Ecdysome 20-hydroxylase from the midgut of the tobacco hornworm (Manduca sexta L.).

An ecdysone 20-hydroxylase enzyme system that converts alpha-ecdysone to 20-hydroxyecdysone was prepared from the midgut of the tobacco hornworm prepupa. This partially purified enzyme is NADPH dependent and is localized in the mitochondrial fraction of the midgut tissue.     

Degradation of encephalitogenic protein in aerobic ascorbic acid solutions.

Aerobic ascorbic acid solutions are capable of extensively cleaving the peptide chain of the myelin basic protein. Cleavage occurred most readily with C-terminal to aspartic acid, serine, threonine, glutamic acid and leucine residues.     

Bowel serotonin levels in the fasted and non-fasted Sprague-Dawley rat

Zusammenfassung Der Serotoningehalt der Darmmucosa aus 14 Schnittpräparaten von Sprague-Dawley Rattenweibchen nach 16-stündigem Fasten und nach normaler Fütterung wurde bestimmt. Grössere Serotoningehalte wurden im oberen Teil des Verdauungstraktus (ausschliesslich den Magenblindsack) der Hungertiere gefunden.     

Hybrid sterility in cattle ticks (Acari: Ixodidae)

Summary Male offspring resulting from interspecific crosses between the cattle ticks,Boophilus annulatus andB. microplus, are sterile. Hybrid females were found to produce sterile sons through 3 backcross generations.

Zusammenfassung An männlichen Ratten wurden nach 2 h Pylorusligatur Magensaftmenge, Volumen/100 g Körpergewicht, pH, Azidität, gesamte Azidität und Säureproduktion bestimmt und als Funktion von ICI-50123 studiert. Eine maximale Stimulation erfolgte bei einer Dosierung von 1000.0 g/kg s.c. Eine Abnahme der Säureproduktion zeigte sich bei supramaximaler Dosis.     

Temperature and acidity for maximal fluorescence of serotonin and serotonin-O.P.T.

Résumé Dans la détermination fluorimétrique de la sérotonine elle-mÊme et du complexe sérotonine-O.P.T. (o-phthaldialdéhyde) un maximum de fluorescence a été atteint pour des normalités de HCl de 3.25–3.50 et 6.25–6.50, respectivement. Pour les deux procédés, on a noté des rapports en sens inverse entre la fluorescence produite et la température de l'échantillon, pour des températures comprises entre 5 à 35 C. Entre la température ambiante (de 20 C) et 5 C, la fluorescence s'est accrue de 30% environ.     

Genome-wide association study identifies a common variant associated with risk of endometrial cancer

Endometrial cancer is the most common malignancy of the female genital tract in developed countries. To identify genetic variants associated with endometrial cancer risk, we performed a genome-wide association study involving 1,265 individuals with endometrial cancer (cases) from Australia and the UK and 5,190 controls from the Wellcome Trust Case Control Consortium. We compared genotype frequencies in cases and controls for 519,655 SNPs. Forty seven SNPs that showed evidence of association with endometrial cancer in stage 1 were genotyped in 3,957 additional cases and 6,886 controls. We identified an endometrial cancer susceptibility locus close to HNF1B at 17q12 (rs4430796, P = 7.1 × 10(-10)) that is also associated with risk of prostate cancer and is inversely associated with risk of type 2 diabetes.     

PALB2, which encodes a BRCA2-interacting protein, is a breast cancer susceptibility gene

PALB2 interacts with BRCA2, and biallelic mutations in PALB2 (also known as FANCN), similar to biallelic BRCA2 mutations, cause Fanconi anemia. We identified monoallelic truncating PALB2 mutations in 10/923 individuals with familial breast cancer compared with 0/1,084 controls (P = 0.0004) and show that such mutations confer a 2.3-fold higher risk of breast cancer (95% confidence interval (c.i.) = 1.4-3.9, P = 0.0025). The results show that PALB2 is a breast cancer susceptibility gene and further demonstrate the close relationship of the Fanconi anemia-DNA repair pathway and breast cancer predisposition.     

The chronic effects of nicotine monomethidide on gastric secretion in pylorus-ligated rats

Résumé L'action de la nicotine monométhyliodine (NMI) sur la sécrétion gastrique basique et sur les concentrations de choline de cervelles, d'acétylcholine et de nicotine et sur l'activité de la transacétylcholinestérase a été étudiée chez des rats. Le NMI, sel quaternaire de nicotine, qui ne traverse pas la barrière hématoencéphalique, a été administré dans de la gélatine à 6% en dosage sous-cutané de 1000 g/ml/kg/jour pendant 14 jours. Le NMI n'a pas changé de paramètres et aucune trace de nicotine n'a été découverte dans les homogénates de cervelles complètes.
