We carried out a multistage genome-wide association study of type 2 diabetes mellitus in Japanese individuals, with a total of 1,612 cases and 1,424 controls and 100,000 SNPs. The most significant association was obtained with SNPs in KCNQ1, and dense mapping within the gene revealed that rs2237892 in intron 15 showed the lowest Pvalue (6.7 x 10(-13), odds ratio (OR) = 1.49). The association of KCNQ1 with type 2 diabetes was replicated in populations of Korean, Chinese and European ancestry as well as in two independent Japanese populations, and meta-analysis with a total of 19,930 individuals (9,569 cases and 10,361 controls) yielded a P value of 1.7 x 10(-42) (OR = 1.40; 95% CI = 1.34-1.47) for rs2237892. Among control subjects, the risk allele of this polymorphism was associated with impairment of insulin secretion according to the homeostasis model assessment of beta-cell function or the corrected insulin response. Our data thus implicate KCNQ1 as a diabetes susceptibility gene in groups of different ancestries.  相似文献

Platelets are the second most abundant cell type in blood and are essential for maintaining haemostasis. Their count and volume are tightly controlled within narrow physiological ranges, but there is only limited understanding of the molecular processes controlling both traits. Here we carried out a high-powered meta-analysis of genome-wide association studies (GWAS) in up to 66,867 individuals of European ancestry, followed by extensive biological and functional assessment. We identified 68 genomic loci reliably associated with platelet count and volume mapping to established and putative novel regulators of megakaryopoiesis and platelet formation. These genes show megakaryocyte-specific gene expression patterns and extensive network connectivity. Using gene silencing in Danio rerio and Drosophila melanogaster, we identified 11 of the genes as novel regulators of blood cell formation. Taken together, our findings advance understanding of novel gene functions controlling fate-determining events during megakaryopoiesis and platelet formation, providing a new example of successful translation of GWAS to function.  相似文献

以锥尾伞滑刃线虫(Bursaphelenchus conicaudatus)作为对照,在韩国不同地域和寄主中采集了15个松材线虫分离物、3 个拟松材线虫的分离物及5个未鉴定的该属的分离物,并对其进行了ITS 和 D2D3 rDNA序列分析。以单条雌线虫的DNA为模板,ITS 和 D2D3 区由PCR仪扩增并克隆其序列。结果表明,所有松材线虫的ITS 和 D2D3 区序列相同,没有种内变异。然而2个采自黑松和红松的拟松材线虫分离物的基因型分别是亚洲型和欧洲型。5个未鉴定线虫的数据表明,它们分别接近B. tusciae、B. lini、B. thailandae、B. doui和 B. hylobianum,该结果同时被形态学结果所支持。利用5种酶(Hinf I、Alu I、Msp I、Hae III、Rsa),PCR产物克隆并测序的数据也可以区分不同种和基因型。  相似文献

Fermionic superfluidity requires the formation of particle pairs, the size of which varies from the femtometre scale in neutron stars and nuclei to the micrometre scale in conventional superconductors. Many properties of the superfluid depend on the pair size relative to the interparticle spacing. This is expressed in 'BCS-BEC crossover' theories, describing the crossover from a Bardeen-Cooper-Schrieffer (BCS)-type superfluid of loosely bound, large Cooper pairs to Bose-Einstein condensates (BECs) of tightly bound molecules. Such a crossover superfluid has been realized in ultracold atomic gases where high-temperature superfluidity has been observed. The microscopic properties of the fermion pairs can be probed using radio-frequency spectroscopy. However, previous work was difficult to interpret owing to strong final-state interactions that were not well understood. Here we realize a superfluid spin mixture in which such interactions have negligible influence and present fermion pair dissociation spectra that reveal the underlying pairing correlations. This allows us to determine that the spectroscopic pair size in the resonantly interacting gas is 20 per cent smaller than the interparticle spacing. These are the smallest pairs so far observed in fermionic superfluids, highlighting the importance of small fermion pairs for superfluidity at high critical temperatures. We have also identified transitions from fermion pairs to bound molecular states and to many-body bound states in the case of strong final-state interactions.  相似文献

Calcium (Ca(2+))-activated chloride channels are fundamental mediators in numerous physiological processes including transepithelial secretion, cardiac and neuronal excitation, sensory transduction, smooth muscle contraction and fertilization. Despite their physiological importance, their molecular identity has remained largely unknown. Here we show that transmembrane protein 16A (TMEM16A, which we also call anoctamin 1 (ANO1)) is a bona fide Ca(2+)-activated chloride channel that is activated by intracellular Ca(2+) and Ca(2+)-mobilizing stimuli. With eight putative transmembrane domains and no apparent similarity to previously characterized channels, ANO1 defines a new family of ionic channels. The biophysical properties as well as the pharmacological profile of ANO1 are in full agreement with native Ca(2+)-activated chloride currents. ANO1 is expressed in various secretory epithelia, the retina and sensory neurons. Furthermore, knockdown of mouse Ano1 markedly reduced native Ca(2+)-activated chloride currents as well as saliva production in mice. We conclude that ANO1 is a candidate Ca(2+)-activated chloride channel that mediates receptor-activated chloride currents in diverse physiological processes.  相似文献

Boc is a receptor for sonic hedgehog in the guidance of commissural axons   总被引:1,自引:0,他引:1  
In the spinal cord, sonic hedgehog (Shh) is secreted by the floor plate to control the generation of distinct classes of ventral neurons along the dorsoventral axis. Genetic and in vitro studies have shown that Shh also later acts as a midline-derived chemoattractant for commissural axons. However, the receptor(s) responsible for Shh attraction remain unknown. Here we show that two Robo-related proteins, Boc and Cdon, bind specifically to Shh and are therefore candidate receptors for the action of Shh as an axon guidance ligand. Boc is expressed by commissural neurons, and targeted disruption of Boc in mouse results in the misguidance of commissural axons towards the floor plate. RNA-interference-mediated knockdown of Boc impairs the ability of rat commissural axons to turn towards an ectopic source of Shh in vitro. Taken together, these data suggest that Boc is essential as a receptor for Shh in commissural axon guidance.  相似文献
