A simple and effective method for total RNA isolation of appressoria in Magnaporthe oryzae

Appressorium formation is an important event in establishing a successful interaction between the rice blast fungus, Magnaporthe oryzae, and its host plant, rice. An understanding of molecular events occurring in appressorium differentiation will give new strategies to control rice blast. A quick and reliable method to extract total RNA from appressorium is essential for studying gene expression during appressorium formation and its mechanism. We found that duplicate film is an efficient substratum for appressorium formation, even when inoculated with high density conidia. When inoculated with conidia at 1 × 106 ml^-1, the percentages of conidium germination and appressorium formation were （97.98±0.67）% and （97.88±0.45）%, respectively. We applied Trizol before appressorium collection for total RNA isolation, and as much as 113.6 lag total RNA was isolated from the mature appressoria at 24 h after inoculation. Functional analysis of two genes, MNH6 and MgATG1, isolated from the cDNA subtractive library, revealed that the quantity of RNA was good enough to construct a cDNA （complementary DNA） library or a cDNA subtractive library. This method may be also applicable for the appressorium RNA isolation of other pathogenic fungi in which conidia differentiate into appressoria in the early stages of host infection.     

Clinical investigation of surgery for intermittent exotropia

Objective： To investigate the time and postoperative binocular vision of strabismus surgery for children with intermittent exotropia （X（T））. Methods： A retrospective investigation was conducted in 80 child patients with intermittent exotropia. Pre- and postoperative angles of deviation fixating at near （33 cm） and distant targets （6 m） were measured with the prolonged alternate cover testing. The binocular function was assessed with synoptophore. Twenty-one patients took the postoperative synoptophore exercise. Results： （1） A week after surgery, 96.2% of the 80 patients had binocular normotopia, while a year after surgery, 91.3% of the 80 patients had binocular normotopia; （2） Preoperatively, 58 patients had near stereoacuity, while postoperatively, 72 patients achieved near stereoacuity （P〈0.05）; （3） Preoperatively, 64 patients had Grade Ⅰ for the synoptophore evaluation and postoperatively, 76 patients achieved Grade Ⅰ. Meanwhile, 55 patients had Grade Ⅱ preoperatively and 72 achieved Grade Ⅱ postoperatively. For Grade Ⅲ, there were 49 patients preoperatively and 64 patients postoperatively （P〈0.05）; （4） Patients of 5-8 years old had a significantly better recovery rate of binocular vision than those of 9-18 years old （P〈0.05）; （5） Patients taking postoperative synoptophore exercise had a better binocular vision than those taking no exercise （P〈0.05）. Conclusions： （1） Strabismus surgery can help to preserve or restore the binocular vision for intermittent exotropia; （2） Receiving the surgery at young ages may develop better postoperative binocular vision; （3） The postoperative synoptophore exercise can help to restore the binocular vision.     

基于集合分析的数据查询方法

针对具有全称量词及蕴含运算查询构造困难的问题,用数学中集合论工具,分析了这两类查询的本质,得到了关于这两类查询应满足的集合条件的两个定理.应用这两个定理,推导出了实现几类复杂查询的统一查询模式.推出的查询集合条件简单明了,从而使具有全称量词和蕴含运算命题查询的构造更容易理解.典型应用示例验证了基于集合分析数据查询方法的有效性.     

基于混沌映射的图像加密硬件实现结构

为了使图像加密硬件化和实用化,针对图像加密系统所需的置换、替代和扩散3种基本要素,提出了一种采用双RAM连接,加/解密过程多轮次、可复用的硬件实现结构.该实现结构通过地址产生器得到置换地址,与地址计数器配合完成像素的位置置换;通过灰度扩散与像素替代单元完成灰度的扩散与替代.验证中采用广义猫映射进行像素位置置换,将单向耦合映射格点用于像素的替代,并与灰度扩散相结合,两种混沌映射多轮次交替使用.分析和实验证明,该结构具有复用性好、吞吐率高且面积节约等特点.现场可编程门阵列验证表明,在200 MHz时钟频率下,进行4轮加/解密时的吞吐率分别达到4.17 Mbyte/s 和3.85 Mbyte/s.     

武夷山土壤有效钾垂直分异研究

通过对武夷山六个典型剖面有效钾含量的分析发现：从山顶到山脚随着海拔的逐渐降低,土壤表层有效钾含量呈降低趋势,2号剖面有效钾含量最高,达88 mg/kg,6号剖面有效钾含量最低,只有39 mg/kg,1,2,5号剖面的有效钾含量处于中度缺乏状态;3,4,6号剖面的有效钾含量处于严重缺乏状态.其成因主要是由于本区位于中亚热带南缘,淋溶作用较强,且随着海拔降低,年均温升高,脱硅富铝化作用加强,同时人为影响也对该分布格局产生一定影响.     

电力工业大气污染治理技术开发 的反思与模式创新

通过回顾我国大气污染治理技术的发展历程以及对我国现有大气污染治理技术的现状分析,针对我国在大气污染治理技术开发模式中存在的瓶颈,提出了一种以全方位多尺度系统级数值仿真模拟技术为核心,以要点试验和工程回归为辅助手段的新型技术开发模式,并实际应用该模式实现了大型火电机组烟气脱硫脱硝技术的开发和大规模工程应用,实现了技术的短周期开发和快速成熟。
