The present study deals with a possible mechanism controlling the transport of manganese (Mn), an essential trace element, from the circulation to the thyroid. Mice were pretreated with propylthiouracil (PTU) or triiodothyronine (T3), and a measurement of the thyroid:serum concentration ratio (T/S) of radioactive manganese (54Mn) was carried out. The T/S of54Mn was greatly enhanced by PTU, but reduced by T3. Several methods were used to demonstrate that the T/S of54Mn depends upon the level of thyroid-stimulating hormone (TSH) in the serum. First, bovine TSH was injected into mice; an increase in the T/S resulted. Secondly, serum thyroxine and T3 levels measured by radioimmunoassay (RIA) suggested that PTU produced an increase in serum TSH and T3 a decrease. However, direct measurement of mouse TSH by RIA for rat TSH failed to produce proof of any changes in TSH level, owing to poor cross-reactivity. Taking all the information into account, it is concluded that Mn-transport into the thyroid is controlled by the thyroid state.  相似文献

The protein kinase MAP kinase, also called MAP2 kinase, is a serine/threonine kinase whose activation and phosphorylation are induced by a variety of mitogens, and which is thought to have a critical role in a network of protein kinases in mitogenic signal transduction. A burst in kinase activation and protein phosphorylation may also be important in triggering the dramatic reorganization of the cell during the transition from interphase to mitosis. The interphase-metaphase transition of microtubule arrays is under the control of p34cdc2 kinase, a central control element in the G2-M transition of the cell cycle. Here we show that a Xenopus kinase, closely related to the mitogen-activated mammalian MAP kinase, is phosphorylated and activated during M phase of meiotic and mitotic cell cycles, and that the interphase-metaphase transition of microtubule arrays can be induced by the addition of purified Xenopus M phase-activated MAP kinase or mammalian mitogen-activated MAP kinase to interphase extracts in vitro.  相似文献

Summary A methanolic extract ofCitrus unshiu induces oviposition by females of aCitrus-feeding swallowtail butterfly,Papilio xuthus L. The chemical factors responsible for stimulating oviposition were isolated and characterized as 5-hydroxy-N-methyltryptamine, adenosine, vicenin-2, narirutin, hesperidin and rutin. An artificial blend of these six components elicited significant oviposition behavior, apparently identical to that induced by contact with intactCitrus leaves.  相似文献

Gelatinous drop-like corneal dystrophy (GDLD; OMIM 204870) is an autosomal recessive disorder characterized by severe corneal amyloidosis leading to blindness, with an incidence of 1 in 300,000 in Japan. Our previous genetic linkage study localized the gene responsible to a 2.6-cM interval on chromosome 1p. Clinical manifestations, which appear in the first decade of life, include blurred vision, photophobia and foreign-body sensation. By the third decade, raised, yellowish-grey, gelatinous masses severely impair visual acuity, and lamellar keratoplasty is required for most patients. Here we report DNA sequencing, cDNA cloning and mutational analyses of four deleterious mutations (Q118X, 632delA, Q207X and S170X) in M1S1 (formerly TROP2 and GA733-1), encoding a gastrointestinal tumour-associated antigen. The Q118X mutation was the most common alteration in the GDLD patients examined, accounting for 33 of 40 (82.5%) disease alleles in our panel of families. Protein expression analysis revealed aggregation of the mutated, truncated protein in the perinuclear region, whereas the normal protein was distributed diffusely in the cytoplasm with a homogenous or fine granular pattern. Our successful identification of the gene that is defective in GDLD should facilitate genetic diagnosis and potentially treatment of the disease, and enhance general understanding of the mechanisms of amyloidosis.  相似文献

