A number of studies have suggested the existence of multiple benzodiazepine binding sites in the brain. We have recently reported the physical separation of two apparent benzodiazepine binding site subtypes, the pharmacological properties, and distribution in tissue sections of which correspond to the putative type I and type II sites. Benzodiazepine and gamma-aminobutyric acid (GABA) receptors have been shown to interact, and lesions of the GABAergic striatonigral pathway, which lead to GABA supersensitivity, both increase the numbers of GABA binding sites and enhance GABA-stimulated benzodiazepine binding. We demonstrate here that degeneration of striatonigral fibres increases the density of putative type I benzodiazepine binding sites in the substantia nigra and decreases the density of the putative type II sites. This suggests that type I sites that increase after denervation are postsynaptic, whereas the type II sites reduced by the lesion may be localized to axons or terminals of the striatonigral pathways.  相似文献

A pair of pentapeptides, Met- and Leu-enkephalin were recently isolated from brain tissue. The two peptides seem to represent endogenous opiate receptor ligands and have by immunocytochemical and radioimmunoassay studies been shown to occur in an extensive system of cerebral and peripheral nerves. The relative proportions between Met- and Leu-enkephalin varies between different brain regions and also between different species, suggesting the existence of separate populations of Met- and Leu-enkephalin nerves. Until now, however, immunocytochemistry has given no support for this notion. We report here evidence of separate populations of Met- and Leu-enkephalin nerves.  相似文献

Zusammenfassung Der Cholinoxidase-Spiegel in Rattenleberbrei wurde nach intravenöser Verabreichung von Cerium gemessen. Die experimentell erhaltenen Werte zeigen, dass die vom Cerium verursachte Fetteinlagerung weder von der Art der Ratten noch von ihrem hormonalen Zustande abhängt. In Fettlebern scheint die Enzymhemmung mehr von ihrem Lipoidgehalt als von ihrer Ceriumkonzentration bestimmt zu werden.

Summary This study was done to delineate the role of - and -adrenergic receptors and cyclic AMP in the mechanism of ethanol effects on insulin release from isolated islets. Rats were given an -adrenergic blocker, phentolamine, or a -adrenergic blocker, propranolol. In addition, ethanol 1 g/kg was given intragastrically 1 h prior to sacrifice. Glucose mediated insulin release from isolated islets was enhanced by phentolamine and decreased by propranolol. Ethanol treatment inhibited glucose-induced insulin release from isolated islets of control rats as well as those given phentolamine and/or propranolol. Insulin release from isolated islets in response to dibutyryl-cyclic AMP was attenuated by ethanol. Theophylline enhanced glucose mediated insulin release from control islets but ethanol treatment produced a significant inhibition of insulin response. The data suggest that the site of action of the deleterious effects of ethanol on insulin release from isolated islets in rat does not involve adrenergic system and cyclic AMP.Supported by the U.S. Veterans Administration  相似文献

Glucocorticoids are released in response to stressful experiences and serve many beneficial homeostatic functions. However, dysregulation of glucocorticoids is associated with cognitive impairments and depressive illness. In the hippocampus, a brain region densely populated with receptors for stress hormones, stress and glucocorticoids strongly inhibit adult neurogenesis. Decreased neurogenesis has been implicated in the pathogenesis of anxiety and depression, but direct evidence for this role is lacking. Here we show that adult-born hippocampal neurons are required for normal expression of the endocrine and behavioural components of the stress response. Using either transgenic or radiation methods to inhibit adult neurogenesis specifically, we find that glucocorticoid levels are slower to recover after moderate stress and are less suppressed by dexamethasone in neurogenesis-deficient mice than intact mice, consistent with a role for the hippocampus in regulation of the hypothalamic-pituitary-adrenal (HPA) axis. Relative to controls, neurogenesis-deficient mice also showed increased food avoidance in a novel environment after acute stress, increased behavioural despair in the forced swim test, and decreased sucrose preference, a measure of anhedonia. These findings identify a small subset of neurons within the dentate gyrus that are critical for hippocampal negative control of the HPA axis and support a direct role for adult neurogenesis in depressive illness.  相似文献

Changes in vegetation including area occupied, canopy cover, and maturity class of cottonwoods ( Populus spp.) within lower-elevation zones of the Colorado River and Rio Grande in Colorado were monitored over 25- and 27-year intervals, respectively, using photo-interpretative methods. Estimated loss of cottonwoods along the Colorado River was 2ha/km (-17.5%), and remaining stands had become more open and older. Cottonwoods along the Rio Grande increased 1.6 ha/km (9.3%) with minor canopy cover and maturity class changes. Area occupied by shrubs and river channel changed little along the Colorado River, but declined along the Rio Grande. Loss of hay meadow occurred along both rivers, whereas developed land increased along the Colorado River and farmland increased along the Rio Grande. Wildlife habitats along the Colorado deteriorated much more rapidly than those along the Rio Grande during monitored intervals.  相似文献
