Structural basis of ultraviolet-B perception by UVR8

The Arabidopsis thaliana protein UVR8 is a photoreceptor for ultraviolet-B. Upon ultraviolet-B irradiation, UVR8 undergoes an immediate switch from homodimer to monomer, which triggers a signalling pathway for ultraviolet protection. The mechanism by which UVR8 senses ultraviolet-B remains largely unknown. Here we report the crystal structure of UVR8 at 1.8?? resolution, revealing a symmetric homodimer of seven-bladed β-propeller that is devoid of any external cofactor as the chromophore. Arginine residues that stabilize the homodimeric interface, principally Arg?286 and Arg?338, make elaborate intramolecular cation-π interactions with surrounding tryptophan amino acids. Two of these tryptophans, Trp?285 and Trp?233, collectively serve as the ultraviolet-B chromophore. Our structural and biochemical analyses identify the molecular mechanism for UVR8-mediated ultraviolet-B perception, in which ultraviolet-B radiation results in destabilization of the intramolecular cation-π interactions, causing disruption of the critical intermolecular hydrogen bonds mediated by Arg?286 and Arg?338 and subsequent dissociation of the UVR8 homodimer.     

B10和NaB10团簇的几何结构和电子性质

应用密度泛函理论（DFT）中的B3LYP方法在6-311+G（d）水平上计算并分析了B₁₀和NaB₉10)团簇的几何结构及电子性质,同时,讨论了团簇的平均结合能、能级间隙、自然电荷分布和极化率.研究表明:B₁₀团簇的基态为半平面构型,NaB₁₀团簇的基态是以B₁₀半平面为配位基,并与Na原子构成的配位络合物.能级间隙和平均结合能结果表明增加一个Na原子改变了B原子之间的成键作用.另外,对平均线性极化率和极化率的各向异性不变量研究表明:基态B₁₀团簇增加一个Na原子后NaB₁₀团簇的电子结构更趋于紧凑.     

温差法制备纳米气泡及温度的影响

通过一系列的实验建立了利用冷水（4±0.1℃）和热水（25～40℃）进行替换即温差法制备界面纳米气泡的方法.原子力显微镜（AFM）观察显示：温差法也能在疏水的高序热解石墨（HOPG）表面上产生稳定的纳米气泡.统计结果显示：热水温度在25～38℃范围内,产生纳米气泡的数量随着热水温度的升高而增多,当热水温度为38℃左右时达到最大值,之后随着热水温度的升高反而减少;每平方微米纳米气泡气体的总体积在37℃时达到最大值;温差法也能产生纳米气层和双层结构,其性质与前人发现的一致.     

钠离子依赖的中性氨基酸转运蛋白SNAT2-HA融合蛋白的构建及鉴定

钠离子依赖的中性氨基酸转运蛋白SNAT2在哺乳动物组织中广泛表达,具有转运中性氨基酸的功能,在谷氨酸-谷氨酰胺循环、肝脏糖质新生等生物通路中发挥重要作用．为了方便测定SNAT2在细胞膜上的表达和定位,本研究采用PCR扩增和酶切连接将HA标签蛋白与SNAT2的C末端连接,构建了真核生物表达载体pBK-CMVA（1098—1300）-SNAT2-HA表达载体．用脂质体转染法将该表达载体瞬时转染到HEK293T细胞中,通过Westernblot法检测到SNAT2-HA融合蛋白在细胞膜上的正确表达和定位．pBK-CMV△（1098-1300）-SNAT2-HA表达载体的成功构建,为今后对SNAT2的结构和功能的研究提供了有效方法．     

基于密度估计的异常电力负荷数据辨识与修正

SCADA系统数据库中一般会有一些异常的电力负荷数据,直接用其来进行短期负荷预测将影响预测结果的准确性,因此有必要对这些异常数据进行辨识和修正.文中同时考虑负荷的横向连续性和纵向连续性,先把负荷数据按照日期排列成二维数据集,然后采用基于密度的方法,在两个维度中对异常数据进行辨识与修正,最后通过实例分析验证了该方法的有效性.     

层次分析结合模糊数学法评价电镀企业清洁生产潜力

电镀行业属于高能耗高污染行业,必须推行清洁生产;评价企业清洁生产潜力非常重要且是基础。采用层次分析法与模糊数学法结合,建立企业清洁生产潜力评价模型,评价电镀企业清洁生产潜力,并通过案例实证。     

The hemocompatibility and the reabsorption function of TiO2 nanotubes biomembranes

A novel high-intensity TiO2 nanotubes array membrane was fabricated via electrochemical anodization of highly pure titanium foil and an open-ended TiO2 nanotubes array membrane was obtained by HF gas etching at the bottom of the nanotubes. Pig tubular epithelial cells (LLC-PK1) and vascular endothelial cells (ECV304) were cultivated on the open-ended TiO2 nanotubes surface through a mixing implantation method and TiO2 nanotubes biomembrane materials with physiological function were successfully produced. Hemocompatibility of glass slides, pure titanium, TiO2 nanotubes without cells and TiO2 nanotubes with culture cells were investigated by the plasma recalcification time method and reabsorption of sodium and potassium were measured by custom-designed devices. The results show that the hemocompatibility of the TiO2 nanotubes array membrane with culture cells was superior to the control group, and the biomembrane has an excellent reabsorption function. This demonstrates that a TiO2 nanotubes array membrane has excellent physiological function and is an ideal candidate material for biological dialysis.

以G85成渝高速公路（永川段）为例,探讨丘陵地区高速公路生态用地的帚观规划以及建设模式．在剖析公路景观中4个典型区域的规划模式及其植被配置方式的基础上,提出大尺度景观的设计理念：根据公路途经地域的不同,设计田园乡村绿化景观、山地丘陵绿化景观和村镇绿化景观,以展示沿途的田园风光、山地风景和村镇绿化景观;结合有色植物的合理配置,展示景观动态．     

基于因素间相互作用的多毒素食品质量安全风险水平评价模型的理论框架

为建立一完整的多毒素食品风险评价模型,本文以关系属性的数学表述和机体系统论模型为基础就建立食品风险评价模型进行了理论探索,从人、食品双方面的因素出发将食品风险评价4个阶段涉及到的变量整合到一个模型之中,形成了一个多毒素地域性食品风险水平评价模型框架,利用独立作用效果经验公式验证了模型的正确性;并考虑食品质量检验、主观因素对食品风险评定的影响完善了模型,引入了模型风险不确定度的评定.同时将联合毒性分为两类,统一了协同作用、增强作用、拮抗作用、独立作用.本文为建立具体评价模型奠定理论基础.
