As end-stage renal disease (ESRD) has a four times higher incidence in African Americans compared to European Americans, we hypothesized that susceptibility alleles for ESRD have a higher frequency in the West African than the European gene pool. We carried out a genome-wide admixture scan in 1,372 ESRD cases and 806 controls and found a highly significant association between excess African ancestry and nondiabetic ESRD (lod score = 5.70) but not diabetic ESRD (lod = 0.47) on chromosome 22q12. Each copy of the European ancestral allele conferred a relative risk of 0.50 (95% CI = 0.39-0.63) compared to African ancestry. Multiple common SNPs (allele frequencies ranging from 0.2 to 0.6) in the gene encoding nonmuscle myosin heavy chain type II isoform A (MYH9) were associated with two to four times greater risk of nondiabetic ESRD and accounted for a large proportion of the excess risk of ESRD observed in African compared to European Americans. 相似文献

Atopic disease, including atopic dermatitis (eczema), allergy and asthma, has increased in frequency in recent decades and now affects approximately 20% of the population in the developed world. Twin and family studies have shown that predisposition to atopic disease is highly heritable. Although most genetic studies have focused on immunological mechanisms, a primary epithelial barrier defect has been anticipated. Filaggrin is a key protein that facilitates terminal differentiation of the epidermis and formation of the skin barrier. Here we show that two independent loss-of-function genetic variants (R510X and 2282del4) in the gene encoding filaggrin (FLG) are very strong predisposing factors for atopic dermatitis. These variants are carried by approximately 9% of people of European origin. These variants also show highly significant association with asthma occurring in the context of atopic dermatitis. Since the creation of Dolly via somatic cell nuclear transfer (SCNT), more than a dozen species of mammals have been cloned using this technology. One hypothesis for the limited success of cloning via SCNT (1%-5%) is that the clones are likely to be derived from adult stem cells. Support for this hypothesis comes from the findings that the reproductive cloning efficiency for embryonic stem cells is five to ten times higher than that for somatic cells as donors and that cloned pups cannot be produced directly from cloned embryos derived from differentiated B and T cells or neuronal cells. The question remains as to whether SCNT-derived animal clones can be derived from truly differentiated somatic cells. We tested this hypothesis with mouse hematopoietic cells at different differentiation stages: hematopoietic stem cells, progenitor cells and granulocytes. We found that cloning efficiency increases over the differentiation hierarchy, and terminally differentiated postmitotic granulocytes yield cloned pups with the greatest cloning efficiency. 相似文献

Sympatric populations of Cophosaurus texanus and Callisaurus draconoides were periodically sampled from March 1973 through April 1974 at Burro Creek, Mohave County, Arizona. Callisaurus were also sampled at Rock Valley, Nye County, Nevada. Sex ratios were skewed in favor of males in the adult Cophosaurus but were equal in both adult populations of Callisaurus. Both species became sexually mature as yearlings. Mean clutch sizes were 3.55 (±0.83) for Cophosaurus, and 4.25 (±1.08) and 5.07 (±1.33) for Callisaurus at Burro Creek and Rock Valley respectively. Evidence of multiple clutches was exhibited by both species. Egg weight/body weight ratios for both species and clutch weight/body weight ratios for Cophosaurus were notably smaller than previously reported. At Burro Creek both species were highly insectivorus, with orthopterans comprising the largest food group of each. Niche overlap for food was high at the ordinal level, but at the familial level it is apparent that Callisaurus probably fed in the more xeric areas of the riparian habitat. No differences were found in the temperature responses of these two lizards. However, minor temporal separations and substantial spatial partitioning were observed. Clethrionomys gapperi were captured in microhabitats with greater densities of overall cover than at noncapture or random sites within the study area. Variables describing cover density and distance from free water were selected in a discriminant function analysis to differentiate between vole capture and noncapture sites. Vole capture sites had greater amounts of cover within 4 dm above ground surface and were further from standing water than noncapture sites. The preferential use by C. gapperi of microhabitats with greater densities of cover is in agreement with laboratory and field assessments of habitat use reported in the literature. 相似文献

Successful infection of white pine species by white pine blister rust (WPBR) is contingent upon environmental conditions that are favorable to the spread and development of Cronartium ribicola . Site and stand factors related to this process have been studied elsewhere within the distribution of the disease, but few studies have concentrated on the high-elevation white pine forests of southern Idaho and western Wyoming. We found that mean summer precipitation, average tree diameter, and elevation were the most important variables in 3 logistic regression models of WPBR presence and intensity. The models were tested on a randomly chosen portion of our data set. The model with 9 variables correctly predicted categories of low-, moderate-, and high-disease incidence in 79% of cases. The 2 models with fewer variables had lower predictive efficiencies but were more parsimonious and generally easy to measure. The ability to use easily measured or remotely sensed site and stand characteristics to predict WPBR spread or intensification could be an important asset to land managers who need to decide where to focus disease mitigation efforts and predict disease effects on water quality, wildlife habitat, recreation potential, and other land-management activities. 相似文献

We examined the effect of elevation on litter-size variation in viviparous lizards of the Sceloporus grammicus complex in 10 states of Mexico. Female snout–vent length (SVL) decreased with increasing elevation, and absolute litter size based on embryos also tended to de crease with increasing elevation. However, after controlling for variation in female body size, we found that litter sizes tended to be relatively larger at higher elevation. Elevation therefore appears to influence litter size in these lizards; however, relatively little of the variation is explained by elevation; thus, other factors are likely making substantial contributions to the observed litter-size variation. The S. grammicus complex appears to be a good model system for examining the underlying causes of geographic and elevational variation in lizard life histories. Non-indigenous African honey bees have invaded most of South and Central America in just over 30 years. The genetic composition of this population and the means by which it rapidly colonizes new territory remain controversial. In particular, it has been unclear whether this 'Africanized' population has resulted from interbreeding between African and domestic European bees, or is an essentially pure African population. Also, it has not been known whether this population expanded primarily by female or by male migration. Restriction site mapping of 62 mitochondrial DNAs of African bees from Brazil, Venezuela and Mexico reveals that 97% were of African (Apis mellifera scutellata) type. Although neotropical European apiary populations are rapidly Africanized by mating with neotropical African males, there is little reciprocal gene flow to the neotropical African population through European females. These are the first genetic data to indicate that the neotropical African population could be expanding its range by female migration. 相似文献
