Chemical genomics involves generating large collections of small molecules and using them to modulate cellular states. Despite recent progress in the systematic synthesis of structurally diverse compounds, their use in screens of cellular circuitry is still an ad hoc process. Here, we outline a general, efficient approach called gene expression-based high-throughput screening (GE-HTS) in which a gene expression signature is used as a surrogate for cellular states, and we describe its application in a particular setting: the identification of compounds that induce the differentiation of acute myeloid leukemia cells. In screening 1,739 compounds, we identified 8 that reliably induced the differentiation signature and, furthermore, yielded functional evidence of bona fide differentiation. The results indicate that GE-HTS may be a powerful, general approach for chemical screening.  相似文献

Past studies of tectonically active mountain ranges have suggested strong coupling and feedbacks between climate, tectonics and topography. For example, rock uplift generates topographic relief, thereby enhancing precipitation, which focuses erosion and in turn influences rates and spatial patterns of further rock uplift. Although theoretical links between climate, erosion and uplift have received much attention, few studies have shown convincing correlations between observable indices of these processes on mountain-range scales. Here we show that strongly varying long-term (>10(6)-10(7) yr) erosion rates inferred from apatite (U-Th)/He cooling ages across the Cascades mountains of Washington state closely track modern mean annual precipitation rates. Erosion and precipitation rates vary over an order of magnitude across the range with maxima of 0.33 mm yr(-1) and 3.5 m yr(-1), respectively, with both maxima located 50 km west (windward) of the topographic crest of the range. These data demonstrate a strong coupling between precipitation and long-term erosion rates on the mountain-range scale. If the range is currently in topographic steady state, rock uplift on the west flank is three to ten times faster than elsewhere in the range, possibly in response to climatically focused erosion.  相似文献

Extracellular Ca(2+) (Ca(2+)(o)) is required for various physiological and developmental processes in animals and plants. In response to varied Ca(2+)(o) levels, plants maintain relatively constant internal Ca(2+) content, suggesting a precise regulatory mechanism for Ca(2+) homeostasis. However, little is known about how plants monitor Ca(2+)(o) status and whether Ca(2+)(o)-sensing receptors exist. The effects of Ca(2+)(o) on guard cells in promoting stomatal closure by inducing increases in the concentration of cytosolic Ca(2+) ([Ca(2+)](i)) provide a clue to Ca(2+)(o) sensing. Here we have used a functional screening assay in mammalian cells to isolate an Arabidopsis complementary DNA clone encoding a Ca(2+)-sensing receptor, CAS. CAS is localized to the plasma membrane, exhibits low-affinity/high-capacity Ca(2+) binding, and mediates Ca(2+)(o)-induced [Ca(2+)](i) increases. CAS is expressed predominantly in the shoot, including guard cells. Repression of CAS disrupts Ca(2+)(o) signalling in guard cells, and impairs bolting (swift upward growth at the transition to seed production) in response to Ca(2+) deficiency, so we conclude that CAS may be a primary transducer of Ca(2+)(o) in plants.  相似文献

Tumour necrosis factor (TNF) was found originally in mouse serum after intravenous injection of bacterial endotoxin into mice primed with viable Mycobacterium bovis, strain Bacillus Calmette-Guerin (BCG). TNF-containing serum from mice is cytotoxic or cytostatic to a number of mouse and human transformed cell lines, but less or not toxic to normal cells in vitro. It causes necrosis of transplantable tumours in mice. TNF also occurs in serum of rat, rabbit and guinea pig. Rabbit TNF has been purified recently to give a single band on SDS-polyacrylamide gel electrophoresis (PAGE). The purified TNF had a relative molecular mass (Mr) 40,000 +/- 5,000 measured by gel filtration, and 17,000 by SDS-PAGE. Its isoelectric point is 5.0 +/- 0.3. The necrotic activity in vivo and the cytotoxicity in vitro are produced by the same substance. The gene encoding TNF has been identified in a human genomic DNA library using as a probe a cloned cDNA encoding a portion of rabbit TNF. The regions of this gene encoding an amino-acid sequence corresponding to mature TNF have been expressed in Escherichia coli and the product of this expression isolated in pure form and shown to produce necrosis of murine tumours in vivo.  相似文献

HLA class II molecules are surface glycoproteins which are essential in the initiation of immune responses. It has been postulated that induction of class II in epithelial cells such as endocrine cells, which are normally class II negative, may result in autoimmunity. In type I diabetes, islet beta cells, the target of the autoimmune process, selectively express class II antigens. But in contrast to most other cell types, islet beta cells are not stimulated to express class II by interferon-gamma (IFN-gamma) and thus the conditions under which this induction occurs have been particularly elusive. The cytotoxins tumour necrosis factor (TNF) and lymphotoxin (LT) synergize with IFN-gamma in a number of activities. We report here that IFN-gamma in combination with either TNF or LT induces islet cell class II expression. This finding has important implications for the pathogenesis of type I diabetes and the understanding of the differential control of class II expression.  相似文献
