Formation of anaphylatoxin in human serum

Zusammenfassung Es ist möglich, auch in menschlichem Serum eine Anaphylatoxinbildung (AT) durch Kontaktaktivierung oder Kobragift zu induzieren. Wegen der geringen Mengen, die entstehen, muss das wirksame Prinzip vor dem biologischen Nachweis angereichert werden. Menschliches AT verhält sich in allen untersuchten Eigenschaften wie AT aus anderen Plasmaarten. Es unterscheidet sich von dem darmkontrahierenden Spaltprodukt aus der menschlichen Komplementkomponente C3, das mithin nicht als AT angesprochen werden kann.     

Nck adaptor proteins link nephrin to the actin cytoskeleton of kidney podocytes

The glomerular filtration barrier in the kidney is formed in part by a specialized intercellular junction known as the slit diaphragm, which connects adjacent actin-based foot processes of kidney epithelial cells (podocytes). Mutations affecting a number of slit diaphragm proteins, including nephrin (encoded by NPHS1), lead to renal disease owing to disruption of the filtration barrier and rearrangement of the actin cytoskeleton, although the molecular basis for this is unclear. Here we show that nephrin selectively binds the Src homology 2 (SH2)/SH3 domain-containing Nck adaptor proteins, which in turn control the podocyte cytoskeleton in vivo. The cytoplasmic tail of nephrin has multiple YDxV sites that form preferred binding motifs for the Nck SH2 domain once phosphorylated by Src-family kinases. We show that this Nck-nephrin interaction is required for nephrin-dependent actin reorganization. Selective deletion of Nck from podocytes of transgenic mice results in defects in the formation of foot processes and in congenital nephrotic syndrome. Together, these findings identify a physiological signalling pathway in which nephrin is linked through phosphotyrosine-based interactions to Nck adaptors, and thus to the underlying actin cytoskeleton in podocytes. Simple and widely expressed SH2/SH3 adaptor proteins can therefore direct the formation of a specialized cellular morphology in vivo.     

复合型固体饮料加工工艺研究

以红枣、花生、枸杞、生姜为主要原料,麦芽糊精为助干剂,通过喷雾干燥技术开发出一种复合固体饮料.通过考察热水浸提、果胶酶浸提以及超声波辅助果胶酶浸提这三种前处理方式,以及喷雾干燥中的料液比、助干剂的添加量、出风温度这三个影响因素,探讨了复合型固体饮料的加工工艺.以出粉率、喷雾干燥效果和感官分值为评定标准,最终得到最佳前处理方式为超声波辅助果胶酶浸提,且其喷雾干燥的最佳条件为料液比1:6,固形物与助干剂的比例1:1,出风温度85℃.     

克拉霉素与阿奇霉素、多西环素治疗生殖道衣原体感染的疗效比较

目的:探讨分析比较克拉霉素、阿奇霉素、多西环素治疗生殖道沙眼衣原体感染的临床疗效、病原体清除情况及其安全性.方法:选择本院皮肤科门诊的150例生殖道沙眼衣原体感染患者,随机分为A、B、C三组,每组各50例,A组口服克拉霉素,B组口服阿奇霉素,C组口服多西环素,连续治疗2周为一个疗程.疗程结束后两周进行疗效评价,记录临床症状、体征及不良反应,并做尿道(宫颈)分泌物沙眼衣原体检测.结果:疗程结束两周后,克拉霉素组疗效总有效率为93.3%,明显高于阿奇霉素组的77.3%和多西环素组的71.1%,差异均有统计学意义(P0.05);阿奇霉素组的有效率虽略高于多西环素组,但无统计学差异.对病原体的清除率分别为93.3%、77.3%、71.1%,统计学分析与上述临床疗效的有效率结果一致.三组不良反应的发生率均较低,且不良反应均较轻微.结论:克拉霉素具有临床疗效好、病原体清除率高、不良反应少等特点,是临床治疗生殖道沙眼衣原体感染的较好药物之一.     

Light-avoidance-mediating photoreceptors tile the Drosophila larval body wall

Photoreceptors for visual perception, phototaxis or light avoidance are typically clustered in eyes or related structures such as the Bolwig organ of Drosophila larvae. Unexpectedly, we found that the class IV dendritic arborization neurons of Drosophila melanogaster larvae respond to ultraviolet, violet and blue light, and are major mediators of light avoidance, particularly at high intensities. These class IV dendritic arborization neurons, which are present in every body segment, have dendrites tiling the larval body wall nearly completely without redundancy. Dendritic illumination activates class IV dendritic arborization neurons. These novel photoreceptors use phototransduction machinery distinct from other photoreceptors in Drosophila and enable larvae to sense light exposure over their entire bodies and move out of danger.     

强震下砌体结构倒塌过程预测及其合理性分析

为了使用计算机仿真技术对一些因条件限制而不能完成的工作进行试验模拟、灾害预测、结构性能评估等,利用显示动力有限元软件ANSYS/LS-DYNA模拟某2层带构造柱的砌体结构振动台试验模型倒塌的过程,同时,通过模态和动力时程分析对结构的周期、振型、加速度、应力应变及位移等进行了分析,并和振动台试验结果进行了对比。结果显示,采用合适的材料模型和失效准则,ANSYS/LS-DYNA能很好地模拟倒塌过程,动力时程分析的结果和试验吻合,验证了上述倒塌过程模拟的合理性,体现了数值振动台的作用。

Biochemical,biophysical and molecular dynamics studies on the proteoglycan-like domain of carbonic anhydrase IX

Human carbonic anhydrase IX (hCA IX) is a tumour-associated enzyme present in a limited number of normal tissues, but overexpressed in several malignant human tumours. It is a transmembrane protein, where the extracellular region consists of a greatly investigated catalytic CA domain and a much less investigated proteoglycan-like (PG) domain. Considering its important role in tumour biology, here, we report for the first time the full characterization of the PG domain, providing insights into its structural and functional features. In particular, this domain has been produced at high yields in bacterial cells and characterized by means of biochemical, biophysical and molecular dynamics studies. Results show that it belongs to the family of intrinsically disordered proteins, being globally unfolded with only some local residual polyproline II secondary structure. The observed conformational flexibility may have several important roles in tumour progression, facilitating interactions of hCA IX with partner proteins assisting tumour spreading and progression.