Reactive oxygen species (ROS) have been identified as central mediators in certain signalling events. In the heart, ROS have important functions in ischaemia/reperfusion-induced cardiac injury and in cytokine-stimulated hypertrophy. Extracellular signal-regulated kinase (ERK) is one of the ROS-responsive serine/threonine kinases. Previous studies showed that tyrosine kinases and small G proteins are involved in the activation of ERK by ROS; however, the initial target protein of ROS that leads to ERK activation remains unknown. Here we show that inhibition of the betagamma-subunit of G protein (G betagamma) attenuates hydrogen peroxide (H2O2)-induced ERK activation in rat neonatal cardiomyocytes. The G betagamma-responsive ERK activation induced by H2O2 is independent of ligands binding to Gi-coupled receptors, but requires phosphatidylinositol-3-kinase and Src activation. In in vitro studies, however, treatment with H2O2 increases [35S]GTP-gammaS binding to cardiac membranes and directly activates purified heterotrimeric Gi and Go but not Gs. Analysis using heterotrimeric Go and its individual subunits indicates that H2O2 modifies G alpha(o) but not G betagamma, which leads to subunit dissociation. We conclude that G alpha(i) and G alpha(o) are critical targets of oxidative stress for activation of ERK.  相似文献

The effects of a 50 Hz magnetic field on experimentally-induced inflammation in rats were studied. Carrageenan edema was inhibited significantly by exposure to magnetic field for 3 h. Adjuvant-induced arthritis in rats was also suppressed by the magnetic field.  相似文献

Genetic differences between 20 species of cichlid fish, representing all the 12 tribes proposed for the cichlid fish fauna of Lake Tanganyika, were studied by allozyme electrophoresis. Most species were genetically very differentiated from each other. Phylogenetic analysis based on the allozyme data indicated that at least seven old, ancestral lineages have contributed to the present cichlid fauna of the lake. Lake Tanganyika, the oldest of the rift-valley lakes, can be recognized as an evolutionary reservoir of major lineages of cichlids in East Africa.  相似文献

The world is full of surfaces, and by looking at them we can judge their material qualities. Properties such as colour or glossiness can help us decide whether a pancake is cooked, or a patch of pavement is icy. Most studies of surface appearance have emphasized textureless matte surfaces, but real-world surfaces, which may have gloss and complex mesostructure, are now receiving increased attention. Their appearance results from a complex interplay of illumination, reflectance and surface geometry, which are difficult to tease apart given an image. If there were simple image statistics that were diagnostic of surface properties it would be sensible to use them. Here we show that the skewness of the luminance histogram and the skewness of sub-band filter outputs are correlated with surface gloss and inversely correlated with surface albedo (diffuse reflectance). We find evidence that human observers use skewness, or a similar measure of histogram asymmetry, in making judgements about surfaces. When the image of a surface has positively skewed statistics, it tends to appear darker and glossier than a similar surface with lower skewness, and this is true whether the skewness is inherent to the original image or is introduced by digital manipulation. We also find a visual after-effect based on skewness: adaptation to patterns with skewed statistics can alter the apparent lightness and glossiness of surfaces that are subsequently viewed. We suggest that there are neural mechanisms sensitive to skewed statistics, and that their outputs can be used in estimating surface properties.  相似文献

Controlling a phase of matter by coherently manipulating specific vibrational modes has long been an attractive (yet elusive) goal for ultrafast science. Solids with strongly correlated electrons, in which even subtle crystallographic distortions can result in colossal changes of the electronic and magnetic properties, could be directed between competing phases by such selective vibrational excitation. In this way, the dynamics of the electronic ground state of the system become accessible, and new insight into the underlying physics might be gained. Here we report the ultrafast switching of the electronic phase of a magnetoresistive manganite via direct excitation of a phonon mode at 71 meV (17 THz). A prompt, five-order-of-magnitude drop in resistivity is observed, associated with a non-equilibrium transition from the stable insulating phase to a metastable metallic phase. In contrast with light-induced and current-driven phase transitions, the vibrationally driven bandgap collapse observed here is not related to hot-carrier injection and is uniquely attributed to a large-amplitude Mn-O distortion. This corresponds to a perturbation of the perovskite-structure tolerance factor, which in turn controls the electronic bandwidth via inter-site orbital overlap. Phase control by coherent manipulation of selected metal-oxygen phonons should find extensive application in other complex solids--notably in copper oxide superconductors, in which the role of Cu-O vibrations on the electronic properties is currently controversial.  相似文献
