Identification of an AFLP marker linked to the stripe rust resistance geneYr10 in wheat

AFLP analysis of near-isogenic lines of the stripe rust resistance gene Yr10 was carried out with 6 PstⅠ- primers and 10 TaqⅠ-primers with the donor parent of Yr10 gene as the check. A total of about 4200 distinguishable bands were amplified, of which 5 were stable. The genetic linkage of the 5 polymorphic DNA fragments with the target gene were tested preliminarily on a segregating F₂ population derived from a cross between the gene donor parent “Moro” and susceptible cultivar “Mingxian 169”. The DNA fragment PT0502 was found closely linked to the Yr10 gene and cloned and sequenced. Based on the sequence specific primers for PCR were designed and synthesized. Genetic linkage analysis with 195 segregating F₂ plants indicated that the genetic distance was 0.5 cM between the main product SC₂₀₀ fragment produced by PCR with the primers and the Yr10 gene. The primers can be used to detect the Yr10 gene quickly, effectively and exactly.     

A novel method for purification of recombinant adenoassociated virus vectors on a large scale

A novel method for recombinant adeno-associated virus (rAAV) purification on large scale is described. The method involves three steps, including chloroform treatment, PEG/NaCl precipitation and chloroform extraction. The whole procedure can be performed in four hours. Using this purification method, we can reproducibly obtain, from 4×10⁹ of proviral cells cultured in roller bottles, purified rAAV-GFP stocks with titers of around 5×10¹³ particles/mL and purity greater than 95%. The infectious titers of the vector stocks were up to 2×10 TU/mL, thus particle-to-infectivity rate was about 25. Under an electronic microscope, most rAAV particles appeared full and a few were in intermediate form. Empty particles were rarely seen. The purified rAAV-GFP stocks have been successfully used in in vitro and in vivo transfection experiments. Therefore, this new method offers a simple, rapid and cost-effective way for large-scale rAAV purification.

为了解西南环境移民示范区农业土地利用对土壤肥力变化的影响 ,以便更好地指导移民对土地进行合理的综合性开发 ,特对肯福环境移民示范区移民迁入前和移民迁入后第三年示范区的土壤养分的动态变化进行了研究 ,分析了耕层土样的 8个土壤养分指标 ,结果表明 ,示范区土壤肥力整体上呈下降趋势 ,但不同的养分元素含量变化又有差别 ,并且不同利用方式的土壤养分元素含量变化也存在差别 ,宜林类用地土壤肥力降低幅度最大。

Early ERP effects on the scaling of spatial attention in visual search

This article describes the examination of the spatial‘scaling＇ effect of visual attention with the technique of event-related potential （ERP）. Eighteen participants were involved in a visual search task in which the cue-target paradigm was used. The search array was three concentric circles consisting of randomly selected English letters that were equally distributed in each circle. The behavioral and ERP data were recorded, respectively. The behavioral results showed that the response time increased and the response accuracy decreased with the increase of precue size. The ERPs amplitude of PI and NI components evoked by search array increased with the reduction of precue size. However, the latencies of these ERP components did not show significant differences between conditions. The hierarchical data of both behavioral assessment and ERPs provided evidence for the spatial ＇scaling＇ effect of visual attention. The amplitudes of PI and N 1 components may be used as indices to examine the effect of spatial ＇scaling＇. In different tasks, the display-set size of stimuli and the task complexity may be important factors that affect the attention allocation.
