Accumulation of DNA damage leading to adult stem cell exhaustion has been proposed to be a principal mechanism of ageing. Here we address this question by taking advantage of the highly specific role of DNA ligase IV in the repair of DNA double-strand breaks by non-homologous end-joining, and by the discovery of a unique mouse strain with a hypomorphic Lig4(Y288C) mutation. The Lig4(Y288C) mouse, identified by means of a mutagenesis screening programme, is a mouse model for human LIG4 syndrome, showing immunodeficiency and growth retardation. Diminished DNA double-strand break repair in the Lig4(Y288C) strain causes a progressive loss of haematopoietic stem cells and bone marrow cellularity during ageing, and severely impairs stem cell function in tissue culture and transplantation. The sensitivity of haematopoietic stem cells to non-homologous end-joining deficiency is therefore a key determinant of their ability to maintain themselves against physiological stress over time and to withstand culture and transplantation. 相似文献

In all complex organisms, the peripheral nerves ensure the portage of information from the periphery to central computing
and back again. Axons are in part amazingly long and are accompanied by several different glial cell types. These peripheral
glial cells ensure electrical conductance, most likely nuture the long axon, and establish and maintain a barrier towards
extracellular body fluids. Recent work has revealed a surprisingly similar organization of peripheral nerves of vertebrates
and Drosophila. Thus, the genetic dissection of glial differentiation in Drosophila may also advance our understanding of basic principles underlying the development of peripheral nerves in vertebrates. 相似文献

Soccer (football) is identified as the most popular sport in the world with a high turnover of economic-financial resources. The different segments of the soccer (football) value chain exert mutual influence on the variables that rule the dynamics of this sector. In this sense it is necessary to identify the variables that represent the soccer (football) value chain systemically; to understand the interrelationships between these variables, the main difficulties derived from this interrelationship and to point out variables that require actions to increment the results of the soccer (football) value chain as a whole. Hence, this study applied a semi-structured questionnaire with open-ended questions about the problems and leveraging variables that exist in this sports modality. A causal linkage diagram was elaborated based on the answers to the interviews, showing the systemic structure of the soccer (football) value chain in Brazil, whose premise it is to identify the existing circularities and the possibly existing points of leverage. The systemic structure pointed to the economic participation of soccer (football) in the Gross Domestic Product as a central variable to be leveraged, which tends to synthetize the results of the joint action of the actors in this chain. Furthermore, the variables that may limit or amplify the growth were identified. The systemic structure showed that the soccer (football) commodity chain is complex because of the great number of variables and interrelationships needed to represent its dynamics. The relationship between the actors also proved to be a factor of impact on economic and social development. 相似文献

Mass coral bleaching events caused by elevated seawater temperatures have resulted in extensive coral mortality throughout the tropics over the past few decades. With continued global warming, bleaching events are predicted to increase in frequency and severity, causing up to 60% coral mortality globally within the next few decades. Although some corals are able to recover and to survive bleaching, the mechanisms underlying such resilience are poorly understood. Here we show that the coral host has a significant role in recovery and resilience. Bleached and recovering Montipora capitata (branching) corals met more than 100% of their daily metabolic energy requirements by markedly increasing their feeding rates and CHAR (per cent contribution of heterotrophically acquired carbon to daily animal respiration), whereas Porites compressa (branching) and Porites lobata (mounding) corals did not. These findings suggest that coral species with high-CHAR capability during bleaching and recovery, irrespective of morphology, will be more resilient to bleaching events over the long term, could become the dominant coral species on reefs, and may help to safeguard affected reefs from potential local and global extinction. 相似文献

Schistosoma mansoni is the primary causative agent of schistosomiasis, which affects 200 million individuals in 74 countries. We generated 163,000 expressed-sequence tags (ESTs) from normalized cDNA libraries from six selected developmental stages of the parasite, resulting in 31,000 assembled sequences and 92% sampling of an estimated 14,000 gene complement. By analyzing automated Gene Ontology assignments, we provide a detailed view of important S. mansoni biological systems, including characterization of metazoa-specific and eukarya-conserved genes. Phylogenetic analysis suggests an early divergence from other metazoa. The data set provides insights into the molecular mechanisms of tissue organization, development, signaling, sexual dimorphism, host interactions and immune evasion and identifies novel proteins to be investigated as vaccine candidates and potential drug targets. 相似文献

The vasculotropic E22Q mutant of the amyloid-β (Aβ) peptide is associated with hereditary cerebral hemorrhage with amyloidosis

Comparative assessment of structural parameters and cell death mechanisms elicited in primary human cerebral endothelial cells
by AβE22Q and wild-type Aβ revealed that only AβE22Q triggered the Bax mitochondrial pathway of apoptosis. AβE22Q neither
matched the fast oligomerization kinetics of Aβ42 nor reached its predominant β-sheet structure, achieving a modest degree
of oligomerization with a secondary structure that remained a mixture of β and random conformations. The endogenous molecule
tauroursodeoxycholic acid (TUDCA) was a strong modulator of AβE22Q-triggered apoptosis but did not significantly change the
secondary structures and fibrillogenic propensities of Aβ peptides. These data dissociate the pro-apoptotic properties of
Aβ peptides from their distinct mechanisms of aggregation/fibrillization in vitro, providing new perspectives for modulation of amyloid toxicity.

Clibanarius symmetricus is a diogenid hermit crab that is highly abundant in rocky intertidal environments, including the rock outcrops of Amazon estuaries. This study characterises the C. symmetricus shell utilisation pattern in the Marapanim River estuary, Pará, Equatorial Amazon, based on the hypothesis that occupancy patterns would differ, in relation to biometry and diversity of occupied shells, from those of other regions in the range of this species due to the distinct and unique environmental characteristics of equatorial estuaries. Monthly samplings were carried out from August 2006 to July 2007, in the upper and lower areas of the mid-littoral during low tides. A total of seven gastropod species were found as shells occupied by hermit crabs: 93.33% were Thaisella trinitatensis shells, 2.00% Nassarius vibex, 2.00% Neritina virginea, 1.33% Natica marochiensis, 0.67% Parvanachis obesa, and, occasionally, Littorina flava and Phalium granulatum shells, each with only one occurrence (0.33%). Juveniles [cephalothoracic shield length (CL) of less than 3.6 mm] occupied a higher variety of shells, while adults occupied shells with larger meristic variation. Males occupied larger shells. The length of the animal was influenced by shell measurements (total width, aperture width and weight). Clibanarius symmetricus showed occupancy patterns generally similar to those of specimens of the same species previously studied in the Brazilian subtropical region, and this occupancy is explained by shell availability, shell size and weight, and hermit size. However, this study showed lower occupied shell species diversity, and the species with highest occupancy frequency (T. trinitatensis) was not reported in any previous studies on this hermit crab.
