重组人脑钠素在大肠杆菌中的表达、纯化与鉴定

人脑钠素(hBNP,humanbrainnatriureticpeptide)是心室分泌的由32个氨基酸组成的多肽激素.临床研究表明,hBNP是治疗充血型心衰竭的一种安全有效的多肽药物.化学合成脑钠素全基因,以pET32a为表达载体,构建了硫氧还蛋白-脑钠素(thioredoxin＿hBNP)融合蛋白表达质粒,并转化至大肠杆菌BL21(DE3),经IPTG诱导,融合蛋白的表达量占全菌蛋白量的25%以上.融合蛋白经肠激酶裂解、Zn＿Sepharose亲和层析和C4反相高效液相层析,从每升培养液中可获取4mgrhBNP,纯度达到95%.经质谱测定,rhBNP样品的分子量为3464Da.体外活性测定结果表明,rhBNP样品对兔胸主动脉条具有显著的血管舒张效应,其ED50为(2.24±0 58)×10-6mg/mL.     

重组人胸腺肽α1在大肠杆菌中的表达纯化和鉴定

用PCR将化学合成的人胸腺肽α1基因扩增并克隆到pMD18-T载体中,经过KpnI／SacI酶切、连接将胸腺肽基因插入到表达载体pET32b中硫氧还蛋白下游．将重组质粒转化至表达宿主BL21(DE3)中,经IPTG诱导,Zn—Sepharose亲和层析纯化以及复性处理,从每升菌液中可获得35mg硫氧还蛋白-胸腺肽α1融合蛋白．经凝血因子Xa酶切、Zn—Sepharose亲和层析以及反相高效液相色谱纯化获得3．8mg重组人胸腺肽．小鼠胸腺细胞凋亡实验表明,重组人胸腺肽能显地抑制地赛米松诱导的小鼠胸腺细胞的凋亡,且该抑制作用与重组人胸腺肽的浓度呈剂量依赖关系．     

Concentrations of “elemental carbon” in samples from the Peking Man Site at Zhoukoudian and the possibility of their application in the development of evidence for the use of fire by humans

Elemental carbon?EC) is a C-rich, O-H-S-N-depleted substance that is necessarily produced in the process of combustion. Due to the logn-term use of fire by cave-inhabiting ape-men, considerable amounts of 揺lemen-tal carbon?would be necessarily left behind in the corre-sponding strata inside the cave. The sepaation of EC was effected by the chemical method and the contents of carbon were determined on an element analyser and a self-made measuring system. The concentration of Ec in the hearth,the exhibited ash samples collected from near the hearth un-earthed in the 1930s, and the three samples of accumulated material collected from the tenth bed are 43.74%, 1.77%,3.88%,1.87% and 1.11%,respectively; those of the equivalent samples from the tenth bed outside the cave and samples from the fourth and seventh beds are at least one order of magnitude lower than the former抯. The above re-sults show that the sampling location of the tenth bed may be close to the hearth. Sampling over a wider range may lead to the finding of a location where the concentrations of EC are corresponding to the maximum values of EC in the hearth. The determination and study of EC may provide new evi-dence for the use of fire by Peking Man.     

<Emphasis Type="Italic">In situ</Emphasis> synthesis of TiC reinforced Cu<Subscript>47</Subscript>Ti<Subscript>34</Subscript>Zr<Subscript>11</Subscript>Ni<Subscript>8</Subscript> bulk metallic glass composites

In situ synthesized TiC particles and β-Ti dendrites reinforced Cu47Ti34Zr11Ni8 bulk metallic glass (BMG) composite ingots were prepared by the suction casting method.The ingots with diameters from 1 up to 4mm were successfully obtained. It was shown that introducing TiC micro-sized particles into the amorphous matrix did not disturb the glass forming ability (GFA) of the matrix,while the yield strength and ductility could be well improved.The phase constitution, microstructure and elements distribution in the composites were studied by OM, XRD, SEM and EDS.It was shown that the in situ synthesized TiC particles acting as heterogeneous nucleation sites promoted the precipitation of β-Ti dendrites, resulting in the formation of the TiC particles and β-Ti dendrites co-reinforced BMG composites. The compressive tests were employed to probe the yield strength and ductility of BMG composites.     

Expression of lysine-rich protein gene and analysis of lysine content in transgenic wheat

Expression vector pBPC102, which carries winged bean lysine-rich protein (wblrp) gene and dihydropicolinate synthase (DHDPS) gene, was transferred into hexaploid winter wheat cv. Jinghua No.l, Jing411, You899 and Yangnongl5 explants of immature inflorescence and immature embryos by particle bombardment. More than 100 transgenic plants were obtained under the selection of s-(2-aminoethyl)-L-cysteine (AEC). Confirmed transgenic plants of To and TI generation by PCR and PCR-Southern blotting analyses showed successful integration of wblrp gene into wheat genome. Analysis of transgenic plant lines of T2 by Northern dot-blotting showed good expression of wblrp gene in offspring seed. The content of free lysine in leaves, contents of bound lysine and total proteins in seeds of T2 transgenie wheat lines were determined and analyzed. Among 34 tested transgenic lines, levels of free lysine content in leaves of 9 transgenic lines are 2~3times higher than un-trans-formed wild-type cultivars. Among 17 analyzed transgenic lines, bound lysine content of 4 transgenic lines is more than 10% higher than that of wild-type cultivars. Our research suggests that introducing wblrp gene into wheat is an effective way to improve its nutrition quality.

Correlation between geometrical parameters of electrical tree and corresponding partial discharge parameters in XLPE

This paper deals with the experimental study on the correlation between the geometrical parameters of electrical tree and corresponding partial discharge (PD) characteristic parameters in the course of electrical tree aging within cross linked polyethylene (XLPE) insulation. The electrical tree aging tests were performed on specimens removed from a section of 220 kV transmission cable. The PD macroscopic characteristic parameters were found to be significantly dependent on the corresponding geometrical parameters of electrical tree channels in the course of aging of XLPE, and different kind of electrical tree has different characteristics, and there is obvious correlation between the type of electrical tree and the pre-applied power-frequency stress. Beside, using regression analysis, the expression of the relation between them were obtained, and from which it can be seen that there is significant nonlinear correlation between geometrical parameters of electrical tree and corresponding PD characteristic parameters in the course of aging of XLPE. Therefore, the aging degree of XLPE can be effectively evaluated by recognizing the changing regularity of the PD rnacroscopic characteristic parameters.     

Synthesis and crystal structure of tetranuclear zinc benzoate

A tetranuclear zinc benzoate Zr4O(C6H5CO2)6 was synthesized and characterized by X-ray single crystal determination. It crystallizes in cubic, space group Ia-3d. Its crystal cell is very large, α=4.10063(18)nm, V=68.953(5)nm^3 and Z=48. The structure is composed of discrete Zr4O(C6H5CO2)6 molecules. In each molecule, four zinc atoms are held together by a central oxygen atom, which results in the formation of a regular tetrahedron. All benzoate ligands coordinate to zinc atoms in a bidentate bridging mode. Each zinc atom is in a slightly distorted tetrahedral geometry, coordinated by three benzoate oxygen atoms and the central oxygen atom. The intermolecular interactions result in the formation of a three-dimensional supramolecular framework, with non-intersecting parallel channels.     

Constructing three-dimension space graph for outlier detection algorithms in data mining

Outlier detection has very important applied value in data mining literature. Different outlier detection algorithms based on distinct theories have different definitions and mining processes. The three-dimensional space graph for constructing applied algorithms and an improved GridOf algorithm were proposed in terms of analyzing the existing outlier detection algorithms from criterion and theory.
