This paper introduces discrete Euler processes and shows their application in detecting and forecasting cycles in non‐stationary data where periodic behavior changes approximately linearly in time. A discrete Euler process becomes a classical stationary process if ‘time’ is transformed properly. By moving from one time domain to another, one may deform certain time‐varying data to non‐time‐varying data. With these non‐time‐varying data on the deformed timescale, one may use traditional tools to do parameter estimation and forecasts. The obtained results then can be transformed back to the original timescale. For datasets with an underlying discrete Euler process, the sample M‐spectrum and the spectra estimator of a Euler model (i.e., EAR spectral) are used to detect cycles of a Euler process. Blood vessel regression is an essential process for ensuring blood vessel networks function at optimal efficiency and for matching blood supply to the metabolic needs of tissues as they change over time. Angiogenesis is the major mechanism by which new blood vessels are produced, but the vessel growth associated with angiogenesis must be complemented by remodeling and maturation events including the removal of redundant vessel segments and cells to fashion the newly forming vasculature into an efficient, hierarchical network. An important and difficult issue is simultaneously identifying the detailed locations of various molecules on the cell surface, as this identification requires a synergistic effect between more than one molecule in a living cell. Au nanoparticles (NPs) with different shapes can be readily recognised under low vacuum scanning electron microscopy (lvSEM). Anisotropic Au nanorods (NRs) possess unique surface plasmon resonance (SPR) properties, which can be further utilised for two photon luminescence (TPL) and other optical imaging techniques. In this paper, Au NRs and Au nanooctahedra (Au NOs) are introduced as biomarkers for ICAM-1 and Integrin β1. 研究混凝土粗集料与砂浆在宏观尺度上的界面特性,并采用砂浆中的砂粒与粗集料的平均间距(简称界面平均间距)进行表征.结合雅砻江锦屏大坝集料品种,研究了球形和片状2种粗集料形状、2组粗集料尺寸,以及粉煤灰和石灰石粉的2个掺量(10%,20%)对界面平均间距的影响.结果表明,采用界面区砂粒与粗集料的平均间距能较好地反映界面区的宏观离析和均质性,测量使用也较简便.对于球形集料,大、小集料上方与下方的界面平均间距无显著差异,表明界面较均匀,离析不明显;而对于片状集料,随着尺寸增大,上、下方的界面平均间距的差异增大,表明大的片状集料下方产生了明显的离析.研究结果还表明,采用粉煤灰或石灰石粉部分等质量取代水泥,可以改善片状集料下方浆体的离析. 相似文献

Atomic force microscopy (AFM) was used to locate CD20 molecules on the surface of lymphoma Raji cells. Rituximab (a monoclonal antibody against CD20) molecules were linked onto the AFM tip via a polyethylene glycol (PEG) linker. Raji cells were adsorbed onto glass slides coated with poly-L-lysine. First, the CD20 distribution in a local area of the cell surface was visualized using the AFM lift scan mode. Second, 16 × 16 force curves were obtained from the same cell area to construct the CD20-rituximab binding force map. Finally, free rituximab was added to block the CD20 molecules on the cell surface and the lift phase image and CD20-rituximab force map were obtained again. The experimental results indicated that when the lift height was greater than the length of the PEG linker, no recognition sites were observed in the lift phase image. However, as the lift height decreased to the length of the PEG linker, some recognition sites were observed in the lift phase image and these sites were generally consistent with the pixels in the force map. After blocking, both the recognition sites in the lift phase image and the gray pixels in the binding force map decreased markedly. These results can improve our understanding of the distribution of protein molecules on the cell surface and facilitate further investigations into cellular functions. 相似文献

Ion transport proteins must remove an ion's hydration shell to coordinate the ion selectively on the basis of its size and charge. To discover how the K+ channel solves this fundamental aspect of ion conduction, we solved the structure of the KcsA K+ channel in complex with a monoclonal Fab antibody fragment at 2.0 A resolution. Here we show how the K+ channel displaces water molecules around an ion at its extracellular entryway, and how it holds a K+ ion in a square antiprism of water molecules in a cavity near its intracellular entryway. Carbonyl oxygen atoms within the selectivity filter form a very similar square antiprism around each K+ binding site, as if to mimic the waters of hydration. The selectivity filter changes its ion coordination structure in low K+ solutions. This structural change is crucial to the operation of the selectivity filter in the cellular context, where the K+ ion concentration near the selectivity filter varies in response to channel gating. 相似文献
