vertebrates also occurs in C. semilaevis as the telomeric regions of Chromosome 2 are always stained positively with C-banding.
