Phosphorylation/dephosphorylation is probably the most common and important reversible post-translational modification of proteins. Analyzing the functional effects of phosphorylation is helpful for understanding the biological functions of proteins. Identification of the phosphorylation sites of phosphorylated protein is a prerequisite for research on phosphorylation. In this work, an effective and simple method of identification of protein phosphorylation sites has been developed. Phosphopeptides were selectively enriched with immobilized metal affinity chromatography (IMAC) and subsequently chemically modified by 4-sulfophenyl isothiocyanate, and then the chemically modified phosphopeptides were sequenced with post-source decay (PSD) matrix-assisted laser desorption/ionization (MALDI) time-of-flight mass spectrometry for detecting phosphorylation sites. The charge of derivatization by 4-sulfophenyl isothiocyanate introduces a negative sulfonic acid group at the N-terminus of a peptide, and enables the selective detection of only a single series of C-terminal y-type ions. This chemically assisted method greatly simplifies the extremely complex pattern of PSD fragment ions and makes the PSD spectra more easier to be interpreted. The phosphorylation sites of a synthesized model phosphopeptide and human c-myc protein have been successfully identified by this method. 相似文献

There is much debate on biological affinities of the phosphatized globular fossils from the Neoproterozoic Doushantuo phosphorites at Weng’an, Guizhou Province. Here we report for the first time the budding structures of some of these globular fossils, which have previously been interpreted as the resting eggs and early cleavage embryos of metazoans. The budding structures are similar to the germinating tubes of the spore and zygote of living algae, suggesting that some globular fossils from the Doushantuo phosphorites bear affinity with algae. The present new information indicated various biological affinity of the phosphatized globular fossils from the Doushantuo phosphorites. 相似文献

The low signal to noise ratio (SNR) of functional MRI (fMRI) prefers more sensitive data analysis methods. Based on stationary wavelet transform and spectrum analysis, a new method with high detective sensitivity was developed for analyzing fMRI time series, which does not require any prior assumption of the characteristics of noises. In the proposed method, every component of fMRI time series in the different time-frequency scales of stationary wavelet transform was discerned by the spectrum analysis, then the components from noises were removed using the stationary wavelet transform, finally the components of real brain activation were detected by cross-correlation analysis. The results obtained from both simulated and in vivo visual experiments illustrated that the proposed method has much higher sensitivity than the traditional cross-correlation method. 相似文献

The object of this paper is to evaluate the removal of disinfection by-products formation potential by artificially intensified biological activated carbon(BAC) process which is developed on the basis of traditional ozone granular activated carbon (GAC). The results show that 23.1% of trihalomethane formation potential (THMFP) and 68% of haloacetic acid formation potential (HAAFP) can be removed by BAC, respectively. Under the same conditions, the removal rates of the same substances were 12.2% and 13-25% respectively only by GAC process. Compared with GAC, the high removal rates of the two formed potential substances were due to the increasing of bioactivity of the media and the synergistic capabilities of biological degradation cooperating with activated carbon adsorption of organic compounds. BAC process has some advantages such as long backwashing cycle time, low backwashing intensity and prolonged activated carbon lifetime, etc. 相似文献
