Environmental biosafety and transgenic potato in a centre of diversity for this crop

The Nuffield Council on Bioethics suggests that introgression of genetic material into related species in centres of crop biodiversity is an insufficient justification to bar the use of genetically modified crops in the developing world. They consider that a precautionary approach to forgo the possible benefits invokes the fallacy of thinking that doing nothing is itself without risk to the poor. Here we report findings relevant to this and other aspects of environmental biosafety for genetically modified potato in its main centre of biodiversity, the central Andes. We studied genetically modified potato clones that provide resistance to nematodes, principal pests of Andean potato crops. We show that there is no harm to many non-target organisms, but gene flow occurs to wild relatives growing near potato crops. If stable introgression were to result, the fitness of these wild species could be altered. We therefore transformed the male sterile cultivar Revolucion to provide a genetically modified nematode-resistant potato to evaluate the benefits that this provides until the possibility of stable introgression to wild relatives is determined. Thus, scientific progress is possible without compromise to the precautionary principle.     

Extreme deuterium enrichment in stratospheric hydrogen and the global atmospheric budget of H2

Molecular hydrogen (H2) is the second most abundant trace gas in the atmosphere after methane (CH4). In the troposphere, the D/H ratio of H2 is enriched by 120 per thousand relative to the world's oceans. This cannot be explained by the sources of H2 for which the D/H ratio has been measured to date (for example, fossil fuels and biomass burning). But the isotopic composition of H2 from its single largest source--the photochemical oxidation of methane--has yet to be determined. Here we show that the D/H ratio of stratospheric H2 develops enrichments greater than 440 per thousand, the most extreme D/H enrichment observed in a terrestrial material. We estimate the D/H ratio of H2 produced from CH4 in the stratosphere, where production is isolated from the influences of non-photochemical sources and sinks, showing that the chain of reactions producing H2 from CH4 concentrates D in the product H2. This enrichment, which we estimate is similar on a global average in the troposphere, contributes substantially to the D/H ratio of tropospheric H2.

Recommendations of the 2006 Human Variome Project meeting

Lists of variations in genomic DNA and their effects have been kept for some time and have been used in diagnostics and research. Although these lists have been carefully gathered and curated, there has been little standardization and coordination, complicating their use. Given the myriad possible variations in the estimated 24,000 genes in the human genome, it would be useful to have standard criteria for databases of variation. Incomplete collection and ascertainment of variants demonstrates a need for a universally accessible system. These and other problems led to the World Heath Organization-cosponsored meeting on June 20-23, 2006 in Melbourne, Australia, which launched the Human Variome Project. This meeting addressed all areas of human genetics relevant to collection of information on variation and its effects. Members of each of eight sessions (the clinic and phenotype, the diagnostic laboratory, the research laboratory, curation and collection, informatics, relevance to the emerging world, integration and federation and funding and sustainability) developed a number of recommendations that were then organized into a total of 96 recommendations to act as a foundation for future work worldwide. Here we summarize the background of the project, the meeting and its recommendations.     

Soft Systems Methodology: Other Voices

This issue of Systemic Practice and Action Research, celebrating the work of Peter Checkland, in the particular nature and development of soft systems methodology (SSM), would not have happened unless the work was seen by others as being important. No significant contribution to thinking happens without a secondary literature developing. Not surprisingly, many commentaries have accompanied the ongoing development of SSM. Some of these are insightful, some full of errors, and some include both insight and absurdity. Checkland (1999, p. A42) opines, in the recently published 30-year retrospective, that "SSM has been ill-served by its commentators." Scrutiny of the secondary literature on SSM provides support for this view and also identifies some general characteristics and trends that are important to the development of SSM and, incidentally, reinforces some existing conclusions.     

Regulation of PDGF signalling and vascular remodelling by peroxiredoxin II

Platelet-derived growth factor (PDGF) is a potent mitogenic and migratory factor that regulates the tyrosine phosphorylation of a variety of signalling proteins via intracellular production of H2O2 (refs 1, 2-3). Mammalian 2-Cys peroxiredoxin type II (Prx II; gene symbol Prdx2) is a cellular peroxidase that eliminates endogenous H2O2 produced in response to growth factors such as PDGF and epidermal growth factor; however, its involvement in growth factor signalling is largely unknown. Here we show that Prx II is a negative regulator of PDGF signalling. Prx II deficiency results in increased production of H2O2, enhanced activation of PDGF receptor (PDGFR) and phospholipase Cgamma1, and subsequently increased cell proliferation and migration in response to PDGF. These responses are suppressed by expression of wild-type Prx II, but not an inactive mutant. Notably, Prx II is recruited to PDGFR upon PDGF stimulation, and suppresses protein tyrosine phosphatase inactivation. Prx II also leads to the suppression of PDGFR activation in primary culture and a murine restenosis model, including PDGF-dependent neointimal thickening of vascular smooth muscle cells. These results demonstrate a localized role for endogenous H2O2 in PDGF signalling, and indicate a biological function of Prx II in cardiovascular disease.

Insights into Operationalizing Communities of Practice from SSM-Based Inquiry Processes

Two systemic inquiries, based on soft systems methodology (SSM), into the potential for using community of practice (CoP) theory by an Australian-government created research and development corporation to change its knowledge management (KM) strategy, are reported. Key staff were engaged in the inquiry into how to build a third-order KM strategy based on CoPs; an exploration of key published work on CoPs yielded four SSM activity models—‘being a community practice system’; ‘doing the work of imagination sub-system’; ‘doing the work of alignment sub-system’ and ‘doing the work of engagement sub-system’. These models can be used as heuristics to aid the purposeful design of CoPs in other settings. SSM, enacted as a systemic inquiry, can be understood as a form of systemic action research, which was well equipped to deal with CoP theory and, when enacted participatively, can generate important systemic insights. The inquiry began the process of fostering an appreciation of third order KM but, on the evidence available, did not lead to on-going commitment to a CoPs-based KM strategy. Future research should acknowledge how the framing of research situations influences the research process, the importance of the design of practice change settings and the limited evidence for purposeful interventions leading to successful CoPs.
